Demethylase-assisted site-specific detection of N^(1)-methyladenosine in RNA

The dynamic RNA modifications have been viewed as new posttranscriptional regulator in modulating gene expression as well as in a broad range of physiological processes.N^(1)-methyladenosine(m^(1)A)is one of the most prevalent modifications existing in multiple types of ***-depth investigation of the functions of m^(1)A requires the site-specific assessment of m^(1)A stoichiometry in ***,we established a demethylase-assisted method(DA-m^(1)A)for the site-specific detection and quantification of m^(1)A in RNA.N^(1)-methyl group in m^(1)A could result in the stalling of reverse transcription at m^(1)A site,thus producing the truncated *** AlkB is a demethylase that can demethylate m^(1)A to produce adenine in RNA,thus generating full-length cDNA from AlkB-treated *** of the produced amounts of full-length cDNA by quantitative real-time PCR can achieve the site-specific detection and quantification of m^(1)A in *** the DA-m^(1)A method,we examined and successfully confirmed the previously well-characterized m^(1)A sites in various types of RNAs with low false positive *** addition,we found that the level of m^(1)A was significantly decreased at the bromodomain containing 2(BRD2)mRNA position 1674 and CST telomere replication complex component 1(CTC1)mRNA position 5643 in human hepatocellular carcinoma *** results suggest that these two m^(1)A sites in mRNA may be involved in liver *** together,the DA-m^(1)A method is simple and enables the rapid,cost-effective,and site-specific detection and quantification of m^(1)A in RNA,which provides a valuable tool to decipher the functions of m^(1)A in human diseases.

Early renal injury indicators can help evaluate renal injury in patients with chronic hepatitis B with long-term nucleos(t)ide therapy

6-Thioguanine(6TG)is a widely used chemotherapeutic agent for the treatment of a variety of human diseases including acute lymphoblastic *** entry into cells,6TG is metabolically converted into 6-thioguanosine(^(S)G)nucleotide that can be incorporated into the genome during DNA replication.^(S)G in genomic DNA could induce cell death by triggering the post-replicative mismatch repair(MMR)***,incorporation of 6TG into the Cp G sites could perturb the global DNA methylation and gene ***,the effect of 6TG on RNA modifications is still ***-toinosine(A-to-I)editing in RNA is one of the most common post-transcriptional modifications in mammals and there is growing evidence showing the significant alteration of A-to-I RNA editing in tumor tissues compared to normal *** the current study,we examined the incorporation of 6TG into RNA and investigated its effect on A-to-I editing of bladder cancer-associated protein(BLCAP)transcript in acute lymphoblastic leukemia *** results demonstrated that ^(S)G could be incorporated into various RNA species,with m RNA having the most abundant ^(S)*** addition,the results showed 6TG treatment elevated A-to-I editing in BLCAP transcript through upregulating adenosine deaminase 2 acting on RNA(ADAR2),which eventually contributes to the decreased cell *** study highlights a new mechanism of the cytotoxicity of 6TG in inducing cell death.

Forced boiling of nonazeotropic immiscible mixture in a supercapillary microchannel array for ultra-high heat flux removal with chip junction temperature below 85℃

In this study,a structure-optimized two-phase microchannel heat sink with sintered submicron nucleation sites was developed and *** copper-based microchannels had a rectangular cross-section with an equivalent hydraulic diameter of 222μ*** subcooled flow boiling characteristics were comprehensively compared between pure HFE-7100 and a non-azeotropic,immiscible binary mixture of HFE-7100 and water,considering heating areas of 1 and 5 cm^(2).The total heating power input to the test section were 100–1500 and 250–3000 W for a 1 and 5 cm^(2)heat source,respectively,with a flow rate ranging from 50 to150 L/*** to pure HFE-7100,the non-azeotropic immiscible binary mixture of HFE-7100/water in the sintered porous microchannels exhibited a much higher overall heat transfer coefficient and lower power *** maintain the junction temperature of a high power electronic chip below 85℃,the proposed supercapillary microchannel heat sink could effectively dissipate the heat flux of 1275 W/cm^(2)over 1 cm^(2)heat source and 500 W/cm^(2)over 5 cm^(2)heat *** addition,the volume ratio of the binary mixture strongly influence the two-phase flow heat transfer *** optimal volume ratio exist in terms of the thermal resistance–pumping power minimization(HFE-7100:water=2:8 is recommended in this study).The findings of this investigation on the flow boiling properties of non-azeotropic immiscible mixtures help fill a gap in the related field.

Functional brain imaging studies commonly use either resting or passive task states as their control conditions, and typically identify the activation brain region associated with a specific task by subtracting the resting from the active task conditions. Numerous studies now suggest, however, that the resting state may not reflect true mental "rest" conditions. The mental activity that occurs during "rest" might therefore greatly influence the functional neuroimaging observations that are collected through the usual subtracting analysis strategies. Exploring the ongoing mental processes that occur during resting conditions is thus of particular importance for deciphering functional brain mapping results and obtaining a more comprehensive understanding of human brain functions. In this review article, we will mainly focus on the discussion of the current research background of functional brain mapping at resting state and the physiological significance of the available neuroimaging data.

Mismatch loss analysis of solar array output power on stratospheric airship during flight

Stratospheric airships are long-endurance aerostats and have broad *** of the energy required for their operation is obtained from solar radiation,which makes accurate calculation of the energy output from the solar array crucial to the design and flight planning of the ***,the status of each photovoltaic module in the solar array may differ due to the airship curvature,resulting in mismatch losses and lowered output power,which has not been widely *** this paper,an irradiation model and a thermal model are established based on the actual arrangement of the *** output power model is established considering the non-uniform radiation in the *** mismatch losses of the array are analyzed under different flight *** output power of the solar array is decreased by up to 31.6%compared to the ideal ***,the proportion of mismatch losses increases with latitude,but the maximum mismatch loss power occurs at ***,an array reconfiguration method is proposed based on the irradiance dispersion index and position dispersion *** reconfigured array increases output power by 11.5%and can maintain energy balance in continuous *** results can be used to correct the overestimation of the output power during the airship design or to guide the configuration of the solar array.

In recent years,deep learning methods have developed rapidly and found application in many fields,including natural language *** the field of aspect-level sentiment analysis,deep learning methods can also greatly improve the performance of ***,previous studies did not take into account the relationship between user feature extraction and contextual *** address this issue,we use data feature extraction and deep learning combined to develop an aspect-level sentiment analysis *** be specific,we design user comment feature extraction(UCFE)to distill salient features from users’historical comments and transform them into representative user feature ***,the aspect-sentence graph convolutional neural network(ASGCN)is used to incorporate innovative techniques for calculating adjacency matrices;meanwhile,ASGCN emphasizes capturing nuanced semantics within relationships among aspect words and syntactic dependency ***,three embedding methods are devised to embed the user feature vector into the ASGCN *** empirical validations verify the effectiveness of these models,consistently surpassing conventional benchmarks and reaffirming the indispensable role of deep learning in advancing sentiment analysis methodologies.

Nano-enabled silicone-rubber articles for feeding or chewing could be a source of metallic nanoparticles(NPs)directly exposed to infants and young ***,the impact of steam disinfection on release of NPs and the related potential risks to children's health are ***,we investigated contents and form of Ag and Zn in 57 nano-enabled silicone-rubber baby bottle teats,pacifiers,and teethers of seven countries and examined the impacts of steam disinfection on in vitro bioaccessibility(IVBA)of Ag and Zn in the *** 89%articles had a mixture of Ag-and Zn-containing NPs and the teethers had relatively high Ag and Zn contents(up to 501 and 254μg/g,respectively).Steam disinfection caused rubber decomposition into micro(nano)plastics(0.54-15.7μm)and NP release from the interior of bulk rubber and micro-sized plastics,thus enhancing the IVBA of Ag and Zn by up to 5.5 *** findings provide insights into mechanisms for NP release by steam *** oral exposure risk assessment suggested low health concerns on individual metal release,our study points out the need to assess the potential health risks of child co-exposure to metallic NPs and micro(nano)plastics.

Analysis of Incidence of Atrial Fibrillation after Implantation of VVI Pacemaker-Long-term Observation of 154 Patients

Incidence of occurrence of atrial fibrillation (Af) after implantation of VVI was examined and the possible mechanism was explored. Eighty cases of atriouentricular block (AVB) and 74 cases of sick sinus syhdrome (SSS) were studied and followed up for 1-14 years after implantation of VVI. The endpoint was the occurrence of permanent Af. The results showed that the incidence of Af among the 154 patients was 14.3 % (22/154). And the incidence was 2. 5 % among patients with AVB (2/80), and 27 % among patients with SSS (20/74). Significant significance was found between patients with AVB and those with SSS (P<0. 01). Among the patients with SSS, the incidence was 33. 3 % (9/27) in type I, 38.1 % (8/21) in type Ⅲ and 11. 5 % (3/26) in type Ⅱ. Significant differences were revealed among patients with Ⅱ, Ⅲ and Ⅱ type SSS (P<0. 05). It is concluded that retrograde conduction of pure ventricular pacing may play the chief role of occurrence of Af.

Resistance-capacitance model of the capillary heat exchanger in subway tunnels

The subway operation generates a large amount of waste heat,which will lead to a disruption of the underground thermal *** remove the heat,a ground source heat pump system adopting capillary net as the front-end heat exchanger can be built in the tunnels to recycle the waste heat and use it for building *** simulation method was used to analyze the heat transfer process in the tunnel and the surrounding *** existing simulation method usually takes a long time for calculation and can not be used for a real-time regulation of the *** on the concept of thermoelectric analogy,a resistance-capacitance(RC)model was proposed for rapid prediction and analysis on the heat transfer of the capillary net in subway *** annual thermal response in the tunnel was analyzed,and the influence of the tunnel air temperature and the capillary inlet water temperature was examined using this *** maximum heat transfer flux of the capillary in summer(release heat)and in winter(extract heat)was 55.27 W/m 2 and 38.33 W/m 2,*** effect of capillary inlet water temperature was more remarkable than that of the tunnel air temperature on the capillary heat exchange process.
