Hepatoprotective and proapoptotic effect of Ecballium elaterium on CCl_4-induced hepatotoxicity in rats

Objective: To investigate the effects of perorally administered juice on tetrachloromethane(CCl4)-induced hepatotoxicity model in rats. Methods: Male Wistar rats were tubeadministrated silymarin, Ecballium juice at 0.2 mL/kg and 0.7 mL/kg daily for 3 consequent days, ie. 3.28 μg and 11.48 μg of cucurbitacin B per kg of body weight respectively. On the third day, liver damage was induced by intraperitoneal application of CCl4. On the fourth day, abdominal cavity was macroscopically examined and liver samples were taken for histopathological and immunochemical evaluation. HPLC was used to determine the content of the active substance cucurbitacin B. Results: The experiment revealed that 0.7 ml/kg juice concentration expressed the highest pro-apoptotic activity, but with prevailing negative effects. Compared with the lower concentration, there was an observable vasodilatation with consequent interstitial hemorrhages and a larger scope of inflammatory damage, which suppressed the hepatoprotective effect. In the 0.2 mL/kg concentration, there was a smaller pro-apoptotic activity but other parameters had better results, and the liver parenchyma damage was reversible. Conclusions: No reactions confirming the potentially allergic effect on laboratory rats were observed; its hepatoprotective and anti-inflammatory effect was confirmed on a model of acute liver damage.

Cytotoxic, kinetics of inhibition of carbohydrate-hydrolysing enzymes and oxidative stress mitigation by flavonoids roots extract of Dicoma anomala(Sond.)

Objective: To investigate the free radical scavenging, antidiabetic, kinetics and cytotoxic potentials of flavonoids extract of Dicoma anomala root by using standard methods. Methods:Antioxidant activity of the flavonoids was investigated at scavenging free radicals such as 1,1-diphenyl-2-picrylhydrazyl, nitric oxide, hydroxyl radical, reducing capabilities, 2,2-azinobis(3-ethylbenzothiazoline-6) sulfonic acid as well as metal chelating capability at different concentrations(0.125-1.000 mg/mL) while the antidiabetic activity was evaluated via the inhibition and kinetics of carbohydrate digestive enzymes including, alpha glucosidase, sucrase and maltase. Brine shrimp lethality assay was also employed to examine the cytotoxic effects of the extract by using different range of concentrations(0.125-2.000 mg/mL). Results: The study revealed the best antioxidant activity of the extract in 1,1-diphenyl-2-picrylhydrazyl,2,2-azino-bis(3-ethylbenzothiazoline-6) sulfonic acid and nitric oxide having IC_(50) values of(386.90±4.91),(736.00±38.12),(629.30±9.62) μg/mL respectively compared with quercetin(standard) with IC_(50) [(522.20±12.38),(1 021.00±15.61) and(1 075.00±29.35) μg/mL]respective values while it was insignificantly(P>0.05) at par with quercetin for reducing power. Similarly, the extract exhibited a moderate inhibition of alpha glucosidase(43.1%),sucrase(33.4%) and maltase(29.9%) activities which were significantly(P<0.05) better than acarbose(18.4%, 12.7% and 24.9% respectively) although acarbose(46.1%) inhibited the higher activity of alpha amylase than the extract(13.7%). The kinetics of mode of inhibition of alpha amylase, alpha glucosidase, sucrase and maltase by flavonoids extract of Dicoma anomala revealed an uncompetitive, non-competitive, competitive and non-competitive inhibition respectively. The result of the lethality assay showed a potent cytotoxic effect of the flavonoids with LC_(50) value 0.510 mg/mL. Conclusions: The results obtained from this stud

Total resection of glioblastoma(GB)tumors is nearly impossible,and systemic administration of temozolomide(TMZ)is often *** study presents a hybrid layered composite nanofiber network(LHN)designed for localized treatment in GB tumor *** LHN,consisting of polyvinyl alcohol and core-shell polylactic acid layers,was loaded with TMZ and *** vitro analysis revealed that LHN^(TMZ) and LHNrutin decelerated epithelial-mesenchymal transition and growth of stem-like cells,while the combination,LHN^(TMZ)+rutin,significantly reduced sphere size compared to untreated and LHNTMZ-treated cells(Ppared to untreated *** changes in tumor mitochondria,reduced membrane potential,and decreased parP expression indicated the activation of apoptotic pathways in tumor cells,which was further confirmed by a reduction in PHH3,indicating decreased mitotic activity of tumor ***,the local application of LHNs in the GB model mitigated aggressive tumor features without causing local tissue inflammation or adverse systemic *** was evidenced by a decrease in the angiogenesismarker CD31,the absence of inflammation or necrosis in H&E staining of the cerebellum,increased production of IFN-γ,decreased levels of interleukin-4 in splenic T cells,and lower serum AST *** findings collectively indicate that LHN^(TMZ)+rutin is a promising biocompatible model for the local treatment of GB.

The cryptic lifestyle of most fungi necessitates molecular identification of the guild in environmental *** the past decades,rapid development and affordability of molecular tools have tremendously improved insights of the fungal diversity in all ecosystems and ***,in spite of the progress of molecular methods,knowledge about functional properties of the fungal taxa is vague and interpretation of environmental studies in an ecologically meaningful manner remains *** order to facilitate functional assignments and ecological interpretation of environmental studies we introduce a user friendly traits and character database FungalTraits operating at genus and species hypothesis *** the information from previous efforts such as FUNGuild and FunFun together with involvement of expert knowledge,we reannotated 10,210 and 151 fungal and Stramenopila genera,*** resulted in a stand-alone spreadsheet dataset covering 17 lifestyle related traits of fungal and Stramenopila genera,designed for rapid functional assignments of environmental *** order to assign the trait states to fungal species hypotheses,the scientific community of experts manually categorised and assigned available trait information to 697,413 fungal ITS *** the basis of those sequences we were able to summarise trait and host information into 92,623 fungal species hypotheses at 1%dissimilarity threshold.
