CD4^(+)T cells can"help"or"license" conventional type 1 dendritic cells(cDC1s)to induce CD8^(+)cytotoxic T lymphocyte(CTL)anticancer responses,as proven in mouse *** recently identified cDC1s with a transcriptomic imprint of CD4^(+)T-cell help,specifically in T-cell-infiltrated human cancers,and these cells were associated with a good prognosis and response to PD-1-targeting ***,we delineate the mechanism of cDC1 licensing by CD4^(+)T cells in *** CD4^(+)T cells produce IFNβvia the STING pathway,which promotes MhC-I antigen(cross-)presentation by cDC1s and thereby improves their ability to induce CTL anticancer *** cooperation with CD40 ligand(L),IFNβalso optimizes the costimulatory and other functions of cDC1s required for CTL response ***-I-producing CD4^(+)T cells are present in diverse T-cell-infiltrated cancers and likely deliver“help”signals to CTLs locally,according to their transcriptomic profile and colocalization with“helped/licensed”cDCs and tumor-reactive CD8^(+)T *** agreement with this scenario,the presence of IFN-I-producing CD4^(+)T cells in the TME is associated with overall survival and the response to PD-1 checkpoint blockade in cancer patients.

Genetic susceptibility to metabolic associated fatty liver disease(MAFLD)is complex and poorly *** characterization of the genetic background of hepatic fat content would provide insights into disease etiology and causality of risk *** performed genome-wide association study(GWAS)on two noninvasive definitions of hepatic fat content:magnetic resonance imaging proton density fat fraction(MRI-PDFF)in 16,050 participants and fatty liver index(FLI)in 388,701 participants from the United Kingdom(UK)Biobank(UKBB).heritability,genetic overlap,and similarity between hepatic fat content phenotypes were analyzed,and replicated in 10,398 participants from the University Medical Center Groningen(UMCG)Genetics Lifelines Initiative(UGLI).Meta-analysis of GWASs of MRI-PDFF in UKBB revealed five statistically significant loci,including two novel genomic loci harboring CREB3L1(rs72910057-T,P=5.40E−09)and GCM1(rs1491489378-T,P=3.16E−09),respectively,as well as three previously reported loci:PNPLA3,TM6SF2,and *** of FLI in UKBB identified 196 genome-wide significant loci,of which 49 were replicated in UGLI,with top signals in ZPR1(P=3.35E−13)and FTO(P=2.11E−09).Statistically significant genetic correlation(rg)between MRI-PDFF(UKBB)and FLI(UGLI)GWAS results was found(rg=0.5276,P=1.45E−03).Novel MRI-PDFF genetic signals(CREB3L1 and GCM1)were replicated in the FLI *** identified two novel genes for MRI-PDFF and 49 replicable loci for *** a difference in hepatic fat content assessment between MRI-PDFF and FLI,a substantial similar genetic architecture was *** is identified as an easy and reliable approach to study hepatic fat content at the population level.

This research aims to analyze the static bending and free vibration behaviors of bidirectional functionally graded porous(BFGP)curved microbeams with elastic boundary conditions resting on a Pasternak’s elastic foundation in a hygro-thermal *** analysis based on non-uniform rational B-splines,first-order shear deformation theory,nonlocal elasticity theory combined with the modified strain gradient theories,modified Timoshenko beam theory are formulated to depict bending and shear deformations of BFGP curved ***,because using the modified Timoshenko beam theory,this study removes the requirement for a shear correction factor and describes shear stress by zero at the upper and lower cross-sectional sites of the BFGP curved *** from traditional boundary conditions,where the beginning and end positions of a curved beam are connected by an elastic system of straight and torsion *** allows for greater flexibility in controlling the stiffness of the springs to obtain arbitrary *** assess the accuracy and convergence of the proposed approach,validation numerical examples were conducted in the various examples.

Background:how do Orange River Francolins(Scleroptila levaillantoides) adapt to an intensive farming landscape with grass(grazing) camps and crop cultivation? To answer this question, a study was carried out in south-east of Johannesburg in South Africa to clarify the interaction of francolins with a landscape consisting of land use ***:A transect-road of 45 km was traversed weekly during November 2015–October 2016 through flat maize(Zea mays) and cattle(Bos primigenius) grazing fields. Francolins were counted in three land use mosaic types along the transect-road(including the road):grass camps on both sides of the road(grass/grass edges); arable fields on both sides(arable/arable edges) and arable land on one side with grass camps on the opposite side of the road(arable/grass edges).Results:Francolins used all three mosaic types, but the arable/arable edges were least frequented. Nonetheless, the arable/arable edges played an important part:francolins, including females with chicks, moved along the arable/arable edges, which enabled contact between sub-populations, and the arable/arable edges provided temporary cover(e.g. stubble and maize plants) and ploughed firebreaks to forage(e.g. for bulbs).Conclusion:Cultivation of crop within grasslands does not constrain breeding, movement and habitat use by the Orange River Francolin. For conservation purposes it is critical that the fine-scale mosaic of grazing and cultivation areas remain intact.

The alpine terrestrials of the Maloti-Drakensberg in southern Africa play crucial roles in ecosystem functions and livelihoods,yet they face escalating degradation from various factors including overgrazing and climate *** study employs advanced Digital Soil Mapping(DSM)techniques coupled with remote sensing to map and assess wetland coverage and degradation in the northern *** model achieved high accuracies of 96%and 92%for training and validation data,respectively,with Kappa statistics of 0.91 and 0.83,marking a pioneering automated attempt at wetland mapping in this *** attributes such as terrain wetness index(TWI)and valley depth(VD)exhibit significant positive correlations with wetland coverage and erosion gully density,Channel Network Depth and slope were negative *** density analysis revealed terrain attributes as dominant factors driving degradation,highlighting the need to consider catchment-specific susceptibility to *** challenge traditional assumptions which mainly attribute wetland degradation to external forces such as livestock overgrazing,ice rate activity and climate *** sensitivity map produced could serve as a basis for Integrated Catchment Management(ICM)projects,facilitating tailored conservation *** research should expand on this work to include other highland areas,explore additional covariates,and categorize wetlands based on hydroperiod and sensitivity to *** comprehensive study underscores the potential of DSM and remote sensing in accurately assessing and managing wetland ecosystems,crucial for sustainable resource management in alpine regions.
