Background Hypertension(HTN)involves genetic variability in the renin-angiotensin system and influences antihypertensive *** previously reported that angiotensinogen(AGT)messenger RNA(mRNA)is endogenously bound by miR-122-5p and rs699 A>G decreases reporter mRNA in the microRNA functional-assay *** AGT promoter variant rs5051 c>T is in linkage disequilibrium(LD)with rs699 A>G and increases AGT *** independent effect of these variants is understudied due to their LD therefore we aimed to test the hypothesis that increased AGT by rs5051 c>T counterbalances AGT decreased by rs699 A>G,and when these variants occur independently,it translates to HTN-related *** We used in silico,in vitro,in vivo,and retrospective models to test this *** In silico,rs699 A>G is predicted to increase miR-122-5p binding affinity by 3%.Mir-ecLIP results show rs699 is 40–45 nucleotides from the strongest microRNA-binding site in the AGT ***,rs699 A>G increases AGT mRNA in an AGT-plasmid-cDNA HepG2 expression ***-Tissue Expression(GTEx)and UK Biobank analyses demonstrate liver AGT expression and HTN phenotypes are not different when rs699 A>G occurs independently from rs5051 c>***,GTEx and the in vitro experiments suggest rs699 A>G confers cell-type-specific effects on AGT mRNA abundance,and suggest paracrine renal renin-angiotensin-system perturbations could mediate the rs699 A>G associations with *** We found that rs5051 c>T and rs699 A>G significantly associate with systolic blood pressure in Black participants in the UK Biobank,demonstrating a fourfold larger effect than in White *** studies are warranted to determine if altered antihypertensive response in Black individuals might be due to rs5051 c>T or rs699 A>*** like this will help clinicians move beyond the use of race as a surrogate for genotype.

Hadal trenches are characterized by enhanced and infrequent high-rate episodic sedimentation events that likely introduce not only labile organic carbon and key nutrients but also new microbes that significantly alter the subseafloor ***,the role of high-rate episodic sedimentation in controlling the composition of the hadal subseafloor microbiosphere is ***,analyses of carbon isotope composition in a~750 cm long sediment core from the challenger Deep revealed noncontinuous deposition,with anomalous ^(14)c ages likely caused by seismically driven mass transport and the funneling effect of trench *** community composition and diverse enzyme activities in the upper~27 cm differed from those at lower depths,probably due to sudden sediment deposition and differences in redox condition and organic matter *** lower depths,microbial population numbers,and composition remained relatively constant,except at some discrete depths with altered enzyme activity and microbial phyla abundance,possibly due to additional sudden sedimentation events of different *** is provided of a unique role for high-rate episodic sedimentation events in controlling the subsurface microbiosphere in Earth’s deepest ocean floor and highlight the need to perform thorough analysis over a large depth range to characterize hadal benthic *** depositional processes are likely crucial in shaping deep-water geochemical environments and thereby the deep subseafloor biosphere.

consider a c1 vector eld together with an ergodic invariant probability that hasℓnonzero Lya-punov *** orthonormal moving frames along a generic orbit we construct a linear system ofℓdi erential equations which is a linearized Liao standard *** show that Lyapunov exponents of this linear system coincide with all the nonzero exponents of the given vector eld with respect to the given ergodic ***,we prove that these Lyapunov exponents have a persistence property meaning that a small perturbation to the linear system(Liao perturbation)preserves both the sign and the value of the nonzero Lyapunov exponents.

We improve estimates for the distribution of primitiveλ-roots of a composite modulus q yielding an asymptotic formula for the number of primitiveλ-roots in any interval I of length |I| >> q1/2+c. Similar results are obtained for the distribution of ordered pairs (x,x-1) with x a primitive A-root, and for the number of primitiveλ-roots satisfying inequalities such as |x - x-1| < B.

How profitable is switchgrass in Illinois, USA? An economic definition of marginal land

Background:Decisions regarding the conversion of land from an existing crop to bioenergy crops are critical for the sustainable production of both food and *** study seeks to establish criteria for delineating land as“economically marginal”,and thus suited for growing ***:In this case study of an Illinois agricultural field,the profitability of switchgrass,with farmgate prices of$44 Mg−1,$66 Mg−1,or$88 Mg−1,was compared to corn and soybean crop ***,the study also evaluates the profitability of switchgrass when replacing corn‐based yield estimates from the Soil Productivity Index(SPI)of ***:Based on a dry‐matter yield of 10.45 Mg ha−1,switchgrass can compete with soybeans only at the high price of$88 Mg−1,but depending on location,can compete with corn at$66 Mg−*** Illinois,at$88 ha−1,all Illinois land with SPIclass c(SPI 100–116)is profitable under *** may not be profitable relative to corn grown in the SPI class A(SPI>133)and only 7%of class B(SPI 117–132).conclusions:Our results show that land with drainage and erosion limitations is economically marginal when corn and soybean yields are low,and the farmgate price for switchgrass is greater than$66 Mg−***,this may not be possible on land where switchgrass is replacing frequent soybean rotations(corn–soybean ratio≤1).Land used to produce only soybeans may only be marginal at the farmgate price of$88 Mg−*** studies need to be conducted to identify how much land can be converted to switchgrass without harming corn production.

Programmable microfluidic genotyping of plant DNA samples for marker-assisted selection

As demands to maintain the global food production continue to mount,multinational seed companies are turning to new DNA marker technologies to accelerate the rate of plant breeding and crop *** key to widespread adoption of molecular breeding is the availability of flexible and cost-effective tools that can perform combinatorial and high-throughput genotyping of single-nucleotide polymorphisms(SNPs)to guide the crop development *** this end,we have developed a programmable,droplet-based microfluidic device for genotyping maize genomic DNA.A unique feature of the microfluidic platform is the nano sample processors(NSPs),which allow the device to sequentially load an unrestricted number of unique DNA samples using only two inlets,overcoming the current limitation to the number of sample inputs due to small device *** and programmable droplet generation within the device allows each sample to be genotyped against a panel of markers on ***,we have successfully implemented the Invader assay for SNP genotyping in flowing,50-nL droplets,thus achieving significant reduction in consumption of reagents per reaction as compared with conventional genotyping *** a demonstration,we performed 240 Invader reactions(testing 8 DNA samples against 10 SNP markers)and achieved greater than 93% accuracy in SNP calling of plant DNA samples in a single droplet-based experiment.
