Background Hypertension(HTN)involves genetic variability in the renin-angiotensin system and influences antihypertensive *** previously reported that angiotensinogen(AGT)messenger RNA(mRNA)is endogenously bound by miR-122-5p and rs699 A>G decreases reporter mRNA in the microRNA functional-assay *** AGT promoter variant rs5051 c>T is in linkage disequilibrium(LD)with rs699 A>G and increases AGT *** independent effect of these variants is understudied due to their LD therefore we aimed to test the hypothesis that increased AGT by rs5051 c>T counterbalances AGT decreased by rs699 A>G,and when these variants occur independently,it translates to HTN-related *** We used in silico,in vitro,in vivo,and retrospective models to test this *** In silico,rs699 A>G is predicted to increase miR-122-5p binding affinity by 3%.Mir-ecLIP results show rs699 is 40–45 nucleotides from the strongest microRNA-binding site in the AGT ***,rs699 A>G increases AGT mRNA in an AGT-plasmid-cDNA HepG2 expression ***-Tissue Expression(GTEx)and UK Biobank analyses demonstrate liver AGT expression and HTN phenotypes are not different when rs699 A>G occurs independently from rs5051 c>***,GTEx and the in vitro experiments suggest rs699 A>G confers cell-type-specific effects on AGT mRNA abundance,and suggest paracrine renal renin-angiotensin-system perturbations could mediate the rs699 A>G associations with *** We found that rs5051 c>T and rs699 A>G significantly associate with systolic blood pressure in Black participants in the UK Biobank,demonstrating a fourfold larger effect than in White *** studies are warranted to determine if altered antihypertensive response in Black individuals might be due to rs5051 c>T or rs699 A>*** like this will help clinicians move beyond the use of race as a surrogate for genotype.

Hadal trenches are characterized by enhanced and infrequent high-rate episodic sedimentation events that likely introduce not only labile organic carbon and key nutrients but also new microbes that significantly alter the subseafloor ***,the role of high-rate episodic sedimentation in controlling the composition of the hadal subseafloor microbiosphere is ***,analyses of carbon isotope composition in a~750 cm long sediment core from the challenger Deep revealed noncontinuous deposition,with anomalous ^(14)c ages likely caused by seismically driven mass transport and the funneling effect of trench *** community composition and diverse enzyme activities in the upper~27 cm differed from those at lower depths,probably due to sudden sediment deposition and differences in redox condition and organic matter *** lower depths,microbial population numbers,and composition remained relatively constant,except at some discrete depths with altered enzyme activity and microbial phyla abundance,possibly due to additional sudden sedimentation events of different *** is provided of a unique role for high-rate episodic sedimentation events in controlling the subsurface microbiosphere in Earth’s deepest ocean floor and highlight the need to perform thorough analysis over a large depth range to characterize hadal benthic *** depositional processes are likely crucial in shaping deep-water geochemical environments and thereby the deep subseafloor biosphere.

Programmable microfluidic genotyping of plant DNA samples for marker-assisted selection

As demands to maintain the global food production continue to mount,multinational seed companies are turning to new DNA marker technologies to accelerate the rate of plant breeding and crop *** key to widespread adoption of molecular breeding is the availability of flexible and cost-effective tools that can perform combinatorial and high-throughput genotyping of single-nucleotide polymorphisms(SNPs)to guide the crop development *** this end,we have developed a programmable,droplet-based microfluidic device for genotyping maize genomic DNA.A unique feature of the microfluidic platform is the nano sample processors(NSPs),which allow the device to sequentially load an unrestricted number of unique DNA samples using only two inlets,overcoming the current limitation to the number of sample inputs due to small device *** and programmable droplet generation within the device allows each sample to be genotyped against a panel of markers on ***,we have successfully implemented the Invader assay for SNP genotyping in flowing,50-nL droplets,thus achieving significant reduction in consumption of reagents per reaction as compared with conventional genotyping *** a demonstration,we performed 240 Invader reactions(testing 8 DNA samples against 10 SNP markers)and achieved greater than 93% accuracy in SNP calling of plant DNA samples in a single droplet-based experiment.

Ultra-fast densification of cNTs reinforced alumina based on combustion reaction and quick pressing

An ultra-fast densification method based on high heating rate from the combustion reaction for producing carbon nanotubes （cNTs） reinforced alumina ceramics is reported. The heat generated by combustion reaction is adopted to act as a high temperature source to the sample, which results in a heating rate of 1660℃/min of the sample. Then a great mechanical pressure is applied to the sample when the sample gets the expected temperature. The densification process is finished in several minutes. The results indicate that the densification method is beneficial to protect the cNTs from destruction and creates good interracial combination between the nanotubes and the matrix. With the addition of 1 wt% cNTs, the fracture toughness of the ceramics prepared increases about 50%.

Analysis of Transcriptome changes Induced by Ptr ToxA in Wheat Provides Insights into the Mechanisms of Plant Susceptibility

To obtain greater insight into the molecular events underlying plant disease susceptibility, we studied transcriptome changes induced by a host-selective toxin of Pyrenophora tritici-repentis, Ptr ToxA （ToxA）, on its host plant, wheat. Transcriptional profiling of ToxA-treated leaves of a ToxA-sensitive wheat cultivar was performed using the Genechip Wheat Genome Array. An improved and up-to-date annotation of the wheat microarray was generated and a new tool for array data analysis （BRAT） was developed, and both are available for public use via a web-based interface. Our data indicate that massive transcriptional reprogramming occurs due to ToxA treatment, including cellular responses typically associated with defense. In addition, this study supports previous results indicating that ToxA-induced cell death is triggered by impairment of the photosynthetic machinery and accumulation of reactive oxygen species. Based on results of this study, we propose that ToxA acts as both an elicitor and a virulence factor.

Optimization for microwave-assisted direct liquefaction of bamboo residue in glycerol/methanol mixtures

Bamboo residues were liquefied in a mixture of glycerol and methanol in the presence of sulfuric acid using microwave energy. We investigated the effects of liquefaction conditions, including glycerol/methanol ratio, liquefaction temperature, and reaction time on the conversion yield. The optimal liquefaction conditions were under the temperature of 120 °c, the reaction time of 7 min, the glycerol–methanol–bamboo ratio of 8/0/2（W/W）, and the microwave power of 300 W. Maximum conversion yield was 96.7 %. The liquid products were separated into two contents（water soluble part and precipitate part） by addition of a sufficient amount of water. By Fourier transform infrared（FT-IR）, the water soluble content mainly contained glycerol and its derivate and carbohydrate degradation products, and the precipitate content was mainly lignin derivatives.

comparison of five video-assisted intubation devices by novice and expert laryngoscopists for use in the aeromedical evacuation environment

Background: The critically ill or injured patient undergoing military medical evacuation may require emergent intubation. Intubation may be life-saving, but it carries *** novice or infrequent laryngoscopist has a distinct disadvantage because experience is critical for the rapid and safe establishment of a secured airway. This challenge is compounded by the austere environment of the back of an aircraft under blackout conditions. This study determined which of five different video-assisted intubation devices(VAIDs) was best suited for in-flight use by U.S. Air Force critical care Air Transport Teams by comparing time to successful intubation between novice and expert laryngoscopists under three conditions, Normal Airway Lights on(NAL), Difficult Airway Lights on(DAL) and Difficult Airway Blackout(DAB), using manikins on a standard military transport stanchion and the floor with a minimal amount of setup time and extraneous light ***: A convenience sample size of 40 participants(24 novices and 16 experts) attempted intubation with each of the 5 different video laryngoscopic devices on high-fidelity airway manikins. Time to tracheal intubation and number of optimization maneuvers used were recorded. Kruskal-Wallis testing determined significant differences between the VAIDs in time to intubation for each particular scenario. Devices with significant differences underwent pair-wise comparison testing using rank-sum analysis to further clarify the difference. Device assembly times, startup times and the amount of light emitted were recorded. Perceived ease of use was surveyed. Results: Novices were fastest with the Pentax AWS in all difficult airway scenarios. Experts recorded the shortest median times consistently using 3 of the 5 devices. The AWS was superior overall in 4 of the 6 scenarios tested. Experts and novices subjectively judged the Glide Scope Ranger as easiest to use. Small water bodies are critical for maintaining freshwater biodiversity,but are among the least investigated aquatic *** examined physical and chemical variables at two reaches in Arroyo Tambillos,a small,high-elevation Andean stream,in NW Mendoza province,Argentina,across four seasons to examine how local factors and seasonality affected the structure of the macroinvertebrate *** Arroyo Tambillos community was numerically dominated by ephemeropterans(mainly Massartellopsis irarrazavali)while the dipteran chironomidae were the most species *** macroinvertebrate abundance was highest in summer,driven mainly by taxa that were secondary in dominance(Austrelmis *** Andesiops peruvianus),while richness did not differ between ***,benthic composition was different in Fall 2013(March)compared to other seasons,largely because of the increased abundance of *** correspondence analysis discriminated chironomidae species distributions in Fall 2013 by substrate type(i.e.,big and small boulder substrate).By contrast,discharge,velocity,and depth were the variables which most affected the macroinvertebrate abundance and *** to our expectations,most community changes observed occurred in fall instead of ***-kryal stream communities like the one described here have become increasingly important for conserving mountain stream biodiversity as anthropogentic impacts and climate change increasingly impact lower stream ***,monitoring high-altitude streams like the Arroyo Tambillos may be critical for preventing the future loss of unique and sensitive stream biota.
