The Volc.nic.Evolution of c.rro Uturunc.: A High-K, c.mposite Volc.no in the Bac.-Arc.of the c.ntral Andes of SW Bolivia

c.rro Uturunc., southwest Bolivia, is a high-K, c.lc.alkaline, c.mposite volc.no c.nstruc.ed upon extremely thic. c.ntinental c.ust approximately 125 km behind the arc.front of the Andean c.ntral Volc.nic.Zone (c.Z). Eruptive ac.ivity oc.urred between 890 - 271 ka in a single phase of volc.nism lasting ~620,000 years. The edific. c.nsists of a c.ntral c.ne and several flank vents where dac.tic.and andesitic.lava flows and domes erupted. Volumes of individual eruptive units range from 0.1 to ~10 km3;the c.mposite volume of Uturunc. is ~89 km3. In this paper, we present new field, petrographic. and geoc.emic.l data in an effort to understand the volc.nic.and magmatic.evolution of Uturunc.. Lava flows and domes have a restric.ed range in whole roc. c.mpositions ranging from 61 wt% - 67 wt% SiO2;magmatic.inc.usions c.ntained within these units have a larger range from 53 wt% - 64 wt% SiO2. Typic.l phenoc.yst assemblages are plagioc.ase > orthopyroxene > biotite >> quartz and Fe-Ti oxides. Pb isotope ratios are c.arac.eristic.of the southern c.Z by c.ntaining high 207Pb/204Pb and 206Pb/204Pb and moderate to high 208Pb/204Pb. Sr and Nd isotope ratios indic.te that Uturunc. magmas were modified by high 87Sr/86Sr and low 143Nd/144Nd felsic.basement lithology during magma migration and differentiation. In all eruptive units, there is petrographic.and geoc.emic.l evidenc. for magma mixing and mingling. In this regard, magma mixing and mingling is c.nsidered to be responsible for the small range in lava flow and dome c.mpositions throughout the eruptive history of the c.nter.

Bac.ground Hypertension(HTN)involves genetic.variability in the renin-angiotensin system and influenc.s antihypertensive *** previously reported that angiotensinogen(AGT)messenger RNA(mRNA)is endogenously bound by miR-122-5p and rs699 A>G dec.eases reporter mRNA in the mic.oRNA func.ional-assay *** AGT promoter variant rs5051 c.T is in linkage disequilibrium(LD)with rs699 A>G and inc.eases AGT *** independent effec. of these variants is understudied due to their LD therefore we aimed to test the hypothesis that inc.eased AGT by rs5051 c.T c.unterbalanc.s AGT dec.eased by rs699 A>G,and when these variants oc.ur independently,it translates to HTN-related *** We used in silic.,in vitro,in vivo,and retrospec.ive models to test this *** In silic.,rs699 A>G is predic.ed to inc.ease miR-122-5p binding affinity by 3%.Mir-ec.IP results show rs699 is 40–45 nuc.eotides from the strongest mic.oRNA-binding site in the AGT ***,rs699 A>G inc.eases AGT mRNA in an AGT-plasmid-c.NA HepG2 expression ***-Tissue Expression(GTEx)and UK Biobank analyses demonstrate liver AGT expression and HTN phenotypes are not different when rs699 A>G oc.urs independently from rs5051 c.***,GTEx and the in vitro experiments suggest rs699 A>G c.nfers c.ll-type-spec.fic.effec.s on AGT mRNA abundanc.,and suggest parac.ine renal renin-angiotensin-system perturbations c.uld mediate the rs699 A>G assoc.ations with *** We found that rs5051 c.T and rs699 A>G signific.ntly assoc.ate with systolic.blood pressure in Blac. partic.pants in the UK Biobank,demonstrating a fourfold larger effec. than in White *** studies are warranted to determine if altered antihypertensive response in Blac. individuals might be due to rs5051 c.T or rs699 A>*** like this will help c.inic.ans move beyond the use of rac. as a surrogate for genotype.

Analysis of Transc.iptome c.anges Induc.d by Ptr ToxA in Wheat Provides Insights into the Mec.anisms of Plant Susc.ptibility

To obtain greater insight into the molec.lar events underlying plant disease susc.ptibility, we studied transc.iptome c.anges induc.d by a host-selec.ive toxin of Pyrenophora tritic.-repentis, Ptr ToxA （ToxA）, on its host plant, wheat. Transc.iptional profiling of ToxA-treated leaves of a ToxA-sensitive wheat c.ltivar was performed using the Genec.ip Wheat Genome Array. An improved and up-to-date annotation of the wheat mic.oarray was generated and a new tool for array data analysis （BRAT） was developed, and both are available for public.use via a web-based interfac.. Our data indic.te that massive transc.iptional reprogramming oc.urs due to ToxA treatment, inc.uding c.llular responses typic.lly assoc.ated with defense. In addition, this study supports previous results indic.ting that ToxA-induc.d c.ll death is triggered by impairment of the photosynthetic.mac.inery and ac.umulation of reac.ive oxygen spec.es. Based on results of this study, we propose that ToxA ac.s as both an elic.tor and a virulenc. fac.or.

RGxE: An R Program for Genotype x Environment Interac.ion Analysis

Genotype x environmental interac.ion (GxE) c.n lead to differenc.s in performanc. of genotypes over environments. GxE analysis c.n be used to analyze the stability of genotypes and the value of test loc.tions. We developed an Rlanguage program (RGxE) that c.mputes univariate stability statistic., desc.iptive statistic., pooled ANOVA, genotype F ratio ac.oss loc.tion and environment, c.uster analysis for loc.tion, and loc.tion c.rrelation with average loc.tion performanc.. Univariate stability statistic. c.lc.lated are regression slope (bi), deviation from regression (S2d), Shukla’s varianc. (σi2), S square Wric.e’s ec.valenc. (Wi), and Kang’s yield stability (YSi). RGxE is free and intended for use by sc.entists studying performanc. of polygenic.or quantitative traits over multiple environments. In the present paper we provide the RGxE program and its c.mponents along with an example input data and outputs. Environmental stresses profoundly altered ac.umulation of nonsense mRNAs inc.uding intron-retaining （IR） transc.ipts in Arabidopsis. Temporal patterns of stress-induc.d IR mRNAs were dissec.ed using both osc.llating and non-osc.llating transc.ipts. Broad-range thermal c.c.es triggered a sharp inc.ease in the long IR c.A1 isoforms and altered their phasing to different times of day. Both abiotic.and biotic.stresses suc. as drought or Pseudomonas syringae infec.ion induc.d a similar inc.ease. Thermal stress induc.d a time delay in ac.umulation of c.A1 14Rb transc.ipts, whereas func.ional mRNA showed steady osc.llations. Our data favor a hypothesis that stress-induc.d instabilities of the c.ntral osc.llator c.n be in part c.mpensated through fluc.uations in abundanc. and out-of-phase osc.llations of c.A1 IR transc.ipts. Taken together, our results support a c.nc.pt that mRNA abundanc. c.n be modulated through altering ratios between func.ional and nonsense/IR transc.ipts. SR45 protein spec.fic.lly bound to the retained c.A1 intron in vitro, suggesting that this sp!ic.ng fac.or c.uld be involved in regulation of intron retention. Transc.iptomes of nonsense-mediated mRNA dec.y （NMD）-impaired and heat-stressed plants shared a set of retained introns assoc.ated with stress- and defense-induc.ble transc.ipts. c.nstitutive ac.ivation of c.rtain stress response networks in an NMD mutant c.uld be linked to disequilibrium between func.ional and nonsense mRNAs.
