Seasonal prediction of extreme high-temperature days over the Yangtze River basin

Extreme high temperatures occur frequently over the densely populated Yangtze River basin(YRB)in China during summer,significantly impacting the local economic development and ecological system.However,accurate prediction of extreme high-temperature days in this region remains a challenge.Unfortunately,the Climate Forecast System Version 2(CFSv2)exhibits poor performance in this regard.Thus,based on the interannual increment approach,we develop a hybrid seasonal prediction model over the YRB(HM_(YRB))to improve the prediction of extreme high-temperature days in summer.The HM_(YRB)relies on the following four predictors:the observed preceding April-May snowmelt in north western Europe;the snow depth in March over the central Siberian Plateau;the CFSv2-forecasted concurrent summer sea surface temperatures around the Maritime Continent;and the 200-hPa geopotential height over the Tibetan Plateau.The HM_(YRB)indicates good capabilities in predicting the interannual variability and trend of extreme high-temperature days,with a markable correlation coefficient of 0.58 and a percentage of the same sign(PSS)of 76% during 1983-2015 in the one-year-out cross-validation.Additionally,the HM_(YRB) maintains high PSS skill(86%)and robustness in the independent prediction period(2016-2022).Furthermore,the HM_(YRB) shows a good performance for years with high occurrence of extreme high-temperature days,with a hit ratio of 40%.These predictors used in HM_(YRB)are beneficial in terms of the prediction skill for the average daily maximum temperature in summer over the YRB,albeit with biases existing in the magnitude.Our study provides promising insights into the prediction of 2022-like hot extremes over the YRB in China.

Human monoamine oxidase B(hMAO-B)has emerged as a pivotal therapeutic target for Parkinson's disease.Due to adverse effects and shortage of commercial drugs,there is a need for novel,highly selective,and reversible hMAO-B inhibitors with good blood-brain barrier permeability.In this study,a high-throughput at-line nanofractionation screening platform was established with extracts from Chuanxiong Rhizoma,which resulted in the discovery of 75 active compounds,including phenolic acids,volatile oils,and phthalides,two of which were highly selective novel natural phthalide hMAO-B inhibitors that were potent,selective,reversible and had good blood‒brain permeability.Molecular docking and molecular dynamics simulations elucidated the inhibition mechanism.Sedanolide(IC_(50)=103 nmol/L;SI=645)and neocnidilide(IC_(50)=131 nmol/L;SI=207)demonstrated their excellent potential as hMAO-B inhibitors.They offset the limitations of deactivating enzymes associated with irreversible hMAO-B inhibitors such as rasagiline.In SH-SY5Y cell assays,sedanolide(EC_(50)=0.962μmol/L)and neocnidilide(EC_(50)=1.161μmol/L)exhibited significant neuroprotective effects,comparable to the positive drugs rasagiline(EC_(50)=0.896μmol/L)and safinamide(EC_(50)=1.079μmol/L).These findings underscore the potential of sedanolide as a novel natural hMAO-B inhibitor that warrants further development as a promising drug candidate.

Mg-doped manganese oxide octahedral molecular sieve(Mg-OMS-2)catalysts were prepared by hydrothermal method.The photothermal degradation performance of these catalysts for formaldehyde(HCHO)in batch system and continuous system was investigated.The light absorption of OMS-2 was increased by Mg-doped,especially for near infrared light,which promoted surface temperature reach a maximum of 214.8℃ under xenon irradiation.At this temperature,the reinforced surface lattice oxygen and oxygen vacancy that formed by lattice distortion via Mg-doped were activated.The best HCHO elimination efficiency was achieved over Mg_(0.2)/OMS-2 catalyst with Mg^(2+)/Mn^(2+)=1/5,which could reduce HCHO from 250 ppm to 10 ppm within 20 min.The in situ DRIFTS was also carried out to monitor the changes in the content of reaction intermediates and analyze the degradation paths of HCHO.It was found the HCHO was attacked by formed·OH and·O^(2-) to generate formate species and carbonate species,and finally transformed to CO_(2) and H_(2)O.This photothermal catalytic oxidation process exhibited a high efficiency purification of HCHO without the help of extra energy consumption.

SEC1A and SEC6 synergistically regulate pollen tube polar growth

Pollen tube polar growth is a key physiological activity for angiosperms to complete double fertilization, which is highly dependent on the transport of polar substances mediated by secretory vesicles.The exocyst and Sec1/Munc18(SM) proteins are involved in the regulation of the tethering and fusion of vesicles and plasma membranes, but the molecular mechanism by which they regulate pollen tube polar growth is still unclear. In this study, we found that loss of function of SEC1A, a member of the SM protein family in Arabidopsis thaliana, resulted in reducing pollen tube growth and a significant increase in pollen tube width. SEC1A was diffusely distributed in the pollen tube cytoplasm, and was more concentrated at the tip of the pollen tube. Through coimmunoprecipitation-mass spectrometry screening,protein interaction analysis and in vivo microscopy,we found that SEC1A interacted with the exocyst subunit SEC6, and they mutually affected the distribution and secretion rate at the tip of the pollen tube. Meanwhile, the functional loss of SEC1A and SEC6 significantly affected the distribution of the SNARE(soluble N-ethylmaleimide-sensitive factor attachment protein receptor) complex member SYP125 at the tip of the pollen tube, and led to the disorder of pollen tube cell wall components. Genetic analysis revealed that the pollen tube-related phenotype of the sec1a sec6 double mutant was significantly enhanced compared with their respective single mutants. Therefore, we speculated that SEC1A and SEC6 cooperatively regulate the fusion of secretory vesicles and plasma membranes in pollen tubes, thereby affecting the length and the width of pollen tubes.

Tsunami hazard and mitigation analysis for bathing beaches in China based on numerical simulations

Bathing beaches are usually the first to suffer disasters when tsunamis occur,owing to their proximity to the sea.Several large seismic fault zones are located off the coast of China.The impact of each tsunami scenario on Chinese bathing beaches is different.In this study,numerical models of the worst tsunami scenarios associated with seismic fault zones were considered to assess the tsunami hazard of bathing beaches in China.Numerical results show that tsunami waves from the Pacific Ocean could affect the East China Sea coast through gaps between the Ryukyu Islands.The Zhejiang and Shanghai coasts would be threatened by a tsunami from Ryukyu Trench,and the coasts of Hainan and Guangdong provinces would be threatened by a tsunami from the Manila Trench.The tsunami hazard associated with the Philippine Trench scenario needs particular attention.Owing to China’s offshore topography,the sequential order of tsunami arrival times to coastal provinces in several tsunami scenarios is almost the same.According to the tsunami hazard analysis results,Yalongwan Beach and eight other bathing beaches are at the highest hazard level.A high-resolution numerical calculation model was established to analyze the tsunami physical characteristics for the high-risk bathing beaches.To explore mitigating effects of a tsunami disaster,this study simulated tsunami propagation with the addition of seawalls.The experimental results show that the tsunami prevention seawalls constructed in an appropriate shallow water location have some effect on reducing tsunami hazard.Seawalls separated by a certain distance work even better.The analysis results can provide a scientific reference for subsequent preventive measures such as facility construction and evacuation.

Background Studying the composition and developmental mechanisms in mammary gland is crucial for healthy growth of newborns. The mammary gland is inherently heterogeneous, and its physiological function dependents on the gene expression of multiple cell types. Most studies focused on epithelial cells, disregarding the role of neighboring adipocytes.Results Here, we constructed the largest transcriptomic dataset of porcine mammary gland cells thus far. The dataset captured 126,829 high-quality nuclei from physiological mammary glands across five developmental stages(d 90 of gestation, G90;d 0 after lactation, L0;d 20 after lactation, L20;2 d post natural involution, PI2;7 d post natural involution, PI7). Seven cell types were identified, including epithelial cells, adipocytes, endothelial cells, fibroblasts cells, immune cells, myoepithelial cells and precursor cells. Our data indicate that mammary glands at different developmental stages have distinct phenotypic and transcriptional signatures. During late gestation(G90), the differentiation and proliferation of adipocytes were inhibited. Meanwhile, partly epithelial cells were completely differentiated. Pseudo-time analysis showed that epithelial cells undergo three stages to achieve lactation, including cellular differentiation, hormone sensing, and metabolic activation. During lactation(L0 and L20), adipocytes area accounts for less than 0.5% of mammary glands. To maintain their own survival, the adipocyte exhibited a poorly differentiated state and a proliferative capacity. Epithelial cells initiate lactation upon hormonal stimulation. After fulfilling lactation mission, their undergo physiological death under high intensity lactation. Interestingly, the physiological dead cells seem to be actively cleared by immune cells via CCL21-ACKR4 pathway. This biological process may be an important mechanism for maintaining homeostasis of the mammary gland. High-performance Cu/Graphene composite wire synergistically strengthened by nano Cr_(3)C_(2) phase was directly synthesized via hot press sintering followed by severe cold plastic deformation, using liquid paraffin and CuCr alloy powder as the raw materials. Since graphene is in situ formed under the catalysis of copper powder during the sintering process, the problem that graphene is easy to agglomerate and difficult to disperse uniformly in the copper matrix has been solved. The nano Cr_(3)C_(2)-particles nailed at the interface favor to improve the interface bonding. The Cu/Graphene composite possesses high electrical conductivity, hardness, and plasticity. The composite wire exhibits high electrical conductivity of 96.93% IACS, great tensile strength of 488MPa, and excellent resistance to softening. Even after annealing at 400℃ for 1 h, the tensile strength can still reach 268 MPa with a conductivity of about 99.14% IACS.The wire's temperature coefficient of resistance(TCR) is largely reduced to 0.0035/℃ due to the complex structure,which leads the wire to present low resistivity at higher temperatures. Such Cu/Graphene composite wire with excellent comprehensive performance has a good application prospect in high-power density motors.

The occurrence of benign prostate hyperplasia(BPH)was related to disrupted sex steroid hormones,and metformin(Met)had a clinical response to sex steroid hormone-related gynaecological disease.However,whether Met exerts an antiproliferative effect on BPH via sex steroid hormones remains unclear.Here,our clinical study showed that along with prostatic epithelial cell(PEC)proliferation,sex steroid hormones were dysregulated in the serum and prostate of BPH patients.As the major contributor to dysregulated sex steroid hormones,elevated dihydrotestosterone(DHT)had a significant positive relationship with the clinical characteristics of BPH patients.Activation of adenosine 5'-monophosphate(AMP)-activated protein kinase(AMPK)by Met restored dysregulated sex steroid hormone homeostasis and exerted antiproliferative effects against DHT-induced proliferation by inhibiting the formation of androgen receptor(AR)-mediated Yes-associated protein(YAP1)-TEA domain transcription factor(TEAD4)heterodimers.Met’s anti-proliferative effects were blocked by AMPK inhibitor or YAP1 overexpression in DHT-cultured BPH-1 cells.Our findings indicated that Met would be a promising clinical therapeutic approach for BPH by inhibiting dysregulated steroid hormone-induced PEC proliferation.

At 12:15 on January 15,2022(Beijing time),a massive eruption of the Hunga Tonga-Hunga Ha'apai volcano produced violent atmospheric fluctuations,which in turn generated a global tsunami through an abrupt air pressure shock upon the sea surface.Two main components of tsunami waves,phase-locked waves and free gravity waves,were identified by significant differences in propagating speeds across the deep ocean.The phase-locked wave propagated through the ocean basin synchronously with the atmospheric Lamb wave at an average speed of approximately 306 m/s,followed by the free gravity wave at a slower speed.The locked wave reached the coast of eastern Taiwan Island at about 20:00 on January 15,in coincidence with the Lamb wave arrival.However,on the coast of Chinese mainland,tidal gauges did not record tsunami signals until at least 2 h after the Lamb wave arrivals.Theoretical analyses and numerical experiments both suggested that as a result of the incoming wave shoaling above the vast continental shelf of Chinese mainland,the locked wave was no longer trapped by the air pressure shock and gradually transformed into freely-propagating shallow water waves by slowing down its propagation.Due to the longlasting planetary atmospheric fluctuations circling the earth many times,the sea level oscillations continuously propagated onto the Chinese shelf,which resulted in the tsunami waves excited along the Chinese coasts for at least 36 h.The maximum wave amplitude recorded on the coast of eastern Taiwan Island was 44 cm at Wushi,while on the coasts of eastern and southern Chinese mainland,the maximum amplitudes were 22 cm at Shipu and 13 cm at Zhuhai.Fourier and wavelet analyses were performed to identify the major components of the tsunami waves on the Chinese coasts.The results indicated that eastern Taiwan Island was impacted mainly by the waves with periods of approximately 10-40 min.Chinese mainland was hit by the evolved shallow water waves and subsequent free waves,with periods of approximately 40-100 and 16-20 min,resp
