1H-NMR based metabolomics approach to evaluate effect of Cydonia oblanga Miller total flavonoid on atherosclerosis apoE(-/-) mice

OBJECTIVE ~1H-NMR-based metabolomics approach was conducted to holistically explore the effect and mechanisms of Cydonia oblanga Mill flavonoids(COMF) on high-fat diet induced Atherosclerosis(AS) apoE-/-*** AS model was established on the apolipoprotein e knockout mice by high-fat *** ApoE-/-mice were split into 6 groups including control group,AS model group,COMF High dose(COMF-H) group,COMF medium dose(COMF-M) group,COMF Low dose(COMF-L) group and Simvastatin group as the positive control *** samples from all groups were analyzed by ~1H-NMR technology and the OPLS-DA was conducted to distinguish the metabolic *** Compared to the control group,serum levels of cholesterol,VLDL,leucine,isoleucine,valine,blood lipid,citrulline,methylamine,glucose,glycine,glycerol,myo-inositol,fructose,phenylalanine,unsaturated lipid,urea and other metabolites content significantly increased,while HDL,lactate,alanine,glutamate,glutamine,pyruvate,carnitine,citrate,choline content *** decreased and the difference was statistically significant(P<0.05).The trend of metabolites in serum samples of COMF low,medium and high group was opposite to that of atherosclerosis model group and the difference was statistically significant(P<0.05).CONCLUSION Through functional analysis of these biomakers,amino acid metabolism,lipid metabolism,cholesterol metabolism,energy metabolism and inflammation reaction were considered as the most relevant pathological biomakers in the serum of AS *** study also demonstrates that COMF had the therapeutic effectiveness on AS through partly reversing the lipid,cholesterol,amino acid,energy metabolism and Inflammation reaction.

Biodegrading Wheat Bran with Agaricus blazei and Its Effects on Intestinal Development Identified with Mice

Agaricus blazei Murill is a well known edible/medicinal mushroom used for immune-nutrient food *** biodegradation of different substrates with this fungus may result in different metabolites and degraded compounds,which may enrich the function of the food *** this study,soluble compounds from the culture of *** grown in liquid media with whole wheat bran and its water filtrate,respectively,were *** soluble sugar,arabinoxylan,protein,and amino acids were significantly higher in the fungal culture resulted from the medium with whole wheat bran （43.54,1.56,0.59,and 2.19 mg mL^-1,respectively） than that from the medium with the bran filtrate （17.28,0.37,0.13,and 1.13 mg mL^-1,respectively）.The biodegraded wheat bran with cultured mycelia was fed to Mus musculus Linnaeus as supplementation or as dietary fiber exclusive *** non-specific food therapies,feeding effects on mice intestinal development were indicated indirectly in growth performance,intestinal absorption and serum *** to feeding uncultured wheat bran,feeding mice with culture mixes of *** resulted in remarkably increase in villus height and villus height/crypt depth ratio,which were increased by 25.4 and 31.0%,respectively,when applied as supplementation,and by 44.3 and 43.4%,respectively,when applied as dietary *** increases are concomitant with the higher level of D-xylose in blood serum about 16.9 and 29.2% as supplementation and dietary fiber,*** results implying that culturing *** with whole wheat bran enhanced extracellular metabolism of the fungus and extensive degradation of wheat bran insoluble fibrous ***,feeding the culture mix including metabolites and degraded wheat bran improved intestinal villus development,proving the nutritional benefits of the *** mycelial cultures.

QL1706 (anti-PD-1 IgG4/CTLA-4 antibody) plus chemotherapy with or without bevacizumab in advanced non-small cell lung cancer: a multi-cohort, phase II study

First-line chemoimmunotherapy(with or without bevacizumab)has improved outcomes in advanced non-small cell lung cancer(NSCLC).Here,this open-label,multi-cohort phase II study(NCT05329025)was done to investigate the safety and efficacy of QL1706(a single bifunctional MabPair product against PD-1 and CTLA-4)and chemotherapy with or without bevacizumab in this *** were enrolled into five different cohorts based on genotype(cohorts 1-4,epidermal growth factor receptor[EGFR]wild-type;cohort 5,EGFR-mutant and progressed on EGFR-tyrosine kinase inhibitors[TKIs]).Between June 11,2021 and December 29,2021,91 patients were *** frequent treatment-related adverse events(TRAEs)included decreased appetite(60[65.9%]),anemia(60[65.9%]),infusion-related reactions(48[52.7%]),and pruritus(44[48.4%]).Grade≥3 TRAEs occurred in 30(33.0%)***-seven(45%)patients with wild-type EGFR achieved partial response(PR)(objective response rate[ORR]=45%)and had a median progression-free survival(mPFS)of 6.8 months(95%CI:5.2-9.7).For 31 patients harboring mutated EGFR,17(54.8%)achieved PR(ORR=54.8%),with an mPFS of 8.5 months(95%CI:5.72-not evaluable).Overall,QL1706 plus chemotherapy,regardless of having bevacizumab,was generally tolerable and had promising antitumor activity for EGFR wild-type advanced NSCLC in first-line ***,QL1706 plus chemotherapy and bevacizumab showed favorable antitumor activity for patients who had EGFR mutated NSCLC but failed in TKI therapy,demonstrating a potential for treating this population.

Tissue elasticity and viscosity are always associated with pathological *** a new imaging method,ultrasound vibro-acoustic imaging is developed for quantitatively measuring tissue elasticity and viscosity which have important significance in early diagnosis of *** paper developed an ultrasound vibro-acoustic imaging research platform mainly consisting of excitation part and detection *** excitation transducer was focused at one location within the medium to generate harmonic vibration and shear wave propagation,and the detection transducer was applied to detect shear wave at other locations along shear wave propagation path using pulse-echo *** received echoes were amplified,filtered,digitized and then processed by Kalman filter to estimate the vibration *** to the phase changes between different propagation locations,we estimated the shear wave speed,and then used it to calculate the tissue elasticity and *** phantom experiments based on this platform show results of phantom elasticity and viscosity close to literature *** experiments are now in progress to obtain quantitative elasticity and viscosity in vitro tissue.

Uric formaldehyde levels are negatively correlated with cognitive abilities in healthy older adults

Recent studies have shown that the abnormal accumulation of endogenous formaldehyde could be a critical factor in age-related cognitive decline. The aim of this study was to estimate the correlation between uric formaldehyde and general cognitive abilities in a community-based elderly population, and to measure the extent and direction in which the correlation varied with demographic characteristics. Using a double-blind design, formaldehyde in human urine was analyzed by high-performance liquid chromatography （n = 604）, and general cognitive abilities were measured using the Montreal Cognitive Assessment （MoCA）. Demographic characteristics, in terms of age, gender, residential region, and education were taken into consideration. We found that uric formaldehyde levels were inversely correlated with the MoCA score, and the concentration varied with demographic features： higher odds of a high formaldehyde level occurred among the less educated and those living in old urban or rural areas. In cytological experiments, the level of cellular formaldehyde released into the medium increased as SH-SY5Y and BV2 cells were incubated for three days. Formaldehyde in excess impairedthe processes of N2a cells and neurites of primary cultured rat hippocampal cells. However, removal of formaldehyde markedly rescued and regenerated the processes of N2a cells. These results demonstrated a negative correlation between the endogenous formaldehyde and general cognitive abilities. High formaldehyde levels could be a risk factor for cognitive impairment in older adults, and could be developed as a non-invasive marker for detection and monitoring of age-related cognitive impairment.

To establish the relationship among reservoir characteristics and rock physical parameters,we construct the well-bore rock physical models firstly,considering the influence factors,such as mineral composition,shale content,porosity,fluid type and *** with analyzing the change rules of elastic parameters along with the above influence factors and the cross-plots among elastic parameters,the sensitive elastic parameters of tight sandstone reservoir are determined,and the rock physics template of sweet spot is constructed to guide pre-stack seismic *** results show that velocity ratio and Poisson impedance are the most sensitive elastic parameters to indicate the lithologic and gas-bearing properties of sweet spot in tight sandstone *** high-quality sweet spot is characterized by the lower velocity ratio and Poisson ***,the actual seismic data are selected to predict the sweet spots in tight sandstone gas reservoirs,so as to verify the validity of the rock physical simulation *** significant consistency between the relative logging curves and inversion results in different wells implies that the utilization of well-bore rock physical simulation can guide the prediction of sweet spot in tight sandstone gas reservoirs.

Environmental exposure to crystalline silica particles can lead to silicosis, which is one of the most serious pulmonary interstitial fibrosis around the world. Unfortunately, the exact mechanism on silicosis is unclear, and the effective treatments are lacking to date. In this study, we aim to explore the molecular mechanism by which interleukin-11(IL-11) affects silica particles-induced lung inflammation and fibrosis. We observed that IL-11 expressions in mouse lungs were significantly increased after silica exposure, and maintained at high levels across both inflammation and fibrosis phase. Immunofluorescent dual staining further revealed that the overexpression of IL-11 mainly located in mouse lung epithelial cells and fibroblasts. Using neutralizing anti-IL-11 antibody could effectively alleviate the overexpression of pro-inflammatory cytokines(i.e., interleukin-6 and tumor necrosis factor-α) and fibrotic proteins(i.e., collagen type I and matrix metalloproteinase-2) induced by silica particles. Most importantly, the expressions of IL-11 receptor subunit α(IL-11Rα), Glycoprotein130(GP130), and phosphorylated extracellular signal-regulated kinase(p-ERK) were significantly increased in response to silica, whereas blocking of IL-11 markedly reduced their levels. All findings suggested that the overexpression of IL-11 was involved in the pathological of silicosis, while neutralizing IL-11 antibody could effectively alleviate the silica-induced lung inflammation and fibrosis by inhibiting the IL-11Rα/GP130/ERK signaling pathway. IL-11 might be a promising therapeutic target for lung inflammation and fibrosis caused by silica particles exposure.

Enhanced mechanical properties and formability of 316L stainless steel materials 3D-printed using selective laser melting

Pseudo-Bonding Interaction between Boron-doped Heterofullerene and Zinc Porphine Predicted by DFT Calculation

Theoretical study on the supramolecular complexes formed between boron-doped het- erofullerene （C59B） and zinc porphine （ZnF）, namely C59B-ZnP and its anion species C59B-ZnP, was performed by density functional theory calculation at wB97XD/6-31G（d） level. Strong interaction between porphyrin and heterofullerene moiety was predicted for these complexes based on geometry and electronic structure analysis. Especially, pseudobonding interaction occurring between the B atom of fullerene and the N atom of porphyrin was predicted to occur in C59B-ZnP complex, but be broken in C59B-ZnP complex. Time-dependent density functional theory calculation manifests the redshift of electron absorption for ZnP upon the interaction with heterofullerene.
