Amino acids are naturally occurring compounds in many edible or medicinal plants,which possess a variety of nutraceutical effects in ***,a method of ultrasound-assisted extraction was optimized using Box-Behnken design of response surface methodology in maximizing the yield of free amino acids deriving from flowers of Apocynum venetum *** the optimal condition of ultrasound-assisted extraction,i.e.187.5 W of ultrasonic power,31.93 min of extraction time,and 0.47 mg/ml of solid-liquid ratio,the experimental yield of extractive was 287.17±0.205 mg/g,which was in close agreement with the value,as predicted by the established *** addition,a hydrophilic interaction ultra-high performance liquid chromatography coupled with QqQ-MS/MS method was developed and validated for simultaneous quantification of 20 types of amino acids without derivatization contained in *** *** analytical method was validated by matrix effect,linearity,limit of detection,limit of quantification,precision,repeatability,stability,and *** analysis results showed that *** flower was rich in free amino acids of~3%of total dried weight,and which contained leucine(13.7 g/mg),isoleucine(7.9 g/mg),and lysine(2.2 g/mg)as the most abundant amino ***,*** flower could provide beneficial nutrient values for human health,*** weight maintenance or glucose homeostasis.

Introduction Patients with stroke-like symptoms may be underutilising emergency medical services and avoiding hospitalisation during the COVID-19 *** investigated a decline in admissions for stroke and transient ischaemic attack(TIA)and emergency department(ED)stroke alert *** We retrospectively compiled total weekly hospital admissions for stroke and TIA between 31 December 2018 and 21 April 2019 versus 30 December 2019 and 19 April 2020 at five US tertiary academic comprehensive stroke centres in cities with early COVID-19 outbreaks in Boston,New York City,Providence and *** collected available data on ED stroke alerts,stroke severity using the National Institutes of Health Stroke Scale(NIHSS)and time from symptom onset to hospital *** Compared with 31 December 2018 to 21 April 2019,a decline in stroke/TIA admissions and ED stroke alerts occurred during 30 December 2019 to 19 April 2020(p trendr each).The declines coincided with state stay-at home recommendations in late *** greatest decline in hospital admissions was observed between 23 March and 19 April 2020,with a 31%decline compared with the corresponding weeks in *** of the five centres with 2019 and 2020 stroke alert data had a 46%decline in ED stroke alerts in late March and April 2020,compared with *** baseline NIHSS during these 4 weeks was 10 in 2020 and 7 in *** was no difference in time from symptom onset to hospital *** At these five large academic US hospitals,admissions for stroke and TIA declined during the COVID-19 *** was a trend for fewer ED stroke alerts at three of the five centres with available 2019 and 2020 *** stroke therapies are time-sensitive,so decreased healthcare access or utilisation may lead to more disabling or fatal strokes,or more severe non-neurological complications related to *** findings underscore the indirect effects of this *** health offici

OBJECTIVES:Proteins are the major component and play a key role in nutritious and therapeutic functions of edible bird’s nest(EBN);however,limited studies have been conducted on the protein due to difficulties in extraction,isolation as well as *** study aimed to provide comprehensive information for the quality evaluation of EBN peptides,which would be a valuable reference for further study on EBN ***:Here,we developed a quality control method using high performance liquid chromatography(HPlC)peptide fingerprints deriving from EBN being digested with simulated gastric *** characteristic peptide peaks were collected and identified by lC-MS/***:The characteristic peptide peaks,corresponding to the protein fragments of acidic mammalian chitinase-like,lysyl oxidase,and Mucin-5AC-like,were identified and ***,the principal component analysis indicated that the fingerprints were able to discriminate colour of EBN(white/red)and production sites(cave/house)of White EBN on the same weight *** proposed by the model developed in this study,Muc-5AC-like and AMCaselike proteins were the markers with the highest discriminative ***:The overall findings suggest that HPlC peptide fingerprints were able to clearly demonstrate peptide profile differences between genuine and adulterated EBN samples;and classify EBN samples by its color and production *** addition,the protein identification results suggested that Muc-5AC-like protein was the major protein in EBN.

lncrNA UNC5B-AS1和mir-381在非小细胞肺癌组织中的表达及临床意义

目的探讨长链非编码rNA(lncrNA)非协调性分子5同源蛋白B-反义rNA1(UNC5B-AS1)和mir-381在非小细胞肺癌(NSClC)组织中的表达及临床意义。方法采用实时荧光定量PCr(qPCr)检测NSClC患者肿瘤组织(实验组)和肺良性肿瘤组织(对照组)lncrNA UNC5B-AS1和mir-381的表达差异;分析lncrNA UNC5B-AS1和mir-381的表达与NSClC临床病理特征之间的关系;采用受试者工作特征(rOC)曲线评估lncrNA UNC5B-AS1和mir-381对NSClC的诊断价值。采用多因素Cox回归模型分析影响NSClC患者预后的独立因素。结果实验组lncrNA UNC5B-AS1的表达明显高于对照组(Pr-381的表达明显低于对照组(PlncrNA UNC5B-AS1和mir-381的表达呈明显负相关(r=-0352,P=0001)。lncrNA UNC5B-AS1、mir-381及两者联合诊断NSClC的曲线下面积(AUC)分别为0860、0850和0924。lncrNA UNC5B-AS1与NSClC患者的TNM分期、淋巴结转移及分化程度有关(Pr-381与与NSClC患者的肿瘤大小、淋巴结转移及分化程度有关(PlncrNA UNC5B-AS1高表达组患者的中位OS为269个月,明显低于低表达组的410个月(Pr-381低表达组患者的中位OS为255个月,明显低于高表达组的420个月(PlncrNA UNC5B-AS1表达及mir-381表达是影响NSClC患者预后的独立因素(PlC组织中lncrNA UNC5B-AS1表达升高,而mir-381表达下降,两者对NSClC有一定的诊断和预后预测价值。

目的:探讨乌苏酸联合吉西他滨对胰腺癌PANC-1细胞增殖和凋亡的影响。方法:以乌苏酸、吉西他滨单独用药作用于体外培养的胰腺癌细胞PANC-1细胞,采用细MTT法检测细胞半抑制浓度(IC_(50)),确定药物给药浓度;以乌苏酸(2μmol/l)、吉西他滨(0.2μmol/l)单独及联合作用于PANC-1细胞,采用锥虫蓝染色法检测PANC-1细胞存活率,PI染色后PANC-1细胞中检测细胞凋亡,采用细胞划痕实验检测PANC-1细胞迁移和伤口愈合情况,采用Western blotting检测对照组、乌苏酸组与吉西他滨组单独及联合用药组细胞中P-JNK、Bcl-2、Il-6、P-Stat 3、NF-κB和Cox-2蛋白的表达。结果:乌苏酸和吉西他滨对胰腺癌PANC-1细胞均有较强的抑制作用,其IC_(50)分别是13.67和2.78μmol/l,据此选择它们的实验浓度分别为2和0.2μmol/l。两者联合用药比分别单独用药能更显著抑制癌细胞的增殖活性[(46.47±5.07)%vs(78.38±8.65)%、(76.12±3.23)%,均Pl-2、Il-6的表达及促进P-JNK的表达。结论:乌苏酸和吉西他滨协同增强抑制胰腺癌PANC-1细胞的增殖和迁移以及促凋亡作用,其机制可能与抑制Bcl-2、Il-6、P-Stat 3和促进P-JNK蛋白表达有关。

城市景观是争议性高、往往也是最显而易见且或许是最具价值的景观设计实践。大多数项目是基于各自不同的城市背景,具有受各种社会、环境、经济因素综合影响的目标的产物。然而,这些项目通过其功能及美学特质为城市环境带来的价值直到21世纪景观设计领域绩效研究的兴起才得到证明与真正认可。本文对两个城市景观项目—位于沃斯堡的圣丹斯广场中心与位于达拉斯的AT＆T表演艺术中心的伊莱恩-查尔斯西蒙斯公园,二者是德克萨斯州北部两座最大的城市数十年来在地区尺度的代表性景观设计作品—所产生的价值进行了记述。此绩效研究是景观设计基金会案例研究调查计划于2014年完成的一项成果。本文主要侧重于记述社会绩效的指标、方法和结果,同时探索项目特质、类型,以及它们对所属城市环境的直接和间接影响。本文还在不断变化的城市景观与城市形态语境下,对这两个标志性项目的景观绩效进行了深入讨论,并向整个社会传播景观设计的社会价值。

Chronic hepatitis B virus monoinfection at a university hospital in Zambia

AIM To characterize antiviral therapy eligibility among hepatitis B virus(HBV)-infected adults at a university hospital in *** Hepatitis B surface antigen-positive adults(n = 160) who were h IV-negative and referred to the hospital after a routine or clinically-driven HBV test were enrolled. Alanine Aminotransferase(AlT),Aspartate Aminotransferase(AST),platelet count,hepatitis B e-antigen,and HBV DNA were measured. liver fibrosis/cirrhosis was assessed by physical examination,AST-to-platelet ratio index,and transient elastography. In antiviral therapy-na?ve individuals,we described hBV stages and antiviral therapy eligibility per World health Organization(WhO) and by hBV test(routine vs clinical). Elevated AlT was > 19 in women and > 30 U/l in men. Among treatmentexperienced individuals,we described medication side effects,adherence,and viral *** The median age was 33 years,71.9% were men,and 30.9% were diagnosed with HBV through a clinicallydriven test with the remainder identified via routine testing(at the blood bank,community events,etc.). Among 120 treatment-na?ve individuals,2.5% were categorized as immune tolerant,11.7% were immune active,35.6% were inactive carriers,and 46.7% had an indeterminate phenotype. Per WhO guidelines,13(10.8%) were eligible for immediate antiviral therapy. The odds of eligibility were eight times higher for those diagnosed at clinical vs routine settings(adjusted odds ratio,8.33; 95%CI: 2.26-29.41). Among 40 treatmentexperienced hBV patients,virtually all took tenofovir,and a history of mild side effects was reported in 20%. Though reported adherence was good,12 of 29(41.4%) had HBV DNA > 20 IU/m l. CONClUSION Approximately one in ten HBV-monoinfected Zambians were eligible for antivirals. Many had indeterminate phenotype and needed clinical follow-up.
