Exosomes derived from pulmonary metastatic sites enhance osteosarcoma lung metastasis by transferring the miR-194/215 cluster targeting MARCKS

Osteosarcoma,a prevalent primary malignant bone tumor,often presents with lung metastases,severely impacting patient survival *** vesicles,particularly exosomes,play a pivotal role in the formation and progression of osteosarcoma-related pulmonary ***,the communication between primary osteosarcoma and exosome-mediated pulmonary lesions remains obscure,with the potential impact of pulmonary metastatic foci on osteosarcoma progression largely *** study unveils an innovative mechanism by which exosomes originating from osteosarcoma pulmonary metastatic sites transport the miR-194/215 cluster to the primary tumor *** transportation enhances lung metastatic capability by downregulating myristoylated alanine-rich C-kinase substrate(MARCKS)*** this phenomenon,in this study we employ cationic bovine serum albumin(CBSA)to form nanoparticles(CBSA-anta-194/215)via electrostatic interaction with antagomir-miR-194/*** nanoparticles are loaded into nucleic acid-depleted exosomal membrane vesicles(anta-194/215@Exo)targeting osteosarcoma lung metastatic *** with bioengineered exosome mimetics(anta-194/215@Exo)not only impedes osteosarcoma progression but also significantly prolongs the lifespan of tumor-bearing *** findings suggest that pulmonary metastatic foci-derived exosomes initiate primary osteosarcoma lung metastasis by transferring the miR-194/215 cluster targeting MARCKS,making the miR-194/215 cluster a promising therapeutic target for inhibiting the progression of patients with osteosarcoma lung metastases.

Aqueous sodium-ion batteries are known for poor rechargeability because of the competitive water decomposition reactions and the high electrode *** have been reported by saltconcentrated and organic-hybridized electrolyte designs,however,at the expense of cost and ***,we report the prolonged cycling of ASIBs in routine dilute electrolytes by employing artificial electrode coatings consisting of NaX zeolite and NaOH-neutralized perfluorinated sulfonic *** as-formed composite interphase exhibits a molecularsieving effect jointly played by zeolite channels and size-shrunken ionic domains in the polymer matrix,which enables high rejection of hydrated Na^(+)ions while allowing fast dehydrated Na^(+)*** this coating to electrode surfaces expands the electrochemical window of a practically feasible 2 mol kg^(-1) sodium trifluoromethanesulfonate aqueous electrolyte to 2.70 V and affords Na_(2)MnFe(CN)_(6)//NaTi_(2)(PO_(4))_(3) full cells with an unprecedented cycling stability of 94.9%capacity retention after 200 cycles at 1 *** with emerging electrolyte modifications,this molecular-sieving interphase brings amplified benefits in long-term operation of ASIBs.

Most in silico evolutionary studies commonly assumed that core genes are essential for cellular function,while accessory genes are dispensable,particularly in nutrient-rich ***,this assumption is seldom tested genetically within the pangenome *** this study,we conducted a robust pangenomic Tn-seq analysis of fitness genes in a nutrient-rich medium for Sinorhizobium strains with a canonical open *** evaluate the robustness of fitness category assignment,Tn-seq data for three independent mutant libraries per strain were analyzed by three methods,which indicates that the Hidden Markov Model(HMM)-based method is most robust to variations between mutant libraries and not sensitive to data size,outperforming the Bayesian and Monte Carlo simulation-based ***,the HMM method was used to classify the fitness *** genes,categorized as essential(ES),advantage(GA),and disadvantage(GD)genes for growth,are enriched in core genes,while nonessential genes(NE)are over-represented in accessory *** ES/GA genes showed a lower fitness effect than core ES/GA *** degrees in the cofitness network decrease in the order of ES,GD,and GA/*** addition to accessory genes,1599 out of 3284 core genes display differential essentiality across test *** the pangenome core,both shared quasi-essential(ES and GA)and strain-dependent fitness genes are enriched in similar functional *** analysis demonstrates a considerable fuzzy essential zone determined by cofitness connectivity degrees in Sinorhizobium pangenome and highlights the power of the cofitness network in understanding the genetic basis of ever-increasing prokaryotic pangenome data.

The shale gas development process is complex in terms of its flow mechanisms and the accuracy of the production forecasting is influenced by geological parameters and engineering parameters. Therefore, to quantitatively evaluate the relative importance of model parameters on the production forecasting performance, sensitivity analysis of parameters is required. The parameters are ranked according to the sensitivity coefficients for the subsequent optimization scheme design. A data-driven global sensitivity analysis(GSA) method using convolutional neural networks(CNN) is proposed to identify the influencing parameters in shale gas production. The CNN is trained on a large dataset, validated against numerical simulations, and utilized as a surrogate model for efficient sensitivity analysis. Our approach integrates CNN with the Sobol' global sensitivity analysis method, presenting three key scenarios for sensitivity analysis: analysis of the production stage as a whole, analysis by fixed time intervals, and analysis by declining rate. The findings underscore the predominant influence of reservoir thickness and well length on shale gas production. Furthermore, the temporal sensitivity analysis reveals the dynamic shifts in parameter importance across the distinct production stages.

Excipients are important components of pharmaceutical preparations that affect their quality, safety, and efficacy. Macrocyclic receptors are a family of supramolecular excipients with several advantages, including molecular-level protection, small sizes,fast kinetics of host-guest recognition, and modular construction. With the continuous advances in the medical field, personalized and precision medicine requires the development of excipients with low dosages, integrated modifying effects, universality,and controlled release. To meet these requirements, we have developed a new family of macrocyclic excipients based on calixarenes by integrating their covalent(broad chemical design space) and noncovalent(wide range of substrates) ***, azocalixarenes(Azo CAs) were designed, showing high binding affinities to a broad spectrum of active pharmaceutical ingredients(APIs), selectivity to interferents, and responsiveness to hypoxic microenvironments. Due to their highly efficient and controllable recognition, Azo CAs serve as low-dose excipients for 30 APIs. Molecular encapsulation by Azo CAs results in the integrated modification of the physicochemical properties of APIs, including solubility, stability, bioavailability,and biocompatibility. Moreover, Azo CAs can be reduced by azoreductases overexpressed in hypoxic microenvironments,leading to the controlled release of APIs. Collectively, Azo CA excipients have broad application prospects for a series of diseases such as enteritis, arthritis, stroke, cancer, bacterial infection and kidney injury, with diverse therapeutic modalities,including chemotherapy, photodynamic therapy, photothermal therapy, immunotherapy, boron neutron capture therapy, radiotherapy, fluorescence imaging, and their combinations.

Experimental Investigation on Heat Transfer and Combustion of a Stirling Engine Combustor Fueled by Reformed Gas and Diesel Fuel

Thermochemical recuperation heat recovery is an advanced waste heat utilization technology that can effectively recover exhaust waste heat from oxy-fuel Stirling *** novel combustor of a Stirling engine with thermochemical recuperation heat recovery system is expected to utilize both reformed gas and diesel fuels as sources of *** this research,the effects of various factors,including the H_(2)O addition,fuel distribution ratio(FDR),excess oxygen coefficient,and cyclone structure on the temperature distribution in the combustor,combustion emissions,and external combustion system efficiency of the Stirling engine were experimentally *** the increase of steam-to-carbon ratio(S/C),the temperature difference between the upper and lower heating tubes reduces and the circumferential temperature fluctuation decreases,and the combustion of diesel and reformed gas remains close to complete *** S/C=2,the external combustion efficiency is 80.6%,indicating a 1.6%decrease compared to conventional *** the increase of FDR,the temperature uniformity of the heater tube is improved,and the CO and HC emissions ***,the impact of the FDR on the maximum temperature difference and temperature fluctuation across the heater is *** the FDR rises from 21%to 38%,the external combustion efficiency increases from 87.4%to92.3%.The excess oxygen coefficient plays a secondary role in influencing temperature uniformity and temperature difference,and the reformed gas and diesel fuel can be burned efficiently at a low excess oxygen coefficient of *** an increase in the cyclone angle,the heater tube temperature increases,while the maximum temperature difference at the lower part decreases,and the temperature fluctuation ***,the CO and HC emissions increase,and the external combustion efficiency experiences a decrease.A cyclone angle of 30°is found to be an appropriate value for achieving optimal

Shale oil of the Qingshankou Formation of the Gulong Sag,the northern Songliao Basin,represents the first attempt at large-scale development of pure-shale-type shale oil in *** integrating the multiscale refined reservoir characterization with macro-micro-scale mechanical testing,it is clarified that the Gulong shale is characterized by high clay mineral content,high rock plasticity,highly-developed bedding,and prominent mechanical anisotropy.A three-dimensional(3D)fracture propagation model of hydraulic fracturing was built for the Gulong shale,which fully captures the hydraulic fracture distribution pattern affected by the high bedding density,in-situ stress,and fracturing treatment *** research showed that due to influences of bedding,hydraulic fracturing in the Gulong shale forms a complex fracture morphology featuring the main fracture with multiple perpendicular branches that have different lengths(like the outdoor directional TV antenna);however,the vertical propagation of fractures is inhibited,and the fracture height is commonly less than 10 *** limited stimulated reservoir volume(SRV)is the main problem facing the fracturing stimulation of the Gulong shale *** density has vital effects on fracture morphology,so case-specific fracturing designs shall be developed for shale intervals with different bedding development *** reservoirs with welldeveloped bedding,it is suggested to properly increase the perforation cluster spacing and raise the volume and proportions of viscous fluids of the pad,so as to effectively promote vertical fracture propagation and improve reservoir stimulation *** study integrates multi-scale fine reservoir characterization and macro-micro-scale mechanical testing,as well as the construction and numerical simulation of hydraulic fracturing models for high-density layered shale reservoirs,providing a new approach and methodological framework for the fracturing research of high-density layer

WPIA: accelerating DNN warm-up in Web browsers by precompiling WebGL programs

In this paper, we study the long warm-up time of GPU acceleration of DNN inference in Web browsers. We analyzed the reason behind the long warm-up time through a measurement study and revealed that compiling WebGL programs takes most of the warm-up time. Inspired by this finding, we proposed WPIA, an approach that suggests precompiling WebGL programs on the server side to avoid compiling them in Web browsers. WPIA tackles the challenges of precompiling by merging WebGL programs and using a record-and-replay technique. Evaluation experiment results show that WPIA can accelerate the DNN warm-up time to an order of magnitude.
