Background： Most eukaryotic protein-coding genes exhibit alternative cleavage and polyadenylation （APA）, resulting in mRNA isoforms with different 3＇ untranslated regions （3＇ UTRs）. Studies have shown that brain cells tend to express long 3＇ UTR isoforms using distal cleavage and polyadenylation sites （PASs）. Methods： Using our recently developed, comprehensive PAS database PolyA_DB, we developed an efficient method to examine APA, named Significance Analysis of Alternative Polyadenylation using RNA-seq （SAAP-RS）. We applied this method to study APA in brain cells and neurogenesis. Results： We found that neurons globally express longer 3＇ UTRs than other cell types in brain, and microglia and endothelial cells express substantially shorter 3＇ UTRs. We show that the 3＇ UTR diversity across brain cells can be corroborated with single cell sequencing data. Further analysis of APA regulation of 3＇ UTRs during differentiation of embryonic stem cells into neurons indicates that a large fraction of the APA events regulated in neurogenesis are similarly modulated in myogenesis, but to a much greater extent. Conclusion： Together, our data delineate APA profiles in different brain cells and indicate that APA regulation in neurogenesis is largely an augmented process taking place in other types of cell differentiation.

以烤烟品种鲁烟1号为材料,在胶州进行大田试验,研究打顶后喷施4种化学调控剂(海藻素、赤霉芸苔素、细胞分裂素和光碳核肥)对上部烟叶开片和产质量的影响。结果表明:4种化学调控剂对调控烟株生长及改善烟叶品质均有一定作用。其中,细胞分裂素最能显著促进上部烟叶长、叶宽和叶面积的增加,促进上部叶开片,提高烟叶产量、产值、均价和上等烟及上中等烟比例等经济性状指标,改善烤后上部烟叶物理性状和化学成分协调性,提高烟叶品质。

The modulation of circularly polarized luminescence(CPL) has become an increasingly prominent area of research. In this study,we propose a unique “one-excitation-to-three-emissions” CPL system through the co-assembly of an upconversion system,incorporating one sensitizer(Pt(II) mesotetraphenyl tetrabenzoporphyrine, Pt TPBP) and two annihilators(R/S-DPA and R/SBDP) within liquid crystals. The chiral nature of the annihilators induces a transformation of the nematic liquid crystal into chiral nematic liquid crystals(N*LC), establishing an excellent chiral matrix. Upon the incorporation of the sensitizer Pt TPBP and subsequent excitation at 635 nm, the system demonstrates two independent triplet–triplet annihilation photon upconversion(TTA-UC) with the emission in blue and yellow, aided by thermally activated triplet–triplet energy transfer(TTET). This is accompanied by the simultaneous generation of upconverting circularly polarized luminescence(UC-CPL) and downshifting near-infrared circularly polarized luminescence(DS-CPL) originating from the residual luminescence of Pt TPBP. Remarkably,fine-tuning the ratio between the two annihilators allows the TTA-UC system to exhibit multicolor CPL emission with an amplified luminescence dissymmetry factor(glum, reaching up to 0.6). Our study unveils a previously unreported “one-excitation-to-three-emissions” system and provides a versatile strategy for modulating CPL emissions, surpassing conventional methodologies.

目的:本研究旨在探索利多卡因联合舒芬太尼静脉自控镇痛(patient controlled intravenous analgesia,PCIA)对腹腔镜结直肠癌根治术患者术后镇痛效果和肠道功能恢复的影响。方法:将135例拟行全身麻醉下腹腔镜结直肠癌根治术患者随机分为舒芬太尼组(S组,n=68)和利多卡因联合舒芬太尼组(LS组,n=67),术后的疼痛管理使用PCIA装置。S组(PCIA:舒芬太尼2.0μg/kg+昂丹司琼16 mg+0.9%盐水配至300 mL),LS组(PCIA:利多卡因48 mg/kg+舒芬太尼2.0μg/kg+昂丹司琼16 mg+0.9%盐水配至300 mL)。主要结果是术后6 h、12 h、24 h、36 h、48 h的VAS疼痛评分。次要结果包括术后48 h内PCIA有效按压次数和补救镇痛发生率;术后首次排气和排便时间;恢复质量(QoR-15)评分;术后住院时间;术后48 h内恶心呕吐(postoperative nausea and vomiting,PONV)的发生率;局麻药中毒不良事件的发生率。结果:LS组患者在术后6 h、12 h、24 h、36 h、48 h的VAS疼痛评分均显著低于S组(P0.05)。LS组患者术后首次排气和排便时间均显著早于S组(P0.05);术后第一天和术后第二天,LS组患者的QoR-15评分均显著高于S组(P<0.05)。LS组患者术后住院时间较S组显著缩短(P<0.05)。LS组患者PONV的发生率显著低于S组(P<0.05)。LS组患者未出现局麻药中毒不良事件。结论:利多卡因联合舒芬太尼PCIA能为腹腔镜结直肠癌根治术患者提供更有效的术后镇痛,加快肠道功能恢复。

Mosquitoes are of great medical significance as vectors of many deadly *** have been widely used in phylogenetic studies,but mitogenome knowledge within the family Culicidae is limited,and Culicidae phylogeny is far from *** this study,we surveyed the mitogenomes of 149 Culicidae species,including 7 newly sequenced *** analysis of 149 mosquito mitogenomes shows gene composition and order to be identical to that of an ancestral insect,and the AT bias,length variation,and codon usage are all consistent with that of other reported Dipteran *** analyses based on the DNA sequences of the 13 protein-coding genes from the 149 species robustly support the monophyly of the subfamily Anophelinae and the tribes Aedini,Culicini,Mansonini,Sabethini,and *** resolve ambiguous relationships between clades within the subfamily Culicinae,we performed topological tests and show that Aedini is a sister to Culicini and that Uranotaenini is a sister to(Mansonini)+(Toxorhynchitini+Sabethini).In addition,we estimated divergence times using a Bayesian relaxation clock based on the sequence data and 3 fossil calibration *** results show mosquitoes diverged during the Early Jurassic with massive Culicinae radiations during the Cretaceous,coincident with the emergence of angiosperms and the burst of mammals and ***,this study,which uses the largest number of Culicidae mitogenomes sequenced to date,comprehensively reveals the mitogenome characteristics and mitogenome-based phylogeny and divergence times of Culicidae,providing information for further studies on the mitogenome,phylogeny,evolution,and taxonomic revision of Culicidae.

Climate and topography regulate the spatial pattern of soil salinization and its effects on shrub community structure in Northwest China

Soil salinization may affect biodiversity and species composition,leading to changes in the plant community ***,few studies have explored the spatial pattern of soil salinization and its effects on shrub community structure at the ecosystem ***,we conducted a transect sampling of desert shrublands in Northwest China during the growing season(June–September)in *** salinization(both the degree and type),shrub community structure(e.g.,shrub density and height),and biodiversity parameters(e.g.,Simpson diversity,Margalf abundance,Shannon-Wiener diversity,and Pielou evenness indices)were used to assess the effects of soil salinization on shrub community *** results showed that the primary degree of soil salinization in the study area was light salinization,with the area proportion of 69.8%.Whereas the main type of soil salinization was characterized as sulfate saline soil,also accounting for 69.8%of the total ***,there was a significant reduction in the degree of soil salinization and a shift in the type of soil salinization from chloride saline soil to sulfate saline soil,with an increase in *** mean annual precipitation(MAP),mean annual evapotranspiration(MAE),elevation,and slope significantly contributed to soil salinization and its geochemical *** soil salinization intensified,shrub community structure displayed increased diversity and evenness,as indicated by the increases in the Simpson diversity,Shannon-Wiener diversity,and Pielou evenness ***,the succulent stems and leaves of Chenopodiaceae and Tamaricaceae exhibited clear advantages under these ***,regional climate and topography,such as MAP,MAE,and elevation,had greater effects on the distribution of shrub plants than soil *** results provide a reference for the origin and pattern of soil salinization in drylands and their effects on the community structure of halophyte shrub species.

Background:Soil acidifcationn caused by anthropogenic activities may aft soil biochemical cydling,bidiversity,productivity,and multiple eosystem-related functions in ***,to date,such information is lacking to support this *** Based on a transect survey of 78 naturally assembled shrub communities,we caloulated acid deposition flux in Northwest China and evaluated its likely ecological ffets by testing three altemnative hypotheses,namely:.nidche complementarity,mass ratio,and vegetation quantity hypotheses Rao's quadratic entopy and community-weighted mean traits were employed to represent the complementary aspect of niche complementarity and mass ratio effects,***:We observed that in the past four decades,the concentrations of exchangeable base cations in soil in Northwest China have decreased significantly to the extent of having faced the risk of depletion,whereas changes in the calium carbonate content and pH of soil were not *** deposition primani ly increased the aboweground biomass and shrub density in shrublands but had no sigmificant effect on shrub richness and ecasystem multifunctionality(EMF),indicating that acid deposition had positive but weak ecological effects on dryland *** wd ghted mean of functional traits(representing the mass ratio hypothesis)correlated negatively with EMF,whereas both Rao's quadratic entropy(representing the niche complementarity hypothesis)and aboveground biomass(representing the vegetation quantity hypothesis)correlated positively but insignifcantly with *** biodiversity-EMF relationships highlight the fragility and instability of drylands relative to forest ***:The findings from this study serve as important reference points to understand the ris of soil acidification in arid regions and its impacts on biodiversity-EMF relationships.
