The inverted triplesalen ligand h6feldMehas been synthesized from 2,4,6-triformyl-phloroglucinol and a ketimine salen half-unit in a convergent synthesis. NMR, IR, and UV-vis spectroscopy reveal that this ligand is not in the O-protonated tautomer but in the N-protonated tautomer with substantial heteroradialene contribution. This ligand and the conventional triplesalen ligand h6talent-Bu2 have been used to synthesize the trinuclear FeⅢcomplexes [(feldMe)(FeCl)3] and [(talent-Bu2)(FeCl)3], respectively. The molecular structures of these complexes were obtained by single-crystal X-ray diffraction. Two trinuclear FeⅢcomplexes of [(feldMe)(FeCl)3] dimerize via two Fe-phenolate bonds, whereas due to steric hindrance no dimerization is observed for [(talent-Bu2)(FeCl)3]. The structural data also reveal some heteroradialene contribution in the trinuclear complexes. Whereas UV-vis and M??bauer spectroscopy are not suitable to distinguish between the two complexes, FT-IR spectra show characteristic features due to the different substitution patterns of the conventional and the inverted triplesalen ligands. Another handle is provided by electrochemistry. Whereas both complexes exhibit an irreversible oxidation wave (0.94 V vs. Fc+/Fc for [(feldMe)(FeCl)3] and 0.84 V vs. Fc+/Fc for [(talent-Bu2)(FeCl)3]), which is assigned to the oxidation of the central backbone, higher potential oxidations are reversible for [(talent-Bu2)(FeCl)3]) but irreversible for [(feldMe)(FeCl)3]. This is attributed to the reversible oxidation of the terminal phenolates in the di-tert-butyl substituted [(talent-Bu2)(FeCl)3] in contrast to the mono-methyl-substituted phenolates in [(feldMe)(FeCl)3]. The magnetic properties of [(talent-Bu2)(FeCl)3] reveal a very small ferromagnetic coupling with significant zero-field splitting of the FeⅢS = 5/2 ions. Membrane gas separation is one of the most promising technologies for the separation of carbon dioxide （CO2） from various gas streams. One application of this technology is the treatment of flue gases from combustion processes for the purpose of carbon capture and storage. For this application, poly（ethylene oxide）-containing block copolymers such as Pebax or PolyActiveTM polymer are well suited. The thin-film composite membrane that is considered in this overview employs PolyActiveTM polymer as a selective layer material. The membrane shows excellent CO2 permeances of up to 4 m^3（STP）.（m^2·h·bar）^-1 （1 bar = 105 Pa） at a carbon dioxide/nitrogen （CO2/N2） selectivity exceeding 55 at ambient temperature. The membrane can be manufactured reproducibly on a pilot scale and mounted into fiat-sheet membrane modules of different designs. The operating performance of these modules can be accurately predicted by specifically developed simulation tools, which employ single-gas permeation data as the only experimental input. The performance of membranes and modules was investigated in different pilot plant studies, in which flue gas and biogas were used as the feed gas streams. The investigated processes showed a stable separation performance, indicating the applicability of PolyActiveTM polymer as a membrane material for industrialscale gas processing.

目的测试体外肺灌注(ex-vivo lung perfusion,EVLP)联合体外抗生素治疗移植前肺炎链球菌急性感染大鼠供肺的疗效,定量分析供肺感染后细菌载量的变化,评估体外肺功能。方法大鼠经气管注入或0.5 ml磷酸盐缓冲液(PBS)(对照组),或0.5 ml含105肺炎链球菌的PBS溶液(感染组、感染治疗组)后获取心肺供体。对照组和感染组不接受抗生素治疗,感染治疗组在常温EVLP灌注液中加入美罗培南和万古霉素。供肺均在体外常温灌注并通气4 h,对灌注液、肺泡灌洗液及供肺组织匀浆进行肺炎链球菌定量分析,并评估体外肺生理功能。采用2-way ANOVA分析肺功能,Dunnett法检验时间效果,Sidak’s法分析组间因素。供肺湿重变化采用1-way ANOVO分析,Tukey’s矫正。灌注液、肺泡灌洗液和肺组织的细菌定量以样品中细菌负荷的绝对浓度表示,并根据临床相关性分类,采用Fisher’s精确检验分析。结果 EVLP转流期间体外抗生素治疗对肺炎链球菌感染供肺静态顺应性无显著改善作用[(0.076±0.020) ml/cmh2O比(0.096±0.035) ml/cmh2O,t=0.984,P>0.05],感染供肺无论是否进行了体外抗生素治疗其氧合能力均随灌注延长逐渐下降[(32.75±12.03) mmhg比(41.31±13.46) mmhg,t=1.060,P>0.05]。对照组和感染治疗组供肺在4 h EVLP灌注液中无细菌污染,感染组EVLP结束时所有供肺灌洗液中均可培养出细菌(χ^(2)=15.00,P0.05)。感染治疗组(细菌中位数=103)比未治疗组(细菌中位数106)肺实质细菌载量低,两组均比对照组具有更高的肺实质细菌定量(χ^(2)=13.00,P<0.05)。结论本模型证明在常温EVLP转流期间进行体外抗生素治疗可减少急性肺炎链球菌感染的供肺和整个EVLP系统的细菌载量,但对肺功能改善不明显。

Objective:Anoctamin 7(ANO7)is a calcium2+-dependent chloride ion channel *** expression is restricted to prostate epithelial *** exact function is *** study aimed to analyze ANO7 expression and its clinical significance in prostate cancer(PCa).Methods:ANO7 expression was assessed by immunohistochemistry in 17,747 clinical PCa ***:ANO7 was strongly expressed in normal prostate glandular cells but often less abundant in cancer ***7 staining was interpretable in 13,594 cancer tissues and considered strong in 34.4%,moderate in 48.7%,weak in 9.3%,and negative in 7.6%.Reduced staining was tightly linked to adverse tumor features[high classical and quantitative Gleason grade,lymph node metastasis,advanced tumor stage,high Ki67 labeling index,positive surgical margin,and early biochemical recurrence(Ph)].The univariate Cox hazard ratio for prostate-specific antigen(PSA)recurrence after prostatectomy in patients with negative *** ANO7 expression was 2.98(95%confidence interval 2.61–3.38).The prognostic impact was independent of established pre-or postoperatively available parameters(Phowed that low ANO7 expression was linked to TMPRSS2:ERG fusions(Phromosomal deletions(Ph).A particularly strong association of low ANO7 expression with phosphatase and tensin homolog(PTEN)deletion may indicate a functional relationship with the PTEN/AKT ***:These data identify reduced ANO7 protein expression as a strong and independent predictor of poor prognosis in ***7 measurement,either alone or in combination,might provide clinically useful prognostic information in PCa.

Skeletal health relies on architectural integrity and sufficient bone mass, which are maintained through a tightly regulated equilibrium of bone resorption by osteoclasts and bone formation by osteoblasts. Genetic studies have linked the gene coding for low-density lipoprotein receptor-related protein1(Lrp1) to bone traits but whether these associations are based on a causal molecular relationship is unknown. here, we show that Lrp1 in osteoblasts is a novel regulator of osteoclast activity and bone *** lacking Lrp1 specifically in the osteoblast lineage displayed normal osteoblast function but severe osteoporosis due to highly increased osteoclast numbers and bone resorption. Osteoblast Lrp1 limited receptor activator of NF-κB ligand(RANKL) expression in vivo and in vitro through attenuation of platelet-derived growth factor(PDGF-BB) signaling. In co-culture, Lrp1-deficient osteoblasts stimulated osteoclastogenesis in a PDGFRβ-dependent manner and in vivo treatment with the PDGFR tyrosine kinase inhibitor imatinib mesylate limited RANKL production and led to complete remission of the osteoporotic phenotype. These results identify osteoblast Lrp1 as a key regulator of osteoblast-to-osteoclast communication and bone mass through a PDGF–RANKL signaling axis in osteoblasts and open perspectives to further explore the potential of PDGF signaling inhibitors in counteracting bone loss as well as to evaluate the importance of functional LRP1 gene variants in the control of bone mass in humans.

Effects of ethanol on the properties of platelets and endothelial cells in model experiments

AIM: To investigate effects of ethanol on activity markers of atherosclerosis in an in vitro endothelial cell model. METhODS: After 24 h incubation with ethanol (0.0095%), human umbilical vein endothelial cells were stimulated for 1 h with lipopolysaccharide, and were then incubated in direct contact with activated platelets. Following this incubation, the expression of CD40L and CD62P on platelets, and the expression of intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), urokinase plasminogen activator receptor (uPAR), and membrane-type 1 matrix metalloproteinase (MT1-MMP) on endothelial cells were measured by flow cytometry. RESULTS: The increased expression of VCAM-1 and uPAR on endothelial cells by proinflammatory stimulation with activated platelets was significantly reduced through pre-incubation with ethanol (P hermore, platelets in direct contact with ethanol and with endothelial cells pre-incubated in ethanol showed a significant reduction in their CD40L expression (P hanol had no significant effect on ICAM-1 and MT1-MMP expression on endothelial cells. CONCLUSION: Ethanol directly attenuates platelet activation and has significant endothelial cell-mediated effects on selected markers of atherosclerosis in vitro. These findings underline possible protective effects of ethanol on atherosclerosis.
