Pediatric autoimmune neuropsychiatric disorders associated with or without streptococcal and other bacterial infections (PANDAS/CANS) are emerging as a featured pediatric disorder. Although there is some controversy regarding treatment approaches, especially related to the behavioral sequelae, we have hypothesized in other published work that it is characterized by the rapid onset of reward Deficiency Syndrome (rDS) in children. We propose utilizing a multi-systems biological approach involving the coupling of genetic addiction risk testing and pro-dopamine regulation (KB220/POLYGEN®) to help induce “dopamine homeostasis” in patients with PANDAS, especially those with known DNA-induced hypodopaminergia. This case study examines a 12-year-old Caucasian male with no prior psychiatric issues who presented with a sudden onset of severe anxiety, depression, emotional liability, and suicidal ideation. The patient underwent genotyping and the genetic addiction risk score (GArS) testing, which revealed risk polymorphisms in the dopamine D2 (-DrD2/ANKK (Taq1A), OPrM1 (A/G), DrD3 (C/T), and MAOA (4r) genes. These polymorphisms have been linked to hypodopaminergia. The patient was subsequently placed on research ID-KB220ZPBMPOLY (POLYGEN®), and albeit the possibility of bias, based upon self and parental assessment, a marked rapid improvement in psychiatric symptoms was observed. In the second phase of treatment (102 days utilizing KB220), the patient received standard antibody testing, which was positive for Lyme. Antibacterial therapy started immediately, and KB220z was discontinued to provide a wash-out period. A monotonic trend analysis was performed on each outcome measure, and a consistently decreasing trend was observed utilizing antibacterial therapy. Our recommendation, albeit only one case, is to utilize and further research a combined therapeutic approach, involving precision-guided DNA testing and pro-dopamine regulation along with antibacterial therapy, as well as

Metabolic syndrome and the urinary microbiome of patients undergoing percutaneous nephrolithotomy

Detecting Android Malware Using Clone Detection

Android is currently one of the most popular smartphone operating systems. However, Android has the largest share of global mobile malware and significant public attention has been brought to the security issues of Android. In this paper, we investigate the use of a clone detector to identify known Android malware. We collect a set of Android applications known to contain malware and a set of benign applications. We extract the Java source code from the binary code of the applications and use NiCad, a near-miss clone detector, to find the classes of clones in a small subset of the malicious applications. We then use these clone classes as a signature to find similar source files in the rest of the malicious applications. The benign collection is used as a control group. In our evaluation, we successfully decompile more than 1 000 malicious apps in 19 malware families. Our results show that using a small portion of malicious applications as a training set can detect 95% of previously known malware with very low false positives and high accuracy at 96.88%. The contribution of the melanin pathway to overall body pigmentation during ontogenesis of Periplaneta americana

The most prominent colors observed in insects are black or brown, whose production is attributed to the melanin pathway. At present, though, the contribution of this pathway to overall body pigmentation throughout ontogenesis is still lacking. To address this question we examined the roles of 2 key melanin genes （TH and DDC）, in embryonic and postembryonic development of the American cockroach, Periplaneta americana. Our results show that the melanin pathway does not contribute to the light brown coloration observed in the first nymphs. However, the dark brown coloration in mid nymphs and adults is produced solely from the melanin pathway. In addition, the DDC rNAi results reveal that it is dopamine melanin, not DOPA melanin, acts as the main contributor in this process. Overall, present study provides a new insight into insect pigmentation suggesting that genetic mechanisms of coloration can change during ontogenesis. Future studies of additional basal insect lineages will be required to assess in more details the generality of this phenomenon.

STEP AP 242 Managed Model-based 3D Engineering:An Application Towards the Automation of Fixture Planning

Fixture design and planning is one of the most important manufacturing activities, playing a pivotal role in deciding the lead time for product development. Fixture design, which affects the part-quality in terms of geometric accuracy and surface finish, can be enhanced by using the product manufacturing information(PMI) stored in the neutral standard for the exchange of product model data(STEP) file, thereby integrating design and manufacturing. The present paper proposes a unique fixture design approach, to extract the geometry information from STEP application protocol(AP) 242 files of computer aided design(CAD) models, for providing automatic suggestions of locator positions and clamping surfaces. Automatic feature extraction software "FiXplan", developed using the programming language C#, is used to extract the part feature, dimension and geometry information. The information from the STEP AP 242 file is deduced using geometric reasoning techniques, which in turn is utilized for fixture planning. The developed software is observed to be adept in identifying the primary, secondary, and tertiary locating faces and locator position configurations of prismatic components. Structural analysis of the prismatic part under different locator positions was performed using commercial finite element method software, ABAQUS, and the optimized locator position was identified on the basis of minimum deformation of the *** area-ratio(base locator enclosed area(%)/work piece base area(%)) for the ideal locator configuration was observed as 33%. Experiments were conducted on a prismatic workpiece using a specially designed fixture, for different locator configurations. The surface roughness and waviness of the machined surfaces were analysed using an Alicona non-contact optical profilometer. Contemporary Approaches for Identifying rare Bone Disease Causing Genes

recent improvements in the speed and accuracy of DNA sequencing, together with increasingly sophisti- cated mathematical approaches for annotating gene networks, have revolutionized the field of human genetics and made these once time consuming approaches assessable to most investigators. In the field of bone research, a particularly active area of gene discovery has occurred in patients with rare bone disorders such as osteogenesis imperfecta （OI） that are caused by mutations in single genes. In this perspective, we highlight some of these technological advances and describe how they have been used to identify the genetic determinants underlying two previously unexplained cases of OI. The widespread availability of advanced methods for DNA sequencing and bioinformatics analysis can be expected to greatly facilitate identification of novel gene networks that normally function to control bone formation and maintenance.
