Comparison of Three Experimental Models for Rat Osteoarthritis Induction

Background: Osteoarthritis is a slowly progressive and debilitating disease with high prevalence in adult population. Knee is one of the joints most affected by this disorder. There are several models for animals’ osteoarthritis induction, however it is not identified any paper that compares these techniques. The present study was aimed to define the most appropriate model for rats osteoarthritis induction. Material and Methods: 40 Wistar rats were distributed into 4 groups of 10 animals each: normality group (NG);meniscectomy group (MG);quinolone group (QG) and iodoacetate group (IG). Radiographic images of the rat’s knees were analyzed as well as the amount of chondrocytes in the epiphyseal and articular cartilage. Results: In the radiographic analysis, there was a low correlation between the raters. Regarding the amount of chondrocytes in the epiphyseal cartilage, it was noticed that the IG and QG groups had fewer chondrocytes than NG, in contrast to MG that reported similar results to normality (p > 0.05). There was no significant difference between IG and QG groups (p > 0.05). Regarding the amount of chondrocytes in articular cartilage, it was noticed that the IG group showed fewer chondrocytes than NG (p 0.05). There was no significant difference between QG and MG groups (p > 0.05). Conclusion: Intraarticular injection of iodoacetate in rats is the model with greatest effect on reduction of chondrocytes amount.

Fourier and Wavelet Spectral Analysis of EMG Signals in 1-km Cycling Time-Trial

Frequency domain analyses in electromyographic (EMG) signals are frequently applied to assess muscle fatigue and similar variables. Moreover, Fourier-based approaches are typically used for investigating these procedures. Nonetheless, Fourier analysis assumes the signal as stationary which is unlikely during dynamic contractions. As an alternative method, wavelet-based treatments do not assume this pattern and may be considered as more appropriate for joint time-frequency domain analysis. Based on the previous statements, the purpose of the present study was to compare the application of Short-Time Fourier Transform (STFT) and Continuous Wavelet Transform (CWT) to assess muscle fatigue in dynamic exercise of a 1-km of cycling (time-trial condition). The results of this study indicated that CWT and STFT analyses have provided similar fatigue estimates (slope) (p> 0.05). However, CWT application represents lesser dispersion (p 0.05) according to different methods, it is important to note that these responses seem to show greater values for CWT compared to STFT for 2 superficial muscles. Thereby, we are capable of considering CWT as a reliable and useful method to take into consideration when non-stationary or oscillating exercise models are evaluated.

Age and Calcium Sources in Laying Hen Feed Affect Calcium Digestibility

The apparent calcium (Ca) digestibility coefficient (ADC) and true digestibility coefficient (TDC) of different inorganic calcium sources were determined in laying hens of different ages. Three Ca digestibility tests were carried out, each assessing 240 Lohmann Brown lineage laying hens distributed in a completely randomized design. Nine dietary treatments were arranged in a 3 × 3 factorial design consisting of three ages (40, 50 and 70 weeks) × three Ca (dicalcium phosphate (DCP) sources, fine (FL) and coarse (CL)) limestone, comprising eight replicates per treatment of six birds per experimental unit. Regarding the DCP, the ADC was higher (P as 0.889, 0.613 and 0.712, respectively. No effect (P > 0.05) of age on the ADC was noted for either FL or CL. Keloid of Ears: Recurrence and Its Complications

In this case report, we present a male patient who arrived at the hospital for the first time at the age of 8 with giant keloids in both ears and diagnosed by the Dermatology and Plastic Surgery as having the “Disease of Jorge Lobo”, which is a fungal infection due to the story of the endemic characteristics. After the first surgical intervention, he received guidance from the team for keloids and left the hospital using elastic mesh and a request to change the place where he lived to reduce his contact with the fungi. Fourteen years later, the patient returned to the Plastic Surgery Service having even larger keloids in both ears, twice the size than the first time. We did the second surgical intervention to remove it, but with the patient’s commitment that he would correctly comply with the guidelines determined by the surgical team. After finishing all the steps and a post-operative for the case, we observed the satisfaction and the increase of the patient’s mood, happier and without the embarrassment of that physical defect.

AIM To evaluate the therapeutic effects of bone marrowderived CD11b+CD14+ monocytes in a murine model of chronic liver damage. METHODS Chronic liver damage was induced in C57BL/6 mice by administration of carbon tetrachloride and ethanol for 6 mo. Bone marrow-derived monocytes isolated by immunomagnetic separation were used for therapy. The cell transplantation effects were evaluated by morphometry,biochemical assessment,immunohistochemistry and enzyme-linked immunosorbent assay. RESULTS CD11b+CD14+ monocyte therapy significantly reduced liver fibrosis and increased hepatic glutathione levels. Levels of pro-inflammatory cytokines,including tumor necrosis factor-α,interleukin(IL)-6 and IL-1β,in addition to pro-fibrotic factors,such as IL-13,transforming growth factor-β1 and tissue inhibitor of metalloproteinase-1 also decreased,while IL-10 and matrix metalloproteinase-9 increased in the monocytetreated group. CD11b+CD14+ monocyte transplantation caused significant changes in the hepatic expression of α-smooth muscle actin and osteopontin. CONCLUSION Monocyte therapy is capable of bringing about improvement of liver fibrosis by reducing oxidative stress and inflammation,as well as increasing antifibrogenic factors.

An Unusual Case of Herpes Simplex Meningitis after Inadvertent Puncture of the Subarachnoid Space

The study seeks to report the clinical evolution of a patient who suffered an inadvertent subarachnoid puncture and evolved with a herpetic viral meningitis, confirmed by cerebrospinal fluid (CSF) analysis. Female, 48 years old, submitted to epidural anesthesia, for surgery, with median puncture, between L2-L3 and inadvertently accessed the subarachnoid space, with clear cerebrospinal fluid. The same puncture was used and the solution with 15 mg hyperbaric bupivacaine and 80 mcg morphine was administered. There was no complication during the entire surgical procedure. On the first postoperative day, she started complaining of headache and nausea, being treated with dipyrone, non-steroidal anti-inflammatory drugs (NSAIDs) and ondansetron. On the third day, she presented two episodes of seizure that ceased with the use of diazepam 10 mg. On the fifth day, the patient presented fever and seizures, which did not stop with medication, requiring sedation, orotracheal intubation and transfer to the intensive care unit. Lumbar puncture was performed for CSF analysis, which was positive for Herpes simplex type I. The patient was extubated, on the second day of hospitalization, maintained with a maintenance dose of phenytoin, sumatriptan, dipyrone, metoclopramide and acyclovir 500 mg, remaining intravenous, without presenting new seizures for 45 days, when she was discharged. After 30 days, he returned for a review consultation without making a complaint.

Postgraduate education among family and community physicians in Brazil:the Trajetórias MFC project

Objective Our objective was to describe the postgraduate education trajectories of family and community physicians in Brazil,where neither primary healthcare nor family and community medicine is recognised as a knowledge area for the purpose of research and postgraduate education(master’s and PhD degrees).Design An observational,exploratory study,using administrative data.A nationwide list of family and community physicians as of late November 2018 was compiled from multiple *** on the mode of specialisation was obtained from the same sources and were correlated with data on master’s and PhD degrees,obtained from the curricula vitae on the Lattes *** This study was set in *** 6238 family and community physicians(58.3%female),of whom 2795 had earned a specialist certificate(identified from the list of physicians certified by Sociedade Brasileira de Medicina de Família e Comunidade)and 3957 had completed medical residency(identified from SisCNRM,the national information system for medical residency).Results A master’s degree was held by 747(12.0%)family and community physicians,and a PhD by 170(2.7%);most degrees were in collective health(47.0%and 42%,respectively).Men were more likely than women to hold a master’s degree(adjusted odds ratio(aOR)1.24,95%uncertainty interval(UI)1.07-1.45)and even more likely to a hold PhD(aOR 1.86,95%UI 1.35-2.59).Family and community physicians were also less likely to hold a PhD degree if their master’s degree was professional(oriented towards jobs outside academia)instead of academic(aOR 0.15,95%UI 0.05-0.39)or in some area other than collective health or medicine(aOR 0.41,95%UI 0.21-0.78,compared with a master’s degree in collective health).The postgraduate degree was more likely to precede specialisation for family and community physicians specialising through certification(master’s degree 39.9%,PhD 33%)than through medical residency(master’s degree 9.1%,PhD 6%).Conclusion Family and community

Fluorescence of Chlorophyll a for Discovering Inhibitors of Photosynthesis in Plant Extracts

Aims: Twelve crude extracts were prepared from different parts of Pluchea sagittalis, Cecropia palmata and Brachiaria brizantha, and their effect on the light reaction of photosynthesis. Study Design: Use of fluorescence of ChL a (Chlorophyll a) to detect inhibitors of photosynthesis in plant extracts. Place and Duration of Study: Instituto de Ciências Ambientais, Químicas e Farmacêuticas, Departamento de Ciências Exatas e da Terra, Universidade Federal de Sao Paulo (UNIFESP), between July 2015 and January 2016. Methodology: The samples had their effect on the light reaction of photosynthesis studied by Chlorophyll a (Chl a) fluorescence transient by the use of chloroplasts isolated from spinach leaves. As negative controls we used the same amounts of DMSO that have been employed to solubilize the samples, and as positive control we have used 10 μM of the commercial herbicide DCMU (Diuron). Results: The extracts inhibited the photosynthesis, and their inhibition sites were located in the range of electron flow from OEC complex and between P680 to QA of PS II, and inhibited the photosystem II (PS II) by inducing the appearance of the transient bands K and J. Conclusion: Our results suggest an indicative that the photochemical apparatus probably is failing at the donor and acceptor sides of PSII, causing damage to electron transport during the photosynthesis phenomenon.

Isolation of Filamentous Fungi from the Caatinga Region and Production of Amylolytic Enzymes of Great Industrial Interest

The industrial importance and the high cost of the commercial amylase require the study of microorganisms that produce these enzymes. For this reason, the objective of this work was to isolate filamentous fungi from a region of the caatinga and evaluate their potential for the production of amylase. Four soil samples were collected from a deactivated dump located in the city of Diamantina, MG, in a region of the caatinga. The analysis of amylolytic production in a submerged medium at the ideal temperature of each microorganism was performed using the saccharification method, and the reducing sugars formed were quantified by DNS. Fourteen filamentous fungi were isolated, which had different morphological aspects. Regarding amylase production, a mean activity of 0.477 U·mL-1 was obtained with the isolates I 1.2.1 and I 4.4.1. These results bring important information regarding the biodiversity of the caatinga, in addition to the isolation of microorganisms that can be used as biological machinery to obtain metabolites with high biotechnological and industrial potential.

BACKGROUND Metabolic dysfunction-associated fatty liver disease corresponds to a clinical entity that affects liver function triggered by the accumulation of fat in the liver and is linked with metabolic *** To evaluate the effects of the intragastric balloon(IGB)in patients with metabolic dysfunction-associated fatty liver disease through the assessment of liver enzymes,imaging and several metabolic *** A comprehensive search was done of multiple electronic databases(MEDLINE,EMBASE,LILACS,Cochrane and Google Scholar)and grey literature from their inception until February *** criteria involved patients with a body mass index>25 kg/m2 with evidence or previous diagnosis of hepatic *** analyzed before and after 6 mo of IGB removal were alanine aminotransferase(IU/L),gamma-glutamyltransferase(IU/L),glycated hemoglobin(%),triglycerides(mg/dL),systolic blood pressure(mmHg),homeostatic model assessment,abdominal circumference(cm),body mass index(kg/m2)and liver volume(cm3).RESULTS Ten retrospective cohort studies evaluating a total of 508 patients were *** 6 mo of IGB placement,this significantly reduced alanine aminotransferase[mean difference(MD):10.2,95%confidence interval(CI):8.12-12.3],gammaglutamyltransferase(MD:9.41,95%CI:6.94-11.88),glycated hemoglobin(MD:0.17%,95%CI:0.03-0.31),triglycerides(MD:38.58,95%CI:26.65-50.51),systolic pressure(MD:7.27,95%CI:4.79-9.76),homeostatic model assessment(MD:2.23%,95%CI:1.41-3.04),abdominal circumference(MD:12.12,95%CI:9.82-14.41)and body mass index(MD:5.07,95%CI:4.21-5.94).CONCLUSION IGB placement showed significant efficacy in improving alanine aminotransferase and gamma-glutamyltransferase levels in patients with metabolic dysfunctionassociated fatty liver disease as well as improving metabolic markers related to disease progression.
