Application of an impact plate-Bedload transport measuring system for high-speed flows

To achieve the sustainable use of dams, the development of methods for sediment management in reservoirs is required. One such method includes the use of Sediment Bypass Tunnels(SBTs) to divert sediment around a dam, thereby preventing sedimentation in the reservoir. However, SBTs are prone to severe invert abrasion caused by the high sediment flux. Therefore, it is necessary to establish a measurement system of the sediment transport rate in these tunnels. One system to measure sediment transport in rivers is the Swiss plate geophone, which can register plate vibrations caused by particle impact. In Japan, the Japanese pipe microphone is used, and sediment transport is measured based on the sound emitted by the particle impact. In this study an attempt was made to optimize the advantages of both systems by fixing a microphone and an acceleration sensor to a steel plate. The results of calibration experiments with this new system are presented and compared with the existing methods. It was found that the acceleration sensor can detect sediment particles larger than 2 mm in ***, a new parameter, referred to as the detection rate, was introduced to describe the correlation between the actual amount of sediment and the registered output. Reservoirs are installed as long-term assets to guarantee water and energy security for decades,if not ***,the effect of siltation undermines reservoirs’sustainability because it significantly reduces the reservoirs’original *** present paper attempts to evaluate the global reservoir siltation problem with the optimism bias theorem introduced by Kahneman and Tversky and applied to infrastructural mega-projects by Flyvbjerg and Ansar using artificial neural networks(ANNs)algorithms for large Japanese *** possesses suitable long-term data and a legal directive concerning the sediment capacity siltation duration,which serves as a valid guide to check whether,over the past 100 years,engineers,planners and managers were capable of judging the sediment input *** ANN models were established to emulate Japanese reservoir siltation *** networks demonstrate that reservoirs in Japan suffer from optimism *** contrast to the law,the dead storage volume of an average dam is supposed to reach capacity after 52 *** finding joins the overall observation that mega-projects generally and globally suffer from optimism *** emulations were subsequently screened for a presumed influence of governance actions,namely,indicating plus monitoring and the change in the market competition *** reservoir siltation appears to continue regardless of the level of competition in public procurement,monitoring directives appear to have a considerable impact on improved siltation management,which demonstrates that dedicated governance action can significantly strengthen the sustainable behavior of key infrastructure elements such as reservoirs.

The relationship between a university and its city is considered strategical to achieve university targeted ambitions and *** university-city relationship is also encouraged for the benefit of the city,as universities unleash their respective cities’potentials to act as driving forces not only for their local communities,but also for the whole ***,maintaining a mutual relationship between the university and the city is considered essential to accomplish strategic goals for ***,the nature of this relationship is quite complex,overlapped,interconnected,and ***,this paper conducted a systematic review of the literature on university-city and campus-city relationships to evaluate recent research trends to uncover the aspects that connect universities with their respective *** search included articles published in 4 different databases from January 1990 to January 2021.A total number of 50 articles were selected in this *** findings uncovered different aspects that could help or hinder university-city relationship based on the physical and functional linkage between the campus and the ***,findings have shown that it is necessary to understand universities according to their contextual differences,as universities have shown different impacts on their respective cities in terms of their sizes and *** have also shown that the impact of the physical connection between the university and the city goes far beyond campus’s accessibility as it deeply affects students’social life as ***,decision makers,stakeholders,and university administrators need to co-design campus development process especially in the early stages to maximize the mutual benefits of campus-city *** main conclusions of this paper address several perspectives and lessons for a more sustainable campus-city relationship.

We examined the degradation of dibromophenols （DBPs）, i.e. 2,4-DBP, 2,6-DBP and 3,5-DBP by ultraviolet （UV） irradiation and estimated the relationship between degradability and molecular orbital properties of each dibromopbenol. The removal of DBPs under a UV lamp system was successfully performed in an aqueous solution. After 5 min of irradiation, the initial DBPs concentration of 20 mg/L was decreased to below 1 mg/L, and about 60% of bromide ion was released. A decrease in the concentration of dissolved organic carbon （DOC） suggested the mineralization of DBPs, The mineralization may occur after release of bromide ions because the decrease of DOC was slower than the release of bromide ions. The degradability of 3,5-DBP was slightly lower than 2,6-DBP and 2,4-DBE Molecular orbital calculation suggested that the electrophilic frontier density and the highest occupied molecular orbital （HOMO） energy may be related to the degradability of DBPs.

Pollen tube guidance is controlled by multiple complex interactions with the female tissues. Here, we show that pollen tubes of Torenia fournieri are regulated by a stylar tissue in a length-dependent manner to receive and respond to attractant LURE peptides secreted from synergid cells. We developed an immunostaining method to visualize LURE peptides bound at the plasma membrane of the tip region of the pollen tube. Using this method, we found that LURE peptides bound specifically to pollen tubes growing through a cut style. The peptides also bound to pollen tubes growing through a shorter style, which were not competent to respond to these peptides. These observations suggested a possibility that acquisition of the LURE peptide reception ability and acquisition of full competency are separable pro- cesses. RNA-Seq suggested that the transcription profile of pollen tubes was affected by both the length of the style and the cultivation period, consistently with physiological changes in binding activity and LURE response ability. The data- base generated from de novo RNA-Seq of Torenia pollen tubes was shown to be useful to identify pollen tube proteins by mass spectrometry. Our studies provide insight and an effective platform for protein identification to understand pollen tube guidance.

Safety and effectiveness of propofol sedation during and after outpatient colonoscopy

AIM:To study the safety and effectiveness of propofol sedation for outpatient ***:Propofol was given by bolus injection with an age-adjusted standard protocol consisting of 60 mg for patients < 70 years old,40 mg for patients age 70-89 years,and 20 mg for those ≥ 90 years,and additional injections of 20 mg propofol were given up to a maximum of 200 *** principal parameters were the occurrence of adverse events within 24 h after colonoscopy and overall satisfaction for this *** parameters included successful procedure,respiratory depression,and other ***:Consecutive patients were entered prospectively and all 2101 entered successfully completed outpatient *** mean dose of propofol used was 96.4 mg(range 40-200 mg).Younger patients required higher doses of propofol than older patients(20-40 years vs ≥ 61 years:115.3 ± 32 mg vs 89.7 ± 21 mg,P < 0.001).Transient supplemental oxygen supply was needed by five patients(0.2%);no other complications *** questionnaires were completed by 1820(87%) of 2101 patients and most rated their overall satisfaction as excellent(80%) or good(17%).The majority(65%) of patients drove home or to their office after their ***(99%) were willing to repeat the same *** incidents occurred within 24 h after ***:Propofol sedation using a dose < 200 mg proved both safe and practical for outpatient colonoscopy.

AIM： To investigate the roles of Bax and p53 proteins in photosensitivity of human colon cancer cells by using lysosome-localizing photosensitizer, ATX-S10Na （Ⅱ）. METHODS： HCT116 human colon cancer cells and Bax-null or p53-null isogenic derivatives were irradiated with a diode laser. Early apoptosis and cell death in response to photodynamic therapy were determined by MTT assays, annexin Ⅴ assays, transmission electron microscopy assays, caspase assays and western blotting. RESULTS： Induction of early apoptosis and cell death was Bax- and p53-dependent. Bax and p53 were required for caspase-dependent apoptosis. The levels of anti-apoptotic Bcl-2 family proteins, Bcl-2 and Bcl-xL, were decreased in Bax- and p53-independent manner. CONCLUSION： Our results indicate that eady apoptosis and cell death of human colon cancer cells induced by photodynamic therapy with lysosome-localizing photosensitizer ATX-S10Na （Ⅱ） are mediated by p53- Bax network and low levels of Bcl-2 and Bcl-xL proteins. Our results might help in formulating new therapeutic approaches in photodynamic therapy.

Development of a competitive enzyme-linked immunosorbent assay for therapeutic drug monitoring of afatinib

Afatinib is an oral tyrosine kinase inhibitor(TKI) approved for treating advanced non-small cell lung cancer. It is necessary to develop a simple quantification method for TKIs in order to facilitate therapeutic drug monitoring(TDM) in clinical settings. This study sought to develop a simple and sensitive competitive enzyme-linked immunosorbent assay(ELISA) to quantify afatinib in plasma for routine pharmacokinetic applications. An anti-afatinib antibody was obtained using(S)-N-4-(3-chloro-4-fluorophenyl)-7-(tetrahydrofuran-3-yloxy)-quinazoline-4,6-diamine(CTQD), which has the same substructure as afatinib, as a hapten. Enzyme labeling of afatinib with horseradish peroxidase was similarly performed using CTQD. A simple competitive ELISA for afatinib was developed based on the principle of direct competition between afatinib and the enzyme marker for the anti-afatinib antibody, which had been immobilized on the plastic surface of a microtiter plate. Plasma afatinib concentrations below the limit of quantification of 30 pg/mL were reproducibly measurable. Also, the values of plasma afatinib levels measured from 20 patients were comparable with those measured by high-performance liquid chromatography, and there was a strong correlation between the values determined by both methods(Y=0.976 X – 0.207, r=0.975). As indicated by its specificity and sensitivity, this newly developed ELISA for afatinib is an important tool for TDM and studies of the pharmacokinetics of afatinib.

Photothermal therapy of tumors in lymph nodes using gold nanorods and near-infrared laser light with controlled surface cooling

Photothermal therapy （PTT） using near-infrared （NIR） laser light and gold nanorods （GNRs） shows promise as a novel cancer treatment modality. However, the laser intensity required to destroy tumor cells located beneath the skin is greater than the threshold intensity that causes skin damage; thus, irradiation with laser light damages the skin as well as the tumor. Here, we show that a temperature control system allows metastatic lymph nodes （LNs） to be treated by PTT using NIR laser light and GNRs, without skin damage. A mouse model of LN metastasis was developed by injection of tumor cells, and the tumor-bearing proper axillary LN was treated with NIR laser light after injection of GNRs. The skin temperature was maintained at 45 ~C during irradiation by using a temperature control system. Bioluminescence imaging revealed that tumor progression was less in LNs exposed to NIR laser light and GNRs than in LNs exposed to NIR laser light alone or controls （no irradiation or GNRs）. Furthermore, the skin and LN capsule were macroscopically intact on day 9 after irradiation with NIR laser light, whereas tumor cells within the LN showed apoptosis. A numerical analysis demonstrated that the high-temperature zone and the LN region showing damage were localized to an area up to 3 mm in depth. The proposed novel PTT technique, using NIR laser light and GNRs with controlled surface cooling, could be applied clinically to treat metastatic LNs located within or outside the area accessible for surgical dissection.
