Attenuated retinoic acid signaling is among the early responses in mouse uterus approaching embryo attachment

The uterus is transiently receptive for embryo *** remains to be understood why the uterus does not reject a semi-allogeneic embryo(to the biological mother)or an allogeneic embryo(to a surrogate)for *** gain insights,we examined uterine early response genes approaching embryo attachment on day 3 post coitum(D3)at 22 hours when blue dye reaction,an indication of embryo attachment,had not manifested in mice.C57BL/6 pseudo-pregnant(control)and pregnant mouse uteri were collected on D3 at 22 hours for microarray *** self-assembling-manifold(SAM)algorithm identified 21,858 unique *** component analysis indicated a clear separation between the pseudo-pregnant and pregnant *** were 106 upregulated and five downregulated protein-coding genes in the pregnant uterus with fold change(fc)>1.5 andq valueene ontology(GO)analysis of the 106 upregulated genes revealed 38 significant GO biological process(GOBP)terms(Pem were associated with immune responses,with a dominant natural killer(NK)cell activation *** the top eight upregulated protein-coding genes,Cyp26a1 inactivates retinoic acid(RA)whileLrat promotes vitamin A storage,both of which are expected to attenuate RA bioavailability;Atp6v0d2 andGjb2 play roles in ion transport and transmembrane transport;Gzmb,Gzmc,andIl2rb are involved in immune responses;andTdo2 is important for kynurenine *** of these genes or their related pathways have functions in immune *** signaling has been implicated in immune tolerance and immune homeostasis,and uterine NK cells have been implicated in immunotolerance at the maternal-fetal interface in the *** mechanisms of immune responses approaching embryo attachment remain to be *** coordinated effects of the early response genes may hold the keys to the question of why the uterus does not reject an implanting embryo.

Toxicity of a seafood toxin， domoic acid， in the retina via modulation of the NRF2 and NF-κB pathways

Domoic acid (DA)， a biotoxin， is produced by several species of marine dinoflagellate and diatom during harmful algal bloom events. DA is a neurotoxin， in humans and non-human primates， oral exposure to DA results in gastrointestinal effects， while DA at higher doses leads to neurological symptoms， seizures and memory deficiency. exposure of humans to DA occurs mainly through consumption of contaminated seafoods containing an accumulation of the toxin. Previously， it was unclear if DA can have toxic effects on the retina. We assessed the toxicity of DA in human retinal cells (ARPe-19) and in zebrafish embryos. DA significantly lowered ARPe-19 cell viability dose-dependently， and decreased anti-oxidative capacity， increased inflammation， and promoted cell death， possibly through modulating the NRF2 and NF-κB signalling pathways. Zebrafish embryos exposed to DA for four days from one day post fertilization (dpf) had an increase in mortality and a decrease in both hatching and heartbeat rate and exhibited morphological abnormalities. DA treatment also significantly downregulated expression of antioxidant genes and upregulated expression of inflammation mediators， as well as causing photoreceptor death in zebrafish embryos. The results demonstrate that consuming seafood containing DA will have potential toxic effects in human retinas.

Dynamic convergent shock compression initiated by return current in high-intensity laser-solid interactions

We investigate the dynamics of convergent shock compression in solid cylindrical targets irradiated by an ultrafast relativistic laser *** particle-in-cell simulations and coupled hydrodynamic simulations reveal that the compression process is initiated by both magnetic pressure and surface ablation associated with a strong transient surface return current with density of the order of 10^(17) A/m^(2) and lifetime of 100 *** results show that the dominant compression mechanism is governed by the plasma β,i.e.,the ratio of thermal pressure to magnetic *** targets with small radius and low atomic number Z,the magnetic pressure is the dominant shock compression *** to a scaling law,as the target radius and Z increase,the surface ablation pressure becomes the main mechanism generating convergent ***,an indirect experimental indication of shocked hydrogen compression is provided by optical shadowgraphy measurements of the evolution of the plasma expansion *** results presented here provide a novel basis for the generation of extremely high pressures exceeding Gbar(100 TPa)to enable the investigation of high-pressure physics using femtosecond J-level laser pulses,offering an alternative to nanosecond kJ-laser pulse-driven and pulsed power Z-pinch compression methods.

Differential roles of seed and sprout regeneration in forest diversity and productivity after disturbance

The Disc Acceleration e Xperiment(DAX)is one of the most recent experimental methods of performance characterization of new energetic materials.A cylindrical explosive charge accelerates a thin metallic disc and its velocity is measured continuously using photonic Doppler *** detonation velocity is measured *** DAX test can be used to obtain the Chapman-Jouguet(CJ)detonation pressure and to describe detonation products expansion using reduced amount of explosive.A series of DAX tests was performed at various charge diameters and disc thicknesses with Semtex 1 A plastic bonded explosive and sensitized *** DAX-like evaluation was also applied to previously measured data of Semtex 1A and A-IX-1 *** optimum disc thickness is determined by the disc to explosive mass ratio of *** repeatability of the Semtex 1 A detonation pressure results is about four times lower compared to the pressed and liquid explosives.

Fibrous dysplasia(FD)is a rare,disabling skeletal disease for which there are no established *** evidence supports inhibiting the osteoclastogenic factor receptor activator of nuclear kappa-B ligand(RANKL)as a potential treatment *** this study,we investigated the mechanisms underlying RANKL inhibition in FD tissue and its likely indirect effects on osteoprogenitors by evaluating human FD tissue pre-and post-treatment in a phase 2 clinical trial of denosumab(NCT03571191)and in murine in vivo and ex vivo preclinical *** analysis of human and mouse tissue demonstrated increased osteogenic maturation,reduced cellularity,and reduced expression of the pathogenic Gαs variant in FD lesions after RANKL *** sequencing of human and mouse tissue supported these *** interaction between osteoclasts and mutant osteoprogenitors was further assessed in an ex vivo lesion model,which indicated that the proliferation of abnormal FD osteoprogenitors was dependent on *** results from this study demonstrated that,in addition to its expected antiosteoclastic effect,denosumab reduces FD lesion activity by decreasing FD cell proliferation and increasing osteogenic maturation,leading to increased bone formation within *** findings highlight the unappreciated role of cellular crosstalk between osteoclasts and preosteoblasts/osteoblasts as a driver of FD pathology and demonstrate a novel mechanism of action of denosumab in the treatment of bone disease.

The reconstruction of images from their corresponding noisy Radon transform is a typical example of an ill-posed linear inverse problem as arising in the application of computerized tomography(CT).As the(naive)solution does not depend on the measured data continuously,regularization is needed to reestablish a continuous *** this work,we investigate simple,but yet still provably convergent approaches to learning linear regularization methods from *** specifically,we analyze two approaches:one generic linear regularization that learns how to manipulate the singular values of the linear operator in an extension of our previous work,and one tailored approach in the Fourier domain that is specific to *** prove that such approaches become convergent regularization methods as well as the fact that the reconstructions they provide are typically much smoother than the training data they were trained ***,we compare the spectral as well as the Fourier-based approaches for CT-reconstruction numerically,discuss their advantages and disadvantages and investigate the effect of discretization errors at differentresolutions.

Synchrophasor-based real-time state estimation and situational awareness system for power system operation

State estimation is a critical functionality of energy management system(eMS) to provide power system states in real-time operations. However, problems such as failure to converge, prone to failure during contingencies,and biased estimates while system is under stressed condition occur so that state estimation results may not be *** unreliable results further impact downstream network and market applications, such as contingency analysis,voltage stability analysis, transient stability analysis, system alarming, and unit commitment. Thus, operators may lose the awareness of system condition in eMS. This paper proposes a fully independent and one-of-a-kind system by integrating linear state estimator into situational awareness applications based on real-time synchrophasor data. With guaranteed and accurate state estimation solution and advanced real-time data analytic and monitoring functionalities, the system is capable of assisting operators to assess and diagnose current system conditions for proactive and necessary corrective actions. The architecture, building components, and implementation of the proposed system are explored in detail. Two case studies with simulated data from the subsystems of electric Reliability Council of Texas(eRCOT) and Los Angeles Department of Water and Power(LADWP) are presented. The test results show the effectiveness and reliability of the system, and its value for realtime power system operations.

Wear dependent virtual flow rate sensor for progressing cavity pumps with deformable stator

This contribution presents a novel wear dependent virtual flow rate sensor for single stage single lobe progressing cavity pumps. We study the wear-induced material loss of the pump components and the impact of this material loss on the volumetric efficiency. The results are combined with an established backflow model to implement a backflow calculation procedure that is adaptive to wear. We use a laboratory test setup with a highly abrasive fluid and operate a pump from new to worn condition to validate our approach. The obtained measurement data show that the presented virtual sensor is capable of calculating the flow rate of a pump being subject to wear during its regular operation.
