Despite emerging contemporary biotechnological methods such as gene-and stem cell-based therapy,there are no clinically established therapeutic strategies for neural regeneration after spinal cord *** previous studies have demonstrated that transplantation of genetically engineered human umbilical cord blood mononuclear cells producing three recombinant therapeutic molecules,including vascular endothelial growth factor(VEGF),glial cell-line derived neurotrophic factor(GDNF),and neural cell adhesion molecule(NCAm)can improve morpho-functional recovery of injured spinal cord in rats and *** investigate the efficacy of human umbilical cord blood mononuclear cells-mediated triple-gene therapy combined with epidural electrical stimulation in the treatment of spinal cord injury,in this study,rats with moderate spinal cord contusion injury were intrathecally infused with human umbilical cord blood mononuclear cells expressing recombinant genes VEGF165,GDNF,NCAm1 at 4 hours after spinal cord *** days after injury,epidural stimulations were given simultaneously above the lesion site at C5(to stimulate the cervical network related to forelimb functions)and below the lesion site at L2(to activate the central pattern generators)every other day for 4 *** subjected to the combined treatment showed a limited functional improvement of the knee joint,high preservation of muscle fiber area in tibialis anterior muscle and increased H/m ratio in gastrocnemius muscle 30 days after spinal cord ***,beneficial cellular outcomes such as reduced apoptosis and increased sparing of the gray and white matters,and enhanced expression of heat shock and synaptic proteins were found in rats with spinal cord injury subjected to the combined epidural electrical stimulation with gene *** study presents the first proof of principle study of combination of the multisite epidural electrical stimulation with ex vivo triple gene therapy(VEGF,GDNF and NCAm

During the last two decadesnumerous research teams demonstratedthat skeletalmuscles function as an exercise-dependent endocrine organ secreting dozens of *** of physiological and pathophysiological implications of skeletalmusclemyokines secretion has been described;however,upstream signals and sensing mechanisms underlying this phenomenon remain poorly *** is well documented that in skeletal muscles intensive exercise triggers dissipation of transmembrane gradient of monovalent cations caused by permanent activation of voltage-gated Nat and Kt ***,we demonstrated that sustained elevation of the[Nat]i/[Kt]i ratio triggers expression of dozens ubiquitous genes including several canonical myokines,such as interleukin-6 and cyclooxygenase 2,in the presence of intra-and extracellular Ca2t *** data allowed us to suggest a novel[Nat]i/[Kt]i-sensitive,Ca2t i-independent mechanism of excitation-transcription coupling which triggers myokine *** pathway exists in parallel with canonical signaling mediated by Ca2t i,AmP-activated protein kinase and hypoxia-inducible factor 1a(HIF-1a).In ourmini-reviewwe briefly summarize data supporting this hypothesis as well as unresolved issues aiming to forthcoming studies.

Objective: We sought to investigate the efficacy of oral dosing in mice with imipramine(7mg/kg/day) via water or in food pellets, and to compare its effects in the paradigms of learned helplessness, locomotion, hedonic state, and anxiety. methods: Water and food consumption were measured to determine daily imipramine dosage in C57BL/6N mice. Next, baseline scores for O-maze, dark/light box, and sucrose tests were measured. mice were then subjected to a 4-week treatment of voluntary ingestion of drinking water or food pellets containing imipramine. Lastly, all groups were subjected to novel cage, open field, O-maze, dark/light box, sucrose test, and forced swim test to assess the effects of the treatment. Results: In na?ve mice, imipramine delivered via food, induced a reduction of total floating and increased latency in the forced swim test, i.e., antidepressant-like effects. No other significant effects were found. Dosing with water did not change behavior in the forced swim, sucrose preference test, anxiety, or locomotor paradigms, but increased exploration in the novel cage. Conclusions: Voluntary ingestion is an effective method of chronic dosing with imipramine in na?ve mice. Delivery of imipramine with food pellets elicits antidepressant-like effects in the forced swim test, with no effects on anxiety, locomotion, or preference behaviors. In contrast, no such effects were observed with treatment via drinking water, suggesting that a higher dose may be required. Our work argues for a broader use of oral delivery using food-treated pellets, in small rodent models of pre-clinical depression. It may substantially improve animal welfare and overcome potential confounds in translational research, which are frequently associated with adverse chronic invasive pharmacotherapies.

Comparison of Debris Flow Characteristics Obtained by Using Video materials and modeling

Velocity of debris flow is one of the most important characteristics for the protective construction design. Since debris flows are rare events, and observations are conducted only on stations in Russia, Ukraine, Italy, Switzerland, USA, China, Japan and New Zealand, the velocity is calculated rather than measured. Nowadays, a large number of videos with passing debris flows have appeared on the Internet. Scientists can use such video materials to obtain qualitative and quantitative characteristics of the debris flow. Therefore, the aim of our research is an attempt to measure the debris flow velocity using video materials and compare the obtained results with the calculated values using various methods. The debris flow that came down in Firgen, Austria on August 4, 2012 was chosen as the object of our study. The video was carried out from several angles, so it was possible to select a section of the channel, through which we could measure the debris flows waves velocity. In addition, we calculated the velocities of waves by formulas adopted in the regulatory documents and compared with the measured by video values. During the video analysis, debris flow velocities at different sites were observed: minimum—7.2 m/s and maximum—10 m/s. The calculated values varied from 4.5 m/s to 11.4 m/s. moreover, we applied model of the transport-shear process of debris flow formation developed by Yu. B. Vinogradov. When we were comparing the obtained debris flow discharges with results from Austrian colleagues, we found out that the values were similar to each other. However, internal scatter in the model changed from 151 to 190 m3/s, while in the report of Austrian colleagues the discharges were from 80 to 250 m3/s.

One of the methods used in practice for the purification of wastewater from vegetable-processing enterprises is biological method of aerobic purification. With the aim to increase air-tank capacity used in traditional systems of biochemical purification of wastewater, different ways effecting to the process of vital activity of microorganisms of active sludge are applied. On the one hand, using the compositions of different salts and complex organic biogenous supplements prevents from the accumulation of some anions in the amounts exceeding the limited concentration, on the other hand- allows different groups of microorganisms to choose biogenous elements, which they assimilate easily and fully. It is known, that compounds of silicon and germanium can cause stimulating effects [1] to the organisms of active sludge. The goal of present studies is the intensification of biological processes of oxidation of organic contamination in wastewater by new bio-degradable chemical compounds or compositions on their basis, and by bio-sorbents.

We in vitro examined the existing prognoses of the dissociation constant, KD, between ТАТА- Binding Protein (TBP) and ТАТА box with single nucleotide polymorphism (SNP) associated with human diseases. Five SNPs of the genes for cytochrome P450 2A6 (associated with lung cancer), β-globin (associated with β-thalassemia), mannose binding lectin (associated with variable immunodeficiency), superoxide dismutase 1 (associated with amyotrophic lateral sclerosis) and triosephosphate isomerase (associated with anemia) fell within the range of –ln(KD;m/KD;WT) between –1.5 and –1 (here KD;WT and KD;m denote the normal ТАТА box and with SNP). The mea-surements using EmSA demonstrated that: 1) all the predictions stating that the affinity between ТВР and ТАТА boxes with SNPs would be reduced were correct;2) the departures of three predictions from the measurements fell within the confidence interval;3) all the predictions consistently underestimated actual mutational damage caused to ТАТА boxes with SNPs (a mial law) and two of these predictions did so significantly (a mation seems to be associated with the damage to the context that modulates ТВP/ТАТА affinity, for example, the contact between the nucleosomal histone H3-Н4 dimer and the core promoter immediately near ТАТА boxes.

Using a novel small-angle X-ray scattering (SAXS) method for determination of fractional and subfractional composition of lipoproteins (LPs), a significant elevation of total cholesterol-lipop- roteins (C-LP) and, especially, total triglyceride- lipoproteins (TG-LP), was shown in this work. Among the LP fractions, poloxamer 407 was shown to significantly increase proatherogenic total C-LDL, TG-LDL and, especially, their precursors C-VLDL and TG-VLDL, while only exhibiting a moderate increase in the antiatherogenic C-HDL and TG-HDL fractions. With regard to the VLDL subfractions, significant elevations were observed in both subfractions studied;namely, C-VLDL1-2 and C-VLDL3-5. Similar chang- es were noted in the TG-VLDL1-2 and TG- VLDL3-5 subfractions. The C-IDL and TG-IDL subfractions were increased significantly (?20- to 30- fold), while the C-LDL1-3 subfraction was moderately (?3- to 5-fold) increased at 48 hrs and at day 4. In the moderately elevated (?2- to 4-fold) anti-atherogenic HDL fraction, the C-HDL2 subfraction was increased more significantly (?4- fold) compared to the C-HDL3 subfraction;how- ever, both C-HDL subfractions returned to base- line by day 4. The elevation in the TG-HDL2 subfraction was observed only at 24 hrs. mouse models of hyperlipidemia and atherosclerosis are useful to evaluate the role of “individual” LPs, as well as their fractions and subfractions, in hyperlipidemia and the genesis of atherosclerosis.
