Polychromatic Fourier-PAM fluorometry and hyperspectral analysis of chlorophyll fluorescence from Phaseolus vulgaris leaves: Effects of green light

Maximal and partial quantum yields of photosystem II(Fv/Fm and Fv–/Fp)in Phaseolus vulgaris leaves were determined in response to the turning on saturating and low(undersaturating)actinic light,*** lights(ML)of blue,green,and red colors were applied simultaneously using a novel method of the polychromatic fast Fourier transforming pulse amplitude modulation(FFT PAM)chlorophyll *** of ML were cross-combined with the colors of low and saturating actinic ***/Fm values measured with the green ML were found to be close to that of red light,whereas application of blue ML leads to lower Fv/*** addition,5-nm resolved excitation spectra were measured to evaluate the dependence between red/far red fluorescence ratios(peak height based–F685/F730,and deconvolved peak square based–F685sq/F730sq)and wavelength of the excitation *** demonstrates that chlorophyll fluorescence ratio red/far red may be dependent on the spectral absorbance of *** data obtained confirm the results of the previous studies which explain high photosynthetic activity of the green light in terms of redistribution of absorbed quanta throughout thickness of the leaf,thus suggesting possible advisability of green light application in greenhouse lighting,especially for the plants having thick leaves.

Peripheral blood lymphocytes DNA in patients with chronic liver diseases

BACKGROUND: Viral replication in blood cells with nucleases may lead to the damage of lymphocytes genetic apparatus and the beginning of immunopathological reactions. AIM: Of this investigation is to reveal the damage to peripheral blood lymphocytes (PBL) DNA in the patients with chronic liver diseases. MATERIALS AND METHODS: Sixteen-nine patients with chronic liver diseases (37 patients with chronic viral hepatitis, 2 patients with liver cirrhosis of mixed etiology (alcohol+virus G), 30 women with primary biliary cirrhosis-PBC) were examined. The condition of DNA structure of PBL was measured by the fluorescence analysis of DNA unwinding (FADU) technique with modification. Changes of fluorescence (in %) reflected the DNA distractions degree (the presence of DNA single-stranded breaks and alkalinelabile sights). RESULTS AND CONCLUSION: The quantity of DNA single-stranded breaks and alkalinelabile sights in DNA in all patients with chronic viral hepatitis didn't differ from the control group, excluding the patients with chronic hepatitis (CH) C+G. Patients with HGV and TTV monoinfection had demonstrated the increase of the DNA single-stranded breaks PBL quantity. This fact may be connected with hypothesis about the viruses replication in white blood cells discussed in the literature. Tendency to increase quantity of DNA PBL damages in the patients with primary biliary cirrhosis (PBC) accordingly to the alkaline phosphatase activity increase was revealed. Significant decrease of the DNA single-stranded breaks and alkalinelabile sights in the PBC patients that were treated with prednisone was demonstrated. Probably, the tendency to increase the quantity of DNA single stranded breaks and alkalinelabile sights in lymphocytes of the PBC patients was depended on the surplus of the blood bile acid content.

Characteristics of Fruit Quality of Peach Cultivars and Hybrids in the Steppe Crimea

The study of peach fruit quality and biochemical composition of cultivars and hybrids were conducted in terms of the steppe zone of Crimea. A total of 40 cultivars and hybrids from five periods of ripening were analyzed for content of basic biochemical components： solids, sugars, acids, polyphenolic compounds and vitamin C. The main fruit quality parameters： the size, the attractive appearance, the consistency flesh, the stone adherence to flesh, taste and others have been evaluated. As a result of studies peach cultivars with a high content of sugar in the fruit under the steppe Crimea have highlighted： Kremlyovskiy （13.6）, Regina （13.2）, Velvet （12.9）, hybrids C7/38 （13.1） and C4/57 （12.9）. Cultivars and hybrids of peach 10607, C1/227 C7/38, C7/6, C8/162, C4/147, C16/87, C4/64, Early Red Haven, Miorita, 7/221, Kremlyovskiy, 10848 are characterized by the best biochemical and morphological indices of fruits. They are recommended for State cultivar testing or use in breeding work to improve the quality of peach fruits.

Report of the restudy of the defined global stratotype of the base of the Silurian System

The Global Stratotype Section and Point （GSSP） for the base of the Silurian System was defined in 1985. Since that time, a number of researchers have suggested that this section has serious deficiencies for use as a GSSP. As a result, in 2000, the Subcommission on Silurian Stratigraphy （SSS） proposed undertaking a formal restu@ of this GSSP, which was subsequently approved by the International Commission of Stratigraphy （ICS）. The result of this restudy was the formal proposal that the current GSSP, at 1.6 m above the base of the Birkhill Shale, at Dob＇s Linn, Scotland, should be maintained as the same locality and stratigraphic level, but the bios- tratigraphical definition of the boundary should be revised. The previously defined, basal Silurian grapto- lite zone, the Parakidograptus acuminatus Zone, should be subdivided into a lower Akidograptus ascensus Zone and a higher E acuminatus Zone. The base of the A. ascensus Zone, marked by the first appearance of A. ascensus, should be regarded as the biostratigraphic mark for the base of the Silurian. This proposal has now been formally approved by the SSS and ICS, and ratified by International Union of Geological Sciences. This is the first GSSP to undergo formal restudy and redefini- tion, and the proposal also included a recommended procedure for restudy of GSSPs.

A Host KH RNA-Binding Protein Is a Susceptibility Factor Targeted by an RXLR Effector to Promote Late Blight Disease

Plant pathogens deliver effector proteins that alter host processes to create an environment conducive to colonization. Attention has focused on identifying the targets of effectors and how their manipulation facil- itates disease. RXLR effector Pi04089 from the potato blight pathogen Phytophthora infestans accumu- lates in the host nucleus and enhances colonization when transiently expressed in planta. Its nuclear local- ization is required for enhanced P. infestans colonization. Pi04089 interacts in yeast and in planta with a putative potato K-homology （KH） RNA-binding protein, StKRBPI. Co-localization of Pi04089 and StKRBP1, and bimolecular fluorescence complementation between them, indicate they associate at nuclear speckles. StKRBP1 protein levels increased when it was co-expressed with Pi04089. Indeed, such accumu- lation of StKRBP1 was observed also on the first day of leaf colonization by the pathogen. Remarkably, overexpression of StKRBP1 significantly enhances P. infestans infection. Mutation of the nucleotide- binding motif GxxG to GDDG in all three KH domains of StKRBP1 abolishes its interaction with Pi04089, its localization to nuclear speckles, and its increased accumulation when co-expressed with the effector. Moreover, the mutant StKRBP1 protein no longer enhances leaf colonization by P. infestans, implying that nucleotide binding is likely required for this activity. We thus argue that StKRBP1 can be regarded as a sus- ceptibility factor, as its activity is beneficial to the pathogen.
