Investigation of the Effect of Ambient Temperature and Geomagnetic Activity on the Vascular Parameters of Healthy Volunteers

Aim of this paper is to reveal whether the geomagnetic activity (GMA) and meteorological factors (MFs) affect vascular parameters of healthy volunteers. As a trial study we used new device “Tonocard,” and new vascular parameters for study—a pulse wave velocity (PWV) and an endothelial function (EnF) in addition to blood pressure measurements. These parameters never investigated before in such aspects. As far as novelty of device itself and investigated parameters we limited ourselves by monitoring only four healthy volunteers (without cardiovascular pathology). To analyze the sensitivity of their aforementioned medical indices to GMA and MFs two independent mathematical approaches were used, one of whom is based on traditional methods of mathematical statistics and the other on the theory of pattern recognition Dependence of physiological characteristics on the atmospheric temperature, revealed by both applied mathematical approaches, showed complex non-linear character of biological replies: the reaction has a different form in different temperature ranges and is manifested in the form of synchronization of slow variations of physiological and atmospheric parameters (trends) with a period of several days, while the daily variations were virtually independent. The systolic blood pressure (SBP), PWV and a difference between two specially selected values of PWV (DPWV) are approximately equally depending on atmospheric temperature, which accounts for an average of 26% to 28% of their variations. Sensitivity to the GMA for this test was found only for PWV.

Role of Helicobacter pylori infection in gastric carcinogenesis:Current knowledge and future directions

Helicobacter pylori(H. pylori) plays a role in the patho-genesis of gastric cancer. The outcome of the infection depends on environmental factors and bacterial and host characteristics. Gastric carcinogenesis is a multistep process that is reversible in the early phase of mucosal damage, but the exact point of no return has not been identified. Therefore, two main therapeutic strategies could reduce gastric cancer incidence:(1) eradication of the already present infection; and(2) immunization(prior to or during the course of the infection). The success of a gastric cancer prevention strategy depends on timing because the prevention strategy must be introduced before the point of no return in gastric carcinogenesis. Although the exact point of no return has not been identified, infection should be eradicated before severe atrophy of the gastric mucosa develops. Eradication therapy rates remain suboptimal due to increasing H. pylori resistance to antibiotics and patient noncompliance. Vaccination against H. pylori would reduce the cost of eradication therapies and lower gastric cancer incidence. A vaccine against H. pylori is still a research challenge. An effective vaccine should have an adequate route of delivery, appropriate bacterial antigens and effective and safe adjuvants. Future research should focus on the development of rescue eradication therapy protocols until an efficacious vaccine against the bacterium becomes available.

Comparative Evaluation of Peptidases Produced by Penicillium corylophilum and Penicillium waksmanii in Solid State Fermentation Using Agro-industrial Residues

In this present work, the best conditions for production of peptidases under solid state fermentation by the fungi Penicillium corylophilum and Penicillium waksmanii, partial purification using Sephadex G-75 gel filtration column, as well as the biochemical characterization of the partial purified enzymes were investigated. P. corylophilum showed the best production in medium containing wheat bran, agro-industrial residue, without additives （egg albumin or casein）, in which peptidase activity reached 520 U mL^-1 and the enzyme displayed the optimum activity between pH range from 7 to 8 and 60 ℃. It also showed high stability in wide pH range and temperature until 45 ℃ for 60 min of incubation. On the other hand, P. waksmanii, the best production was noted in a medium containing wheat bran （95%） and casein （5%）, reaching 545 U mL^-1, with proteolytic optimum activity at pH 7.5 and 55 ℃. The enzyme was mainly stable in pH range from 8 to 9 and at temperatures until 45 ℃ for 60 rain of incubation. The peptidases secreted by both fungi were inhibited in the presence of phenylmethane sulfonyl fluoride, showing that they belong to the subclass of serine peptidases.

A continuously or/and discontinuously tunable LiNbO3 Optical Parametric Oscillator (OPO) with the spectral range of 1.42 - 4.24 um and shifting of 0…12 nm is created and investigated. The OPO resonator ring circuit provides for the radiation energy output value of up to 50 mJ. Radiation bandwidth narrowing of up to 0.7 cm–1 by introducing of the Fabry-Perot etalon into the OPO resonator has been obtained.

Increased density of tolerogenic dendritic cells in the small bowel mucosa of celiac patients

AIM: To investigate the densities of dendritic cells(DCs) and FOXP3+ regulatory T cells(Tregs) and their interrelations in the small bowel mucosa in untreated celiac disease(CD) patients with and without type 1 diabetes(T1D).METHODS: Seventy-four patients(45 female, 29 male, mean age 11.1 ± 6.8 years) who underwent small bowel biopsy were studied. CD without T1 D was diagnosed in 18 patients, and CD with T1 D was diagnosed in 15 patients. Normal small bowel mucosa was found in two T1 D patients. Thirty-nine patients(mean age 12.8 ± 4.9 years) with other diagnoses(functional dyspepsia, duodenal ulcer, erosive gastritis, etc.) formed the control group. All CD patients had partial or subtotal villous atrophy according to the Marsh classification: Marsh grade Ⅲa in 9, grade Ⅲb in 21 and grade Ⅲc in 3 cases. Thirty-nine patients without CD and 2 with T1 D had normal small bowel mucosa(Marsh grade 0). The densities of CD11c+, IDO+, CD103+, Langerin(CD207+) DCs and FOXP3+ Tregs were investigated by immunohistochemistry(on paraffin-embedded specimens) and immunofluorescence(on cryostat sections) methods using a combination of mono- and double-staining. Sixtysixserum samples were tested for Ig A-tissue transglutaminase(t TG) using a fully automated Eli ATM Celikey Ig A assay(Pharmacia Diagnostics, Freiburg, Germany). RESULTS: The density of CD11c+ DCs was significantly increased in CD patients compared with patients with normal mucosa(21.67 ± 2.49 vs 13.58 ± 1.51, P = 0.007). The numbers of FOXP3+ cells were significantly higher in CD patients(10.66 ± 1.50 vs 1.92 ± 0.37, P = 0.0002) and in patients with CD and coexisting T1D(8.11 ± 1.64 vs 1.92 ± 0.37, P = 0.002) compared with patients with normal mucosa. The density of FOXP3+ cells significantly correlated with the histologicalgrade of atrophic changes in the small bowel mucosa according to the March classification(r = 0.62; P < 0.0001) and with levels of Ig A antibody(r = 0.55; P < 0.0001). A Quantitative Analysis of Dose Distributions of Two Tangential Whole Breast Irradiation Techniques

We compared DVHs （dose volume histograms） of two tangential irradiation techniques for whole breast RT （radiotherapy） CT （computed tomography） based 2D （two dimensional） technique with wedge filters and 3D CRT （three dimensional conformal radiotherapy） with segmented fields. These two modalities are commonly used in external beam radiotherapy of breast cancer in Clinical Center Nis, Clinic of Oncology, Serbia. Therapy plans that employed two techniques were generated for eighteen patients. Dosimetric outcomes of each technique were investigated. The maximum dose of breast CTV was significantly reduced from 55.83 Gy for 2D to 54.1 Gy for 3D CRT. Also, the maximum dose of lung tissue decreased from 51.81 Gy for 2D to 49.61 Gy for 3D. The dose conformity and uniformity were much better with 3D CRT segmented field technique.

Colonization of germ-free mice with a mixture of three lactobacillus strains enhances the integrity of gut mucosa and ameliorates allergic sensitization

Increasing numbers of clinical trials and animal experiments have shown that probiotic bacteria are promising tools for allergy prevention. Here, we analyzed the immunomodulatory properties of three selected lactobacillus strains and the impact of their mixture on allergic sensitization to Bet v I using a gnotobiotic mouse model. We showed that Lactobacillus （L.） rhamnosus LOCK0900, L. rhamnosus LOCK0908 and L. casei LOCK0919 are recognized via Toll-like receptor 2 （TLR2） and nucleotide-binding oligomerization domain-containing protein 2 （NOD2） receptors and stimulate bone marrow-derived dendritic cells to produce cytokines in species- and strain-dependent manners. Colonization of germ-free （GF） mice with a mixture of all three strains （Lmix） improved the intestinal barrier by strengthening the apical junctional complexes of enterocytes and restoring the structures of microfilaments extending into the terminal web. Mice colonized with Lmix and sensitized to the Bet v I allergen showed significantly lower levels of allergen-specific IgE, IgG 1 and IgG2a and an elevated total IgA level in the sera and intestinal lavages as well as an increased transforming growth factor （TGF）-β level compared with the sensitized GF mice. Splenocytes and mesenteric lymph node cells from the Lmix-colonized mice showed the significant upregulation of TGF-β after in vitro stimulation with Bet v 1. Our results show that Lmix colonization improved the gut epithelial barrier and reduced allergic sensitization to Bet v 1. Furthermore, these findings were accompanied by the increased production of circulating and secretory IgA and the regulatory cytokine TGF-β. Thus, this mixture of three lactobacillus strains shows potential for use in the prevention of increased gut permeability and the onset of allergies in humans,

Variability of anti-human transglutaminase testing in celiac disease across Mediterranean countries

To verify the precision and accuracy of transglutaminase antibodies (TGA) assays across Mediterranean countries. METHODSThis study involved 8 referral centres for celiac disease (CD) in 7 Mediterranean countries. A central laboratory prepared 8 kits of 7 blinded and randomized serum samples, with a titrated amount of Human TGA IgA. Each sample was analysed three times on three different days, with each centre running a total of 21 tests. The results were included in a blindly coded report form, which was sent to the coordinator centre. The coordinator estimated the mean coefficient of Variation (CoVar = σ/μ), the mean accuracy (Accur = Vobserved - Vreal) and the mean percent variation (Var% = [(Vobserved - Vreal)/Vreal] × 100). RESULTSThe analysis showed that 79.17% of the mean variation fell between -25% and +25% of the expected value, with the accuracy and precision progressively increasing with higher titres of TGA. From values 1.25 times greater than the normal cut-off, the measurements were highly reliable. CONCLUSIONTGA estimation is a crucial step for the diagnosis of CD; given its accuracy and precision, clinicians could be confident in establishing a diagnosis.
