Relative importance of biological and human-associated factors for alien plant invasions in Hokkaido, Japan

Aims The invasion success of alien plants is strongly affected by both biological and human-associated *** of the rela-tive contribution of each factor is important not only for the further understanding of invasion processes but also for the better manage-ment of invasion risk,particularly in protected areas of high con-servation ***,we quantified the relative importance of species biological traits and association with a human activity,***,in explaining the invasion success of alien plants across the entire region and in protected areas in Hokkaido,*** As a quantitative measure of invasion success,the distribution ex-tent of naturalized populations across the entire prefecture and in protected areas was calculated for 63 alien species with equal residence time based on species occurrence records at a spatial resolution of 5-km mesh grid *** each species,we identi-fied seven biological traits(seed mass,dispersal mode,maximum plant height,capability of vegetative reproduction,flowering start time and period and life span)and two human-associated factors(introduction purpose and cultivation frequency for agricultural use).Cultivation frequency was determined based on the frequency of seed-sowing in pastures:(1)not sown,(2)accidentally sown as a seed contaminant and(3)intentionally sown for commercial *** importance of biological traits and human-associated factors in explaining the distribution extent was determined using an information-theoretic *** Findings In explaining the distribution extent across the entire prefecture,species biological traits and human-associated factors showed comparable importance;cultivation frequency exhibited the high-est importance value closely followed by seed mass,maximum height and flowering *** contrast,when focusing on pro-tected areas,human association was more important than bio-logical traits,as indicated by the greatest importance of cultiva

Arginine kinase in Toxocara canis:Exon-intron organization,functional analysis of site-directed mutants and evaluation of putative enzyme inhibitors

Objective: To determine exon/intron organization of the Toxocara canis(T. canis) AK(TCAK) and to test green and black tea and several other chemicals against the activity of recombinant TCAK in the guanidino-specific region by site-directed mutants. Methods: Amplification of genomic DNA fragments containing introns was carried out by PCRs. The open-reading frame(1 200 bp) of TCAK(wild type) was cloned into the BamH 1/SalI site of pM AL-c2X. The maltose-binding protein-TCAK fusion protein was expressed in Escherichia coli TB1 cells. The purity of the expressed enzyme was verified by SDS-PAGE. Mutations were introduced into the guanidino-specific region and other areas of pM AL/TCAK by PCR. Enzyme activity was measured with an NADH-linked assay at 25℃ for the forward reaction(phosphagen synthesis). Results: Arginine kinase in T. canis has a seven-exon/six-intron gene structure. The lengths of the introns ranged from 542 bp to 2 500 bp. All introns begin with gt and end with ag. Furthermore, we measured the enzyme activity of site-directed mutants of the recombinant TCAK. The K_m value of the mutant(Alanine to Serine) decreased indicating a higher affinity for substrate arginine than the wild-type. The K_m value of the mutant(Serine to Glycine) increased to 0.19 mM. The Km value(0.19 mM) of the double mutant(Alanine-Serine to Serine-Glycine) was slightly greater than in the wild-type(0.12 mM). In addition, several other chemicals were tested; including plant extract Azadiracta indica(A. indica), an aminoglycoside antibiotic(aminosidine), a citrus flavonoid glycoside(rutin) and a commercially available catechin mixture against TCAK. Green and black tea(1:10 dilution) produced 15% and 25% inhibition of TCAK, respectively. The extract of A. indica produced 5% inhibition of TCAK. Moreover, green and black tea produced a non-competitive type of inhibition and A. indica produced a mixed-type of inhibition on TCAK. Conclusions: Arginine kinase in T. canis has a seven-exon/six

Potential antioxidant bioactive peptides from camel milk proteins

Camel milk is traditionally considered to have medicinal characteristics that it has potential health benefits and could help to treat several illnesses. Particularly, it is closest to human breast milk and has high levels of nutrients and bioactive components. The aim of this study was to explore the antioxidant peptides derived from protein fractions of camel milk. Camel milk proteins(CMP) were fractionated into camel casein protein(CCP) and camel whey protein(CWP), which were hydrolyzed with pepsin to produce peptic digests P-CCP and P-CWP, respectively. RP-HPLC was used for fractionation of the peptides from the P-CCP and P-CWP. The antioxidant activities were evaluated using superoxide anion generating system of xanthine oxidase(XOD)and 2,2-diphenyl-1-picrylhydrazyl(DPPH) scavenging assay. Active peptides were analyzed using matrixassisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS) whereas a number of antioxidant peptides, with masses ranging from 913 to 2,951 Da, derived mainly from alpha-casein, lactophorin and lactoferrin, were identified. When yeast cells are used as a system for modeling mitochondrial disease, the peptides in caseins and whey fractions significantly enhanced the tolerance of yeast cells against peroxide-induced oxidative stress. The results show that both caseins and whey proteins of camel milk possess bioactive peptides with significant radical-scavenging activities and thus herald a fascinating opportunity for their potential as nutraceuticals or therapeutic peptides for prevention and treatment of oxidative stress-associated diseases.

Ectopic porcine spermatogenesis in murine subcutis： tissue grafting versus cell-injection methods

Fragments of testis tissue from immature animals grow subcutaneous areas of immunodeficient mice. The same results testes of rodents are injected into the subcutis of nude mice. and develop spermatogenesis when grafted onto are obtained when dissociated cells from immature Those cells reconstitute seminiferous tubules and facilitate spermatogenesis. We compared these two methods, tissue grafting and cell-injection methods, in terms of the efficiency of spermatogenesis in the backs of three strains of immunodeficient mice, using neonatal porcine testicular tissues and cells as donor material. Nude, severe combined immunodeficient （SCID） and NOD/Shi- SCID, IL-2Rγc^null （NOG） mice were used as recipients. At 10 months after surgery, the transplants were examined histologically. Both grafting and cell-injection methods resulted in porcine spermatogenesis on the backs of recipient mice; the percentage of spermatids present in the transplants was 67% and 22%, respectively. Using the grafting method, all three strains of mice supported the same extent of spermatogenesis. As for the cell- injection method, although SCID mice were the best host for supporting reconstitution and spermatogenesis, any difference from the other strains was not signifcant. As NOG mice did not show any better results, the severity of immunodeficiency seemed to be irrelevant for supporting xeno-ectopic spermatogenesis. Our results confirmed that tubular reconstitution is applicable to porcine testicular cells. This method as well as the grafting method would be useful for studying spermatogenesis in different kinds of animals.

Mast cells are the main effector cells in IgE-associated allergic disorders,and we have reported that mucosal mast cells(MMCs)play a more important role in the development of food allergy(FA).IgE with antigen or calcium ionophore stimulation can lead to the activation of MMCs via a calcium-dependent *** purpose of the present study was to identify gene signatures with IgE/antigen(dinitrophenyl-bovine serum albumin,DNP-BSA)or calcium ionophore(A23187)on the activation of *** expressed genes between the two types of samples were identified with microarray *** ontology functional and pathway enrichment analyses of differentially expressed genes were performed using the database for annotation,visualization,and integrated discovery *** results showed that IgE/antigen and A23187 could induce degranulation,increase vacuoles,and elevate the cytosolic calcium concentration in ***,GeneChip analysis showed that the same 134 mRNAs were altered with IgE/DNP-BSA and A23187,suggesting that DNP-BSA/IgE and A23187 affect the same signal pathway partly in *** analysis showed that the data were enriched in NF-κB,TNF,MAPK,transcription factor activity,DNA binding,and nucleic acid binding,suggesting that activation of MMCs is a complex *** results provide new insights on MMCs activation.

Helicobacter pylori-negative,non-steroidal anti-inflammatory drug:Negative idiopathic ulcers in Asia

Since the discovery of Helicobacter pylori(***)infection in the stomach,the bacteria infection and non-steroidal anti-inflammatory drugs(NSAIDs)use had been considered to be the 2 main causes of peptic ***,there have been recent reports of an increase in the proportion of peptic ulcers without these known risk factors;these are termed idiopathic peptic *** trend was firstly indicated in 1990s from some reports in North *** Asia,numerous studies reported that idiopathic ulcers accounted for a small percentage of all ulcers in the 1990s,but in the2000s,multiple studies reported that the proportion of idiopathic ulcers had reached 10%-30%,indicating that the incidence of idiopathic ulcers in Asia has also been rising in recent *** a decline in *** infection rates of general population in Asia is seen as the main reason for the increased incidence of idiopathic ulcers,it is also possible that the absolute number of idiopathic ulcer cases has *** age,serious systemic complication,and psychological stress are considered to be the potential risk factors for idiopathic *** of idiopathic ulcers is challenging,at present,because there is no effective preventative measure against recurrence in contrast with cases of ***-positive ulcers and NSAIDs-induced *** it is expected that *** infection rates in Asia will decline further in the future,measures to treat idiopathic ulcers will also likely become more important.

Potent antioxidant peptides derived from honey major protein enhance tolerance of eukaryotic cells toward oxidative stress

Honey is known for its medicinal benefits and receiving renewed attention as natural *** on health benefits of honey attributed its antioxidant activity to phenolic compounds,but the contribution of proteins and peptides to the antioxidant activity of honey is *** aim of this study was to explore the contribution of proteins and peptides to the antioxidant activity of honey,which remained obscure for *** honey proteins(THP)were isolated by dialysis method and hydrolyzed through simulated gastrointestinal *** hydrolysates were fractionated using size-exclusion *** antioxidant activity was determined by using superoxide radical-scavenging,DPPH reduction and intracellular ROS *** was shown to exhibit superoxide-scavenging activity but its pepsin-hydrolysate(HP-p)showed superior scavenging *** HP-p produced five peptide fractions(P1~P5)when fractionated on Sephacryl S-100 size-exclusion *** five fractions showed superoxide-scavenging activities and DPPH reducing activities,whereas the slow-eluting peptide fractions(P3 and P4)were the most ***-TOF/MS analysis identified a pentapeptide(TSNTF)as the dominant peptide in the active fractions P3 and *** colonic epithelial cells treated with P3 and P4 peptides exhibited lower intracellular ROS,when oxidative stress was induced by H2O2 or diethyl maleate(DEM),indicating strong tolerance to oxidative *** viabilities of human cells or yeast cells were largely decreases under oxidative stress,but treated cells with P3 and P4 showed higher viability compared with the untreated *** results are the first to describe a novel antioxidant peptide from honey that confer ex vivo anti-oxidative function within a complicated milieu of eukaryotic cells and pave the way for its potential as nutraceutical or therapeutic peptide for risk-reduction of oxidative-stress and related diseases.

The last appearance datum of the radiolarian Kilinora spiralis is recorded above the first appearance datum of the ammonite Ataxioceras (A.) kurisakense in the Todoro Section of the Kurisaka Formation, Southern Kurosegawa Terrane, Shikoku, SW Japan. The constraint by ammonite age prolongs the range of the Kilinora spiralis Zone, a remarkable Jurassic radiolarian zone in Japan-NW Pacific region, into the lower Kimmeridgian. The direct correlation of the Kilinora spiralis zone with the Late Jurassic ammonite faunal succession in the Kurisaka Formation will provide a clue to the still pending chronological difference between European and North American radiolarian zones.

Distinctive Precursory Air–Sea Signals between Regular and Super El Ni os

Statistically different precursory air–sea signals between a super and a regular El Ni no group are investigated, using observed SST and rainfall data, and oceanic and atmospheric reanalysis data. The El Ni no events during 1958–2008 are first separated into two groups： a super El Ni no group（S-group） and a regular El Ni no group（R-group）. Composite analysis shows that a significantly larger SST anomaly（SSTA） tendency appears in S-group than in R-group during the onset phase[April–May（0）], when the positive SSTA is very small. A mixed-layer heat budget analysis indicates that the tendency difference arises primarily from the difference in zonal advective feedback and the associated zonal current anomaly（u）.This is attributed to the difference in the thermocline depth anomaly（D） over the off-equatorial western Pacific prior to the onset phase, as revealed by three ocean assimilation products. Such a difference in D is caused by the difference in the wind stress curl anomaly in situ, which is mainly regulated by the anomalous SST and precipitation over the Maritime Continent and equatorial Pacific.

Does a combined intervention program of repetitive transcranial magnetic stimulation and intensive occupational therapy affect cognitive function in patients with post-stroke upper limb hemiparesis?

Low-frequency repetitive transcranial magnetic stimulation（LF-r TMS） to the contralesional hemisphere and intensive occupational therapy（i OT） have been shown to contribute to a significant improvement in upper limb hemiparesis in patients with chronic stroke. However, the effect of the combined intervention program of LF-r TMS and i OT on cognitive function is unknown. We retrospectively investigated whether the combined treatment influence patient＇s Trail-Making Test part B（TMT-B） performance, which is a group of easy and inexpensive neuropsychological tests that evaluate several cognitive functions. Twenty-five patients received 11 sessions of LF-r TMS to the contralesional hemisphere and 2 sessions of i OT per day over 15 successive days. Patients with right- and left-sided hemiparesis demonstrated significant improvements in upper limb motor function following the combined intervention program. Only patients with right-sided hemiparesis exhibited improved TMT-B performance following the combined intervention program, and there was a significant negative correlation between Fugl-Meyer Assessment scale total score change and TMT-B performance. The results indicate the possibility that LF-r TMS to the contralesional hemisphere combined with i OT improves the upper limb motor function and cognitive function of patients with right-sided hemiparesis. However, further studies are necessary to elucidate the mechanism of improved cognitive function.
