Assessment of Endometrial Carcinoma Markers and Hormone Receptors Profile before and after Bariatric Surgery: A Clinico-Pathological-Immunohistochemical Study

Background: Obesity is a major risk factor for endometrial carcinoma, and we aim to assess markers of carcinogenesis including PTEN and Ki-67 and hormone receptors profile including ER, PR and AR before and after bariatric surgery to find out its effects in reducing endometrial carcinoma risk in morbid obese females. Patients and methods: The study included 80 females with morbid obesity (BMI > 40 Kg/m2) who underwent bariatric surgery. All were sampled by Pipelle biopsy at baseline and 12 months after operation and examined histopathologically and immunohistochemically for Ki-67, PTEN, ER, PR and AR. Results: Sixty two out of 80 (62/80) females showed no pathological abnormalities;4 had polyps;7 had simple endometrial hyperplasia;4 had atypical endometrial hyperplasia and 3 had endometrial carcinoma. In total, 34 females underwent gastric bypass operation (42.5%) and 46 underwent a sleeve gastrectomy operation (57.5%). There was a statistically significant difference between baseline weight and BMI before and after surgery (p < 0.001). Of the 7 women with simple hyperplasia, resolution occurred in 5 within 7 months of surgery. Three of 4 females with atypical hyperplasia (AH) showed resolution after 9 months. Mean Ki-67 score was lower at 12 months (p < 0.001) after surgery. 43/77 (55.8%) baseline biopsies were glandular PTEN null, including 9/15 of the women with baseline endometrial abnormalities, of whom 5/15 regained glandular PTEN expression as their endometrial abnormalities resolved. There was a significant reduction in ER score after surgery (p < 0.001). PR H-scores were not significantly different post-operatively (p = 0.193). AR H-scores were higher significantly in pre-operative biopsies than post-operative ones (p < 0.001). Conclusion: Females with morbid obesity have a higher risk of harboring endometrial abnormalities even if asymptomatic. Utilization of Cement Kiln Dust (CKD) with Silica Fume (SF) as a Partial Replacement of Cement in Concrete Production

This research aimed to clarify the role of by-product materials, such as CKD with SF as partial replacement by weight of cement in concrete manufacturing and inclusion on different characteristics of concrete. Concrete test specimens were mixed with 0%, 5%, 10%, 15%, 20% and 25% (CKD) with 15% (SF) as partial replacement by weight of Cement (CEM I-52.5N). Fresh concrete properties have been evaluated by workability measurement slump test. While hardened concrete properties have been evaluated by compressive, split tensile and flexural strengths tests at ages 7, 28 and 56 days, but evaluated for bond strength, modulus of elasticity and chemical composition measurement with X-Ray Fluorescence at age of 28 days. The test results have revealed that the increase of CKD amount with fixed amount of SF in concrete mixtures as partial replacement by weight of cement leads to gradual decrease of fresh concrete workability. In concrete mixtures, 20% CKD in the presence of 15% SF as partial replacement by the weight of cement are the optimum ratios which can be used without any negative effect on mechanical properties compressive, indirect tensile, flexural and bond strength at all the ages of concrete. Also modulus of elasticity and bond strength increased by 8.81% and 0.69% respectively at the age 28 days compared with control mixture.

Molecular docking of amphetamine,cathine and cathinone with dihydrofolate reductase:a computational analysis of inhibition of dihydrofolate reductase by khat alkaloids

Interaction of amphetamine,cathine and cathinone with the enzyme dihydrofolate reductase was studied by molecular docking using AutoDock 4.2 as the docking software *** 4.2 software serves as a valid and acceptable docking application to study the interactions of small compounds with *** of amphetamine,cathine and cathinone with dihydrofolate reductase were compared to those of methotrexate,a known inhibitor of the *** calculated free energy of binding(ΔG binding)shows that the three ligands(ΔG=-6.87 to-7.21 kcal/mol;Ki=9.15 to 5.18μM)bind with affinity slightly lower than methotrexate(ΔG=-8.78 kcal/mol;Ki=363 nM).Binding interactions of the three ligands with active site residues of the enzyme are also *** the ligands appear to bind in a similar conformation making extensive VDW contacts in the active site of the *** bonding and pi-pi interaction with key active site residues is also ***,a probable inhibition of dihydrofolate reductase by khat alkaloids can be explained on the basis of this in silico binding and khat alkaloids can be considered as potential lead compounds in the development of new inhibitors of dihydrofolate reductase which is a potential target of anti-cancer *** results of these studies can serve as a starting point for further computational and experimental studies.
