Asymmetric reduction of the conjugated C=C bonds by the old yellow enzymes(OYEs)presents a promising field in the synthesis of chiral ***,few natural OYEs have been applied in large-scale applications due to the requirement of costly NADPH and low operational ***,a stable and efficient fusion of YqjM from Bacillus subtilis and glucose dehydrogenase(GDH)from Bacillus megaterium was constructed to stereoselectively reduce the conjugated C=C bonds in a self-sufficient continuous *** effects of the enzyme order and different linkers on the fusions were investigated by structural analysis and all-atom molecular dynamics *** best fusion YqjM_G_GDH gave 98% conversion of 100 mmol/L 2-methylcyclopentenone with an excellent ee value(>99%)in 3 h,while the mixture of individual enzymes only obtained 68% conversion after more than 8 *** improved substrate conversion of YqjM_G_GDH fusion was probably attributed to the increased flexibility of each fused enzyme and the shortening of the diffusion distance of NADPH regenerated.A one-pot process was designed to purify and immobilize the fusion on the Ni2t-nitrilotriacetic acid functionalized magnetic mesoporous silica *** resulting immobilized biocatalyst not only catalyzed the asymmetric reduction of various α,β-unsaturated ketones(20 mmol/L)continuously with only 50μmol/L NADPt to initiate the whole process,but also retained more than 82%of the initial activity after seven cycles,serving as a good candidate for the industrial applications.

Refractories have an important effect on the cleanliness of molten *** interaction between CeAlO_(3) refractories and Ce treated steel was explored and compared with that between Al_(2)O_(3) refractories and Ce treated *** results show that the Ce content in steel decreases sharply after contact with Al_(2)O_(3) refractories and the pick-up of dissolved Al *** with the test of Al_(2)O_(3) refractories,the loss of Ce content is less in the test of CeAlO_(3) refractory,and the total oxygen content and dissolved Al content in steel increase *** dense and continuous Ce_(2)O_(2)S interface layer is observed at the CeAlO_(3) refractories/steel interface,which can serve as the metallurgical isolation layer and hinder the further interaction between refractories and ***,the higher steel cleanliness and less penetration of refractories by molten steel are obtained in the test of CeAlO_(3) *** CeAlO_(3) refractories show a promising prospect in the production of clean Ce treated steel.

Electrocatalytic performance of CNts/graphene composited rare earth phthalocyanines(M=La,Y,Yb,Sc)

In this study,a class of rare earth composite sandwich phthalocyanines(MPcs,M=La,Y,Yb,Sc) were prepared and compounded with graphene and carbon nanotubes to obtain MPc/Gr and MPc/CNts *** electrocatalytic behaviors of MPc/Gr and MPc/CNts electrodes were further *** results show that the central rare earth metal has a large influence on the electrocatalytic *** the MPcs/Gr samples,ScPc with the smallest ionic radius and molecular size can be more uniformly dispersed in graphene,and the hydrogen precipitation overpotential of ScPc/Gr electrode is514 mV,corresponding to a tafel slope of 148 mV/dec,with better electrocatalytic performance than other rare earth metal *** for the MPc/CNts composites,LaPc,which has the largest ionic radius and molecular size,is more uniformly dispersed on the surface of CNts,so that the LaPc/CNt electrode exhibits the best LSV performance with the smallest corresponding tafel slope(176 mV/dec).the main reason is the different binding modes of MPcs molecules in Gr and *** rare earth phthalocyanine is combined with layered graphene,the smallest size of rare earth phthalocyanine(ScPc)is more easily embedded in the graphene layer,resulting in better homogeneity of the composite,larger effective contact area and better electrocatalytic *** contrast,when rare earth phthalocyanine is bound to carbon nanotubes in a tubular structure,it is mainly bound by attaching to the surface or being entangled by the carbon *** this case,the rare-earth phthalocyanine molecules(LaPc)with larger layer spacing can provide more contact area with CNts,forming a more uniform and effective composite,which eventually provides more active sites and better electrocatalytic performance.

Comparative observation of the effectiveness and safety of remimazolam besylate versus dexmedetomidine in gastrointestinal surgery in obese patients

BACKGROUND Surgery for obese patients carries a higher risk of anesthesia complications compared with surgery for nonobese ***,a safe and effective anesthesia strategy is necessary to improve the medical experience of such patients and ensure their *** to compared the effectiveness and safety of remimazolam besylate versus dexmedetomidine(DEX)in gastrointestinal surgery in obese *** the study cohort included 60 obese patients undergoing gastrointestinal surgery between July 2021 and April 2023,comprising 30 patients who received DEX intervention(control group)and 30 patients who received remimazolam besylate intervention(research group).Heart rate(HR),respiratory rate(RR),mean arterial pressure(MAP),blood oxygen saturation(SpO_(2)),safety(nausea and vomiting,bradycardia,hypotension,and apnea),anesthesia and examination indices[induction time,anesthesia recovery time,and postanesthesia care unit(PACU)discharge time],sedation effect(Ramsay Sedation Scale),and postoperative pain visual analog scale were comparatively analyzed before anesthesia(t0),during anesthesia(t1),and after anesthesia(t2).RESULtS At t1,the research group showed significantly smaller changes in HR,RR,MAP,and SpO_(2) than the control group,with a significantly lower adverse reaction rate and shorter induction,anesthesia recovery,and PACU discharge ***,the intra-and postoperative Ramsay Sedation Scale scores were statistically higher in the research group than in the control *** Remimazolam besylate was significantly more effective than DEX in gastrointestinal surgery in obese patients and had a higher safety profile and value in clinical promotion.

Ultrathin and flexible MXene-contained electromagnetic interference shielding composite paper designed with a protective hydrogel film

MXene-contained paper is a good choice to design ultrathin and flexible electromagnetic interference(EMI)shielding ***,the deficiencies in strength and stability of MXene-contained paper impede its practical ***,a composite paper was proposed to address the problems,in which a filter paper was modified with a three-layer structured surface via a facile layer-by-layer coating ***,the tEMPO-oxidized cellulose nanofibers(tOCN)/cationic starch(CS)/MXene gel layer and tOCN/MXene nacre structure layer ensured the good EMI shielding and mechanical performances of the composite paper,while the uppermost tOCN/CS hydrogel film layer mainly protected *** composite paper achieved an EMI SE of 40.3 dB at a thickness of merely 0.1894 mm(SE/t value of ca.212.8 dB mm^(−1),SSE/t values of ca.13216 dB cm 2 g^(−1))and the total MXene dosage was 20 g m^(−2).Its tensile strength could be up to 11.7 MPa while the original filter paper was 6.4 *** pieces of this composite papers could be easily packed together to attain an EMI SE of nearly 70 ***,the hydrogel film layer efficiently protected the MXene and maintained the EMI shielding performance of the composite paper when immersed in different liquids including water,HCl(1 M)and ethanol,due to the dense and compact structure of hydrogel film *** work provides a practical way to develop ultrathin,flexible and durable EMI shielding materials.

two-dimensional (2D) materials exhibit exceptionally strong nonlinear optical responses, benefiting from their reduced dimensionality, relaxed phase-matching requirements, and enhanced light-matter interaction. With additional degrees of freedom in the modulation of the physical properties by stacking 2D layers together, nonlinear optics of 2D heterostructures becomes increasingly fascinating. In this perspective, we provide a brief overview of recent advances in the field of nonlinear optics of 2D heterostructures, with a particular focus on their remarkable capabilities in characterization and modulation. Given the recent advances and the emergence of novel heterostructures, combined with innovative tuning knobs and advanced nonlinear optical techniques, we anticipate deeper insights into the underlying mechanisms and more associated applications in this rapidly evolving field.

GRK2 mediated degradation of SAV1 initiates hyperplasia of fibroblast-like synoviocytes in rheumatoid arthritis

Hyperplasia and migration of fibroblast-like synoviocytes(FLSs)are the key drivers in the pathogenesis of rheumatoid arthritis(RA)and joint *** Yes-associated protein(YAP),which is a powerful transcription co-activator for proliferative genes,was observed in the nucleus of inflammatory FLSs with unknown upstream *** Gene Expression Omnibus database analysis,it was found that Salvador homolog-1(SAV1),the pivotal negative regulator of the Hippo-YAP pathway,was slightly downregulated in RA ***,SAV1 protein expression is extremely ***,it was revealed that SAV1 is phosphorylated,ubiquitinated,and degraded by interacting with an important serine-threonine kinase,G protein-coupled receptor(GPCR)kinase 2(GRK2),which was predominately upregulated by GPCR activation induced by ligands such as prostaglandin E2(PGE2)in *** process further contributes to the decreased phosphorylation,nuclear translocation,and transcriptional potency of YAP,and leads to aberrant FLSs *** depletion of GRK2 or inhibition of GRK2 by paroxetine rescued SAV1 expression and restored YAP phosphorylation and finally inhibited RA FLSs proliferation and ***,paroxetine treatment effectively reduced the abnormal proliferation of FLSs in a rat model of collagen-induced arthritis which was accompanied by a significant improvement in clinical ***,these results elucidate the significance of GRK2 regulation of Hippo-YAP signaling in FLSs proliferation and migration and the potential application of GRK2 inhibition in the treatment of FLSs-driven joint destruction in RA.

Piperazine-derived ionizable lipids for enhanced mRNA delivery and cancer immunotherapy

Messenger ribonucleic acid(mRNA)-based therapeutics hold great prospects in disease treatment and lipid nanoparticles(LNPs)are the most extensively applied non-viral platform for RNA delivery in *** the clinical success of LNPs as vehicles have been achieved,developing LNPs with enhanced mRNA transmembrane delivery and transfection efficiency in a non-toxic manner is highly desirable and *** this study,we designed a series of new ionizable amino lipids with piperazinederived headgroups and constructed a group of LNPs to promote the transfection activity of mRNA *** them,LNP formulated with lipid 10(L10-LNP)can efficiently package mRNA and perform superior transfection efficiency both in vitro and in vivo,which is mainly attributed to the improved intracellular uptake and effective endosomal *** verified that a single administration of L10-LNP packaging interleukin(IL)-12 mRNA induced tumor shrink and even regression by robust activation of immune effector CD8^(+)t cells and stimulating the generation of IFN-γwithout causing systemic toxicity,which provides a promising platform for clinical cancer immunotherapy.

Magnetic resonance imaging(MRI)plays an important role in precision medicine that is hampered by the lack of contrast agents with high efficiency and the ability to translate diagnostic accuracy into therapeutic ***,we demonstrate a DNA-based MRI probe that overcomes previous single-mode enhancement and provides a mechanism of action for aggregationinduced dual-modal MRI signal enhancement.A facile method is developed to produce aggregated t_(1)/t_(2)dual-modal NaGdF_(4):Dy@PDA-DNA(PDA=polydopamine)MRI *** aggregated,this probe can further amplify MRI signal intensity and exhibit improved geometrical and positional stability in *** performance of the NaGdF_(4):Dy@PDA-DNA MRI probe toward MRI-guided preoperative planning and visualization-guided surgery is verified using an orthotopic tumor-bearing mouse *** result shows that the rapid metabolism of the degraded probe leads to the mitigation of long-term toxic ***,the developed high-performance MRI probe is of great significance for enhancing MRI diagnostic accuracy into precision medical therapeutic interventions.
