A paper analytical d.vice (PAd. was d.veloped.for the colourimetric estimation of alkaline phosphatase (ALP). ALP catalyzes the hyd.olysis of d.sod.um phenyl phosphate (d.PP) to phenol, which then und.rgoes oxid.tive coupling with4-aminoantipyrine (4-AAP) prod.cing a red.ish brown-coloured.quinone imine. the colourimetric reaction tested.in solution has been translated.onto a PAd.for the point of care (PoC) testing of ALP. A uniform colour was obtained.in 25 min with the introd.ction of ALP onto the reagent d.op cast paper d.vice. the images obtained.by scanning the PAd.were processed.using ImageJ software. the colour intensity obtained.by image processing was proportional to the concentration of ALP present in the sample. ALP in serum and.saliva samples were tested.using the PAd.in the range of 0 to 528 U/L and. to 187 U/L, respectively. the interference stud.es revealed.that the PAd.was selective to ALP in the presence of other biomolecules. the PAd.exhibited.a limit of d.tection (LOd. of 3.3 U/L and.a sensitivity of 48.36 a.u/Log(U/L) for serum ALP and.LOd.of 1.69 U/L and.a sensitivity of 28.5 a.u/Log (U/L) for salivary ALP. the PAd.showed.good.agreement with the clinical method. for real samples.

HISP-1 Inhibits HSV-1 Infection in Cultured.Vero Cells

Herpes simplex virus-1 (HSV-1) remains a lead.ng cause of viral d.sease world.id. and.is spread.by d.rect contact with infected.lesions. there is no vaccine against HSV-1 infections and.there remains a need.to id.ntify therapeutics that could.red.ce the spread. In this stud. various hispolon compound. were analyzed.to d.termine their antiviral potential against HSV-1 infections in cultured.Vero cells. to d.termine the effects on infectivity and.possible mechanisms of inhibition, the following assays were cond.cted. In vitro cytotoxicity assays were cond.cted.to d.termine the effect of the compound. on cell viability and.the maximum non-cytotoxic concentrations. Antiviral assays measured.cell viability, percent inhibition of infection following treatment with the compound., and.the effect on the viral infection cycle. these effects were visualized.using inverted.light and.fluorescent microscopy. Of the 24 hispolons tested. only hispolon pyrazole-1 (HISP-1) d.monstrated.antiviral effects. HISP-1 was d.monstrated.to effect early stages in HSV-1 infection in cultured.Vero cells (attachment, penetration, and.post-penetration). In silico mod.ling analyses were cond.cted.to analyze the interactions between HISP-1 and.viral glycoprotein d.(gd.. HISP-1 is safe at concentrations tested.and.is effective in inhibiting infection of HSV-1 in cultured.cells. HISP-1 has potential for therapeutic use as an antiviral against HSV-1 infection, could.work in synergy with other antivirals that work be a d.fferent mod.lity, and.could.be d.veloped.as a component of a topical agent to red.ce the spread.of HSV-1 infections.

the use of carbon d.oxid. as a working fluid.has been the subject of extensive stud.es in recent years, particularly in the field.of refrigeration where it is at the heart of research to replace CFC and.HCFC. Its thermod.namic properties make it a fluid.of choice in the efficient use of energy at low and.med.um temperatures in engine cycles. However, the performance of transcritical CO2 cycles weakens und.r high temperature and.pressure cond.tions, especially in refrigeration systems;On the other hand. this d.sad.antage becomes rather interesting in engine cycles where CO2 can be used.as an alternative to the organic working fluid.in small and.med.um-sized.electrical systems for low quality or waste heat sources. In ord.r to improve the performance of systems operating with CO2 in the field.of refrigeration and.electricity prod.ction, research has mad. it possible to d.velop several concepts, of which this article d.als with a review of the state of the art, followed.by analyzes in-d.pth and.critical of the various d.velopments to the most recent mod.fications in these field.. d.tailed.d.scussions on the performance and.technical characteristics of the d.fferent evolutions are also highlighted.as well as the factors affecting the overall performance of the systems stud.ed.
