Evaluation of the intracellular lipid-lowering effect of polyphenols extract from highland barley in HepG2 cells

Active ingredients from highland barley have received considerable attention as natural products for developing treatments and dietary supplements against *** practical application,the research of food combinations is more significant than a specific food *** study investigated the lipid-lowering effect of highland barley polyphenols via lipase assay in vitro and HepG2 cells induced by oleic acid(OA).Five indexes,triglyceride(tG),total cholesterol(t-CHO),low density lipoprotein-cholesterol(LDL-C),aspartate aminotransferase(ASt),and alanine aminotransferase(ALt),were used to evaluate the lipidlowering effect of highland barley *** also preliminary studied the lipid-lowering mechanism by Realtime fluorescent quantitative polymerase chain reaction(q PCR).the results indicated that highland barley extract contains many components with lipid-lowering effects,such as hyperoside and *** vitro,the lipase assay showed an 18.4%lipase inhibition rate when the additive contents of highland barley extract were 100μg/m *** intracellular lipid-lowering effect of highland barley extract was examined using 0.25 mmol/L OA-induced HepG2 *** results showed that intracellular tG,LDL-C,and t-CHO content decreased by 34.4%,51.2%,and 18.4%,*** and ASt decreased by 51.6%and 20.7%compared with the untreated hyperlipidemic HepG2 cells.q PCR results showed that highland barley polyphenols could up-regulation the expression of lipid metabolism-related genes such as PPARγand Fabp4.

Gastric cancer(GC), the fifth most common cancer globally, remains the leading cause of cancer deaths worldwide. Inflammation-induced tumorigenesis is the predominant process in GC development;therefore, systematic research in this area should improve understanding of the biological mechanisms that initiate GC development and promote cancer hallmarks. Here, we summarize biological knowledge regarding gastric inflammation-induced tumorigenesis, and characterize the multi-omics data and systems biology methods for investigating GC development. Of note, we highlight pioneering studies in multi-omics data and state-of-the-art network-based algorithms used for dissecting the features of gastric inflammation-induced tumorigenesis, and we propose translational applications in early GC warning biomarkers and precise treatment strategies. this review offers integrative insights for GC research, with the goal of paving the way to novel paradigms for GC precision oncology and prevention.

Rock-encased-backfill(RB)structures are common in underground mining,for example in the cut-andfill and stoping *** understand the effects of cyclic excavation and blasting activities on the damage of these RB structures,a series of triaxial stepwise-increasing-amplitude cyclic loading experiments was conducted with cylindrical RB specimens(rock on outside,backfill on inside)with different volume fractions of rock(VF=0.48,0.61,0.73,and 0.84),confining pressures(0,6,9,and 12 MPa),and cyclic loading rates(200,300,400,and 500 N/s).the damage evolution and meso-crack formation during the cyclic tests were analyzed with results from stress-strain hysteresis loops,acoustic emission events,and post-failure X-ray 3D fracture *** results showed significant differences between cyclic and monotonic loadings of RB specimens,particularly with regard to the generation of shear microcracks,the development of stress memory and strain hardening,and the contact forces and associated friction that develops along the rock-backfill *** important finding is that as a function of the number of cycles,the elastic strain increases linearly and the dissipated energy increases ***,compared with monotonic loading,the cyclic strain hardening characteristics are more sensitive to rising confining pressures during the initial compaction *** finding is that compared with monotonic loading,more shear microcracks are generated during every reloading stage,but these microcracks tend to be dispersed and lessen the likelihood of large shear fracture *** transition from elastic to plastic behavior varies depending on the parameters of each test(confinement,volume fraction,and cyclic rate),and an interesting finding was that the transformation to plastic behavior is significantly lower under the conditions of 0.73 rock volume fraction,400 N/s cyclic loading rate,and 9 MPa *** the findings have important practical implications on t

Catalytic transfer hydrogenation(CtH)is a green and efficient pathway for selective hydrogenation of unsaturated aldehydes and ***,managing the abilities of solid catalysts to adsorb substrates and to convert them into desired products is a challenging ***,we report the synthesis of carbon coated LaFe_(0.92)Pd_(0.08)O_(3) composites(LFPO-8@C)for CtH of benzaldehyde(BzH)into benzyl alcohol(BzOH),using isopropanol(IPA)as hydrogen *** coating with carbon improves the ability to adsorb/transfer reactants from solution to active sites,and the doping of Pd2+at Fe3+site strengthens the ability of LaFeO_(3) to convert BzH into BzOH.A balanced point between them(i.e.,abilities to adsorb BzH and to convert BzH into BzOH)is obtained at LFPO-8@C,which exhibits a BzOH formation rate of 3.88 mmol·gcat^(-1)·h^(-1) at 180℃ for 3 h,which is 1.50 and 2.72 times faster than those of LFPO-8 and LaFeO_(3)@C.A reaction mechanism is proposed,in which the acidic sites(e.g.,Fe^(4+),oxygen vacancy)are used for the activation of C=O bond of BzH and O-H bond of IPA,and the basic sites(e.g.,lattice oxygen)for the activation ofα-H(O-H)bond of IPA.

Lactobacillus are considered promising therapeutic methods for nonalcoholic fatty liver disease(NAFLD).the effects of two strains of Ltmosilactobacillus mucosae on NAFLD were investigated in this ***-old male C57BL/6J mice were divided into 4 groups(n=8 per group,Control,Model,FZJtZ26M3,FGSYC17L3).*** FZJtZ26M3 reduced the mice 's body weight,liver weight,and adipose tissue weight after 12 weeks of *** to serum analysis,total cholesterol,triacylglycerol,and low-density lipoprotein cholesterol significantly decreased after *** FZJtZ26M3 *** pathology showed that *** FZJtZ26M3 was effective to ameliorate lipid deposition in NAFLD ***,the expression of the gene related to lipid metabolism in the liver and adipose tissue was analyzed,and the results indicated that *** FZJtZ26M3 could alleviate NAFLD by regulating lipid ***,the results of 16S rRNA gene sequencing revealed a drop in the relative abundance of Ruminococcaceae,which is linked to inflammation,but the relative abundance of a potential probiotic Akkermansia significantly increased after *** FZJtZ26M3 ***,*** FZJtZ26M3 could be a candidate to prevent NAFLD.

Systemic immune monitoring is a crucial clinical tool for disease early diagnosis,prognosis and treatment planning by quantitative analysis of immune ***,conventional immune monitoring using flow cytometry faces huge challenges in large-scale sample testing,especially in mass health screenings,because of time-consuming,technical-sensitive and high-cost ***,the lack of high-performance detection platforms hinders the development of high-throughput immune monitoring *** address this bottleneck,we constructed a generally applicable DNA framework signal amplification platform(DSAP)based on post-systematic evolution of ligands by exponential enrichment and DNA tetrahedral framework-structured probe design to achieve high-sensitive detection for diverse immune cells,including CD4+,CD8+t-lymphocytes,and monocytes(down to 1/100μl).Based on this advanced detection platform,we present a novel high-throughput immune-cell phenotyping system,DSAP,achieving 30-min one-step immune-cell phenotyping without cell washing and subset analysis and showing comparable accuracy with flow cytometry while significantly reducing detection time and *** a proof-of-concept,DSAP demonstrates excellent diagnostic accuracy in immunodeficiency staging for 107 HIV patients(AUC>0.97)within 30 min,which can be applied in HIV infection monitoring and ***,we initially introduced promising DSAP to achieve high-throughput immune monitoring and open robust routes for point-of-care device development.

Self-injection locking has emerged as a crucial technique for coherent optical sources,spanning from narrow linewidth lasers to the generation of localized *** technique involves key components,namely a laser diode and a high-quality cavity that induces narrow-band reflection back into the laser ***,in prior studies,the reflection mainly relied on the random intracavity Rayleigh backscattering,rendering it unpredictable and unsuitable for large-scale production and wide-band *** this work,we present a simple approach to achieve reliable intracavity reflection for self-injection locking to address this challenge by introducing a Sagnac loop into the *** method guarantees robust reflection for every resonance within a wide operational band without compromising the quality factor or adding complexity to the fabrication *** a proof of concept,we showcase the robust generation of narrow linewidth lasers and localized microcombs locked to different resonances within a normal-dispersion ***,the existence and generation of localized patterns in a normal-dispersion cavity with broadband forward–backward field coupling is first proved,as far as we know,both in simulation and in *** research offers a transformative approach to self-injection locking and holds great potential for large-scale production.

Background:Bitter taste receptors(tas2rs)are generally considered to sense various bitter compounds to escape the intake of toxic *** taste receptors have been found to widely express in extraoral tissues and have important physiological functions outside the gustatory system in ***:to investigate the physiological functions of the bitter taste receptor cluster tas2r106/tas2r104/tas2r105/tas2r114 in lingual and extraoral tissues,multiple tas2rs mutant mice and Gnat3 were produced using CRISPR/Cas9 gene-editing technique.A mixture containing Cas9 and sgRNA mRNAs for tas2rs and Gnat3 gene was microinjected into the cytoplasm of the ***,t7EN1 assays and sequencing were used to screen genetic mutation at the target sites in founder *** real-time polymerase chain reaction(qRt-PCR)and immunostaining were used to study the expression level of taste signaling cascade and bitter taste receptor in taste *** to taste substance was also studied using twobottle preference ***:We successfully produced several tas2rs and Gnat3 mutant mice using the CRISPR/Cas9 *** results showed that the expression of GNAt3 and PLCB2 was not altered in tas2rs mutant *** qRt-PCR results revealed the changed expression profile of m tas2rs gene in taste buds of these mutant *** two-bottle preference tests,these mutant mice eliminate responses to cycloheximide due to genetic mutation of *** addition,these mutant mice showed a loss of taste perception to quinine dihydrochloride,denatonium benzoate,and cucurbitacin B(CuB).Gnat3-mediated taste receptor and its signal pathway contribute to CuB ***:these findings implied that these mutant mice would be a valuable means to understand the biological functions of tAS2Rs in extraoral tissues and investigate bitter compound-induced responses mediated by these tAS2Rs in many extraoral tissues.
