Men with obesity often present with low testosterone(t)and sex hormone-binding globulin(SHBG)*** mechanisms for this have been proposed,but as SHBG is secreted by hepatocytes and sex steroids undergo hepatic metabolization,this study investigates whether severity and histological components of nonalcoholic fatty liver disease(NAFLD)are associated with sex steroid levels in obese *** cross-sectional study included 80 obese men(age:46±11 years;body mass index:42.2±5.5 kg m^-2).Serum levels of total t and estradiol(E.)were measured using liquid chromatography coupled with tandem mass spectroscopy(LC/MS-MS)and SHBG and gonadotropins by *** biopsies were evaluated using ***,and Fibrosis *** with steatohepatitis had similar median(1st quartile-3rd quartile)total t levels(7.6[5.0-11.0]nmol l^-1 vs 8.2[7.2-10.9]nmol l^-1;P=0.147),lower calculated free t(cFt)levels(148.9[122.9-188.8]pmol 1-1 s 199.5[157.3-237.6] pmol l^-1;P=0.006),and higher free E.t ratios(10.0[6.4-13.9]×10^-3 vs 7.1[5.7-10.7]×10^-3;P=0.026)compared to men with only nonalcoholic fatty *** the histological components of *** steatosis was independently associated with total t(r=-0.331.P=0.003)and cFt levels(r=-0.255.P=0.025).Obese men *** have even lower cFt levels compared to those without,an association mainly driven by grade of *** this reflects a subgroup of men with a more severe obesity-related phenotype or results from direct relations between hepatic steatosis and sex steroid metabolism needs further investigation.

Winners and losers among tree species in Xishuangbanna: which traits are most important?

Massive human interference in natural ecosystems is resulting in a few ＂winners＂ and many ＂losers＂. However, the drivers of this winner-loser replacement pattern remain poorly understood. the aim of the study reported here was to identify winners among the tree flora of Xishuangbanna and compare their functional traits, specific leaf area （SLA）, wood density （WD）, seed mass （SM） and maximum height （MH） with previously identified losers （i.e., endangered species）. Fifteen native tree species were identified as winners from expert opinion, plot-based surveys of secondary forests and plotless surveys along roads. twelve endangered tree species for which trait information could be obtained were used for comparison. traits were compared with a Wilcoxon rank-sum test. Winners had significantly higher SLA, but lower WD. SM and MH did not differ significantly between groups. When the effects of phylogeny were removed by using phylogenetic generalized least squares, the difference in SLA became marginally insignificant. Principal component analysis resulted in two overlapping groups, showing that the selected traits were insufficient to distinguish winners and losers. Our results suggest that the ＂few winners, many losers＂ paradigm applies to trees in Xishuangbanna, with15 species accounting for most trees in the disturbed habitats sampled.

Arrays of chemical vapor sensors based on graphene field effect transistors functionalized with single-stranded DNA have been demonstrated. Standard photolithographic processing was adapted for use on large-area graphene by including a metal protection layer, which protected the graphene from contamination and enabled fabrication of high quality field-effect transistors （GFEts）. Processed graphene devices had hole mobilities of 1,640 ± 250 cm2.V-1.s-1 and Dirac voltages of 15 ± 10 V under ambient conditions. Atomic force microscopy was used to verify that the graphene surface remained uncontaminated and therefore suitable for controlled chemical functionalization. Single-stranded DNA was chosen as the functionalization layer due to its affinity to a wide range of target molecules and π-π stacking interaction with graphene, which led to minimal degradation of device characteristics. the resulting sensor arrays showed analyte- and DNA sequence-dependent responses down to parts-per-billion concentrations. DNA/GFEt sensors were able to differentiate among chemically similar analytes, including a series of carboxylic acids, and structural isomers of carboxylic acids and pinene. Evidence for the important role of electrostatic chemical gating was provided by the observation of understandable differences in the sensor response to two compounds that differed only by the replacement of a （deprotonating） hydroxyl group by a neutral methyl group. Finally, target analytes were detected without loss of sensitivity in a large background of a chemically similar, volatile compound. these results motivate further development of the DNA/graphene sensor family for use in an electronic olfaction system.

In order to investigate the relationship between restenosis and the morphology detected by coronary angioscopy （CASC） and introvascular ultrasound imaging （IVUS）,17 patients were detected by CASC and IVUS immediately and 3 months after percutaneous transluminai coronary angioscopy （PtCA）.the results showed that the dilation index by IVUS （Dlu） was significantly lower in restenosis patients than in non-restenosis patients （0.42±0.08 versus 0.78±0.16, Pthat the elastic recoil （ER） was higher in restenosis patients than in non-restenosis patients （4.51±1.42mmversus 1.63±1.20 mm, Pthat the elastic recoil rate （ERR） was also higher in restenosis patients than in non-restenosis patients （57.3±8.07% venus 21.80±16.84% Pthat coronary dissection,atheromatous plaque and calcification as well as the coiour of inner coronary artery had no relation with chronic *** conclusion,the elastic recoil is one of the important factors of chronic restcnosis after PtCA.

the objective of the present study was to investigate the effects of genistein and equol on 3β-hydroxysteroid de- hydrogenase （3β-HSD） and 17β-hydroxysteroid dehydrogenase 3 （17β-HSD3） in human and rat testis microsomes. these enzymes （3β-HSD and 17β-HSD3）, along with two others （cytochrome P450 side-chain cleavage enzyme and cytochrome P450 17α-hydroxylase/17-20 lyase）, catalyze the reactions that convert the steroid cholesterol into the sex hormone testosterone. Genistein inhibited 3β-HSD activity （0.2 μmol L^-1 pregnenolone） with half-maximal inhibition or a half-maximal inhibitory concentration （IC50） of 87 ± 15 （human） and 636 ± 155 nmol L^-1 （rat）. Genistein＇s mode of action on 3β-HSD activity was competitive for the substrate pregnenolonrge and noncompetitive for the cofactor NAD＋. there was no difference in genistein＇s potency of 3β-HSD inhibition between intact rat Leydig cells and testis microsomes. In contrast to its potent inhibition of 3β-HSD, genistein had lesser effects on human and rat 17β-HSD3 （0.1 μmol L^-1 androstenedione）, with an IC50 〉 100μmol L^-1. On the other hand, equol only inhibited human 3β-HSD by 42%, and had no effect on 3β-HSD and 17β-HSD3 in rat tissues. these observations imply that the ability of soy isoflavones to regulate androgen biosynthesis in Leydig cells is due in part to action on Leydig cell 3β- HSD activity. Given the increasing intake of soy-based food products and their potential effect on blood androgen levels, these findings are greatly relevant to public health.

the number of available protein sequences in public databases is increasing ***,a sig-nificant percentage of these sequences lack functional annotation,which is essential for the understanding of how bio-logical systems ***,we propose a novel method,Quantitative Annotation of Unknown Structure(QAUSt),to infer protein functions,specifically Gene Ontology(GO)terms and Enzyme Commission(EC)*** uses three sources of information:structure information encoded by global and local structure similarity search,biological network information inferred by protein–protein interaction data,and sequence information extracted from functionally discriminative sequence *** three pieces of information are combined by consensus averaging to make the final *** approach has been tested on 500 protein targets from the Critical Assessment of Functional Annotation(CAFA)benchmark *** results show that our method provides accurate functional annotation and outperforms other prediction methods based on sequence similarity search or *** further demonstrate that a previously unknown function of human tripartite motif-containing 22(tRIM22)protein predicted by QAUSt can be experimentally validated.
