Structure-based Comparative Analysis and Prediction of N-linked Glycosylation Sites in Evolutionarily Distant Eukaryotes

the asparagine-X-serine/threonine （NXS/t） motif, where X is any amino acid except proline, is the consensus motif for N-linked glycosylation. Significant numbers of high-resolution crystal structures of glycosylated proteins allow us to carry out structural analysis of the N-linked glycosylation sites （NGS）. Our analysis shows that there is enough structural information from diverse glycoproteins to allow the development of rules which can be used to predict NGS. A Python-based tool was developed to investigate asparagines implicated in N-glycosylation in five species： Homo sapiens, Mus musculus, Drosophila melanogaster, Arabidopsis thaliana and Saccharo- myees cerevisiae. Our analysis shows that 78 % of all asparagines of NXS/t motif involved in N-gly- cosylation are localized in the loop/turn conformation in the human proteome. Similar distribution was revealed for all the other species examined. Comparative analysis of the occurrence of NXS/t motifs not known to be glycosylated and their reverse sequence （S/tXN） shows a similar distribu- tion across the secondary structural elements, indicating that the NXS/t motif in itself is not bio- logically relevant. Based on our analysis, we have defined rules to determine NGS. Using machine learning methods based on these rules we can predict with 93% accuracy if a particular site will be glycosylated. If structural information is not available the tool uses structural prediction results resulting in 74% accuracy. the tool was used to identify glycosylation sites in 108 human proteins with structures and 2247 proteins without structures that have acquired NXS/t site/s due to non-synonymous variation. the tool, Structure Feature Analysis tool （SFAt）, is freely available to the public at http：//***/tools/sfat.

the good quality CuInGaSe2 (CIGS) thin film solar cells were fabricated on molybdenum metal coated soda lime glass substrate. three-stage co-evaporation method was utilized for the fabrication of high quality p-type CIGS thin film absorber layer. Further, n-type CdS layer, high resistive intrinsic ZnO layer and transparent conducting AlZnO layers were fabricated by CBD method and vacuum sputtering methods. We made three various top metal sandwich grid patterns, i.e. Al, Al/Cu and Cu/Al which were utilized to investigate the metal sandwich layer oriented efficiency enhancement superiority on CuInGaSe2 thin film solar cells. the investigated specific CIGS solar cell device efficiency with respect to various top metal grid sandwich patterns is presented and discussed.

Using 16S rDNA gene sequencing technique, three different species of non-symbiotic bacteria of entomopathogenie nematodes （EPNs）（Steinernema sp. and Heterorhabditis sp.） were isolated and identified from infected insect cadavers （Galleria mellonella larvae） after 48-hour post infections. Sequence similarity analysis revealed that the strains SRK3, SRK4 and SRK5 belong to Ochrobactrum cytisi, Schineria larvae and Ochrobactrum anthropi, respectively. the isolates O. anthropi and S. larvae were found to be associated with Heterorhabditis indica strains BDU-17 and Yer-136, respectively, whereas O. cytisi was associated with Steinernema siamkayai strain BDU-87. Phenotypically, temporal EPN bacteria were fairly related to symbiotic EPN bacteria （Photorhabdus and Xenorhabdus genera）. the strains SRK3 and SRK5 were phylogeographically similar to several non-symbionts and contaminated EPN bacteria isolated in Germany （LMG3311t） and China （X-14）, while the strain SRK4 was identical to the isolates of S. larvae （L1/57, L1/58, L1/68 and L2/11） from Wohlfahrtia magnifica in Hungary. the result was further confirmed by RNA secondary structure and minimum energy calculations of aligned sequences. this study suggested that the non-symbionts of these nematodes are phylogeographicaily diverged in some extent due to phase variation. therefore, these strains are not host-dependent, but environment-specific isolates.

We have successfully attempted to solve the equations of full-MHD model within the framework of Ψ- ωformulation with an objective to evaluate the performance of a new higher order scheme to predict better values of control parameters of the flow. In particular for MHD flows, magnetic field and electrical conductivity are the control parameters. In this work, the results from our efficient high order accurate scheme are compared with the results of second order method and significant discrepancies are noted in separation length, drag coefficient and mean Nusselt number. the governing Navier-Stokes equation is fully nonlinear due to its coupling with Maxwell’s equations. the momentum equation has several highly nonlinear body-force terms due to full-MHD model in cylindrical polar system. Our high accuracy results predict that a relatively lower magnetic field is sufficient to achieve full suppression of boundary layer and this is a favorable result for practical applications. the present computational scheme predicts that a drag-coefficient minimum can be achieved when β=0.4 which is much lower when compared to the value β=1 as given by second order method. For a special value of β=0.65, it is found that the heat transfer rate is independent of electrical conductivity of the fluid. From the numerical values of physical quantities, we establish that the order of accuracy of the computed numerical results is fourth order accurate by using the method of divided differences.

AIM: the objective of the present study was to determine the total phenolic and total flavonoid contents, and to evaluate the antioxidant potential of different leaf extracts of Meyna spinosa Roxb. ex Link, a traditional medicinal plant of India. MEtHODS: Free radical scavenging and antioxidant potential of the methanol, ethyl acetate, and petroleum ether extracts of Meyna spinosa leaves were investigated using several in vitro and ex vivo assays, including the 2, 2-diphenyl-picrylhydrazyl radical scavenging, superoxide anion scavenging, hydroxyl radical scavenging, nitric oxide radical scavenging, hydrogen peroxide scavenging activity, metal chelating assay, and reducing power ability method. total antioxidant activity of the extracts was estimated by the ferric thiocyanate method. Inhibition assay of lipid peroxidation and oxidative hemolysis were also performed to confirm the protective effect of the extracts. total phenolic and total flavonoid contents of the extracts were estimated using standard chemical assay procedures. RESULtS: Methanol extracts showed the highest polyphenolic content and possessed the better antioxidant activity than the other two extracts. total phenolic and total flavonoid contents in the methanol extract were (90.08 ± 0.44) mg gallic acid equivalents/g and (58.50 ± 0.09) mg quercetin equivalents/g, respectively. the IC 50 of the methanol extract in the DPPH , superoxide anion, hydroxyl radical, nitric oxide radical, hydrogen peroxide scavenging activity and metal chelating assays were (16.4 ± 0.41), (35.9 ± 0.19), (24.1 ± 0.33), (23.7 ± 0.09), (126.8 ± 2.92), and (117.2 ± 1.01) μg·mL-1 , respectively. the methanol extract showed potent reducing power ability, total antioxidant activity, and significantly inhibit lipid peroxidation and oxidative hemolysis which was similar to that of standards. High-Velocity Impact Studies on Scaled Leading Edges of Horizontal tail with Smart Composite Layers

Bird strike studies on typical aluminium leading edges of the Horizontal tail (Ht) with and without Glass Fibre Shape Memory Polymer (GF-SMP) layers are carried out. A one-fifth scaled model of Ht is designed and fabricated. the parameters like bird dimension and energy requirements are accordingly scaled to conduct the bird strike tests. two leading-edge components have been prepared, namely one with AL 2024-t3 aluminium alloy and the other specimen of the same dimension and material, additionally having GF-SMP composite layers inside the metallic leading edge, in order to enhance its impact resistance. Bird strike experiments are performed on both the specimens, impacting at the centre of the leading edge in the nose tip region with an impact velocity of 115 m/s. the test component is instrumented with linear post-yield strain gauges on the top side and the PZt sensors on the bottom. Furthermore, the impact scenario is monitored using a high-speed camera at 7000 fps. the bird strike event is simulated by an equation of state model, in which the mass of the bird is idealized using smooth particle hydrodynamics element in PAMCRASH? explicit solver. the strain magnitude and its pattern including time duration are found to be in a good correlation between test and simulation. Key metrics are evaluated to devise an SHM scheme for the load and impact event monitoring using strain gauges and PZt sensors. GF-SMP layers have improved the impact resistance of the aluminium leading edge which is certainly encouraging towards finding a novel solution for the high-velocity impact.
