In vitro and in silico studies on fibrinolytic activity of nattokinase： A clot buster from Bacillus sp.

BACKGROUND： Nattokinase （NK） is a serine protease enzyme of the subtilisin family. It exhibits a strong fibrinolytic activity. The fibrinolytic enzymes from Bacillus sp. have attracted interest as thrombolytic agents because of their efficiency in the fibrinolytic process including plasmin activation. METHODS： In the present study, VIT garden soil was collected and subjected to isolation process in order to screen for the NK production. Screening for NK enzyme was performed by radial caseinolytic assay. The production of NK enzyme was done in two different production medium for comparative studies. The NK enzyme was purified by gel permeation chromatography. The activity of the purified NK was checked by clot lysis and casein digestion assay. To investigate the structural basis of NK and fibrinogen interaction and also to identify the best binding mode, molecular dynamics and docking studies were performed. RESULTS： Based on the morphological and biochemical characterization, the isolate was identified as Bacillus sp. The overall purification fold of NK was about 3 with the specific activity of 664U/mg and 9.9% yield. Homogeneity of the purified enzyme was analyzed and confirmed by the single band obtained in SDS-PAGE. Molecular weight of the purified protease was estimated as 25 kDa. Purified NK enzyme exhibited 97% of effective clot lysis activity. The NK was docked in to the knob region of the fibrinogen at its binding site using Dock server. A total of 26 residues of fibrinogen and 29 residues of NK constitute the interface region. However, 9 residues offibrinogen （THR238, MET264, LYS266, ARG275, THR277, ALA279, ASN308, MET310, and LYS321） and 8 residues ofNK （GLY61, SER63, THR99, PHE189, LEU209, TYR217, ASN218, and MET222） are involved in intact binding. CONCLUSIONS： A significant amount of NK enzyme was obtained from Bacillus sp. Existence of Equilibrium Points in the R3BP with Variable Mass When the Smaller Primary is an Oblate Spheroid

The paper deals with the existence of equilibrium points in the restricted three-body problem when the smaller primary is an oblate spheroid and the infinitesimal body is of variable mass. Following the method of small parameters;the co-ordinates of collinear equilibrium points have been calculated, whereas the co-ordinates of triangular equilibrium points are established by classical method. On studying the surface of zero-velocity curves, it is found that the mass reduction factor has very minor effect on the location of the equilibrium points;whereas the oblateness parameter of the smaller primary has a significant role on the existence of equilibrium points.

Libration Points in the R3BP with a Triaxial Rigid Body as the Smaller Primary and a Variable Mass Infinitesimal Bod

The paper deals with the existence of the coplanar libration points in the restricted three-body problem when the smaller primary is a triaxial rigid body and the infinitesimal body is of variable mass. Following small parameter method, the coordinates of collinear libration points are established whereas the coordinates of triangular libration points are established by classical method. It is found that the mass reduction factor has small effect but triaxiality parameters of the smaller primary have great effects on the coordinates of the libration points.

Synthesis and catalytic activity of SBA-15 supported catalysts for styrene oxidation

Cu(II) and Mn(II) metals embedded on mesoporous SBA-15 were synthesized by co-precipitation *** support and catalysts were characterized by SEM–EDX,TEM,BET,XRD and ICP-AES *** catalytic activity of these catalysts was evaluated for styrene oxidation at various reaction conditions such as styrene to TBHP mole ratio,temperature,catalyst amount by using TBHP as an oxidizing *** reaction products were styrene oxide and benzaldehyde and highest styrene conversion(97.3%) was observed at styrene to TBHP mole ratio of 1:4,temperature at 80 °C and 20 mg of ***,the recyclability of the catalysts was observed and found that they can be recycled three times without major loss in their activity and selectivity.

Current Updates on Salpingectomy for the Prevention of Ovarian Cancer and Its Practice Patterns Worldwide

A paradigm shift of the origin of ovarian cancer to fallopian tube has brought more focus on bilateral salpingectomy as a preventive method for ovarian cancer. Bilateral salpingectomy has shown a dramatic reduction in the risk of ovarian cancer. Bilateral salpingo-oophorectomy has been a long-used practice to prevent ovarian cancer, but it brings surgical menopause and an increased mortality rate to women undergoing such a surgery at the age of <47.5. With the prophylactic bilateral salpingectomy, however, the ovarian function remains unaltered. Recent studies have shown that prophylactic salpingectomy was helpful not only in preventing high-grade serous type ovarian cancer, but also in decreasing adnexal pathologies. With the publication of committee opinion, more practitioners have accepted this proposal, but some are more concerned about its disadvantages. This review illustrates the latest updates on salpingectomy as a preventive method for ovarian cancer, including its advantages and disadvantages, clinicians' opinions, public opinions, so as to find out Obstetricians' and Gynecologists' practice pattern related to opportunistic salpingectomy worldwide.

Fire monitoring in coal mines using wireless underground sensor network and interval type-2 fuzzy logic controller

From the view of underground coal mining safety system, it is extremely important to continuous monitoring of coal mines for the prompt detection of fires or related problems inspite of its uncertainty and imprecise characteristics. Therefore, evaluation and inferring the data perfectly to prevent fire related accidental risk in underground coal mining (UMC) system are very necessary. In the present article, we have proposed a novel type-2 fuzzy logic system (T2FLS) for the prediction of fire intensity and its risk assessment for risk reduction in an underground coal mine. Recently, for the observation of underground coal mines, wireless underground sensor network (WUSN) are being concerned frequently. To implement this technique IT2FLS, main functional components are sensor nodes which are installed in coal mines to accumulate different imprecise environmental data like, temperature, relative humidity, different gas concentrations etc. and these are sent to a base station which is connected to the ground observation system through network. In the present context, a WUSN based fire monitoring system is developed using fuzzy logic approach to enhance the consistency in decision making system to improve the risk chances of fire during coal mining. We have taken Mamdani IT2FLS as fuzzy model on coal mine monitoring data to consider real-time decision making (DM). It is predicted from the simulated results that the recommended system is highly acceptable and amenable in the case of fire hazard safety with compared to the wired and off-line monitoring system for UMC. Legitimacy of the suggested model is prepared using statistical analysis and multiple linear regression analysis.

Re-re-treatment of hepatitis C virus: Eight patients who relapsed twice after direct-acting-antiviral drugs

AIM: To determine risk factors associated with hepatitis C virus(HCV) treatment failure after direct acting antivirals in patients with complex treatment ***: All HCV mono-infected patients who received treatment at our institution were *** was restricted to patients who previously failed treatment with boceprevir(BOC) or telaprevir(TVR) and started simeprevir(SMV) and sofosbuvir(SOF) ± ribavirin(RBV) between December 2013 and June 2014. Patients with human immunodeficiency virus(HIV)/HCV co-infection or patients who received a liver transplant in the past were excluded. Viral loads were recorded while on treatment and after treatment. Data collection continued until December,31 st 2014 when data analysis was initiated. Patients missing virologic outcomes data were not included in the analysis. Analysis of 35 patients who had virologic outcome data available resulted in eight patients who were viral load negative at the end of treatment with SMF/SOF but later relapsed. Data related to patient demographics,HCV infection,and treatment history was collected in order to identify risk factors shared among patients who failed treatment with SMF/***: Eight patients who were treated with the first generation HCV protease inhibitors BOC or TVR in combination with pegylated-interferon(PEG) and RBV who failed this triple therapy were subsequently retreated with an off-label all-oral regimen of SMV and SOF for 12 wk,with RBV in seven cases. Treatment was initiated before the Food and Drug Administration approved a 24-wk SMV/SOF regimen for patients with liver cirrhosis. All eight patients had an end of treatment response,but later relapsed. Eight(100%) patients were male. Mean age was 56(range,49-64). Eight(100%) patients had previously failed PEG/RBV dual therapy at least once in addition to prior failure with triple therapy. Total number of times treated ranged from 3-6(mean 3.8).
