Characterization of Leaf Transcriptome in Banksia hookeriana

Abstract Banksia is a significant element in vegetation of southwestern Australia, a biodiversity hotspot with global significance. In particular, Banksia hookeriana represents a species with significant economic and ecological importance in the region. For better conservation and management, we reported an overview of transcriptome of B. hookeriana using RNA-seq and de novo assembly. We have generated a total of 202.7 million reads （18.91 billion of nucleotides） from four leaf sam- ples in four plants of B. hookeriana, and assembled 59,063 unigenes （average size = 1098 bp） through de novo transcriptomc assembly. Among them, 39,686 unigenes were annotated against the Swiss-Prot, Clusters of Orthologous Groups （COG）, and NCBI non-redundant （NR） protein databases. We showed that there was approximately one single nucleotide polymorphism （SNP） per 5.6-7.1 kb in the transcriptome, and the ratio of transitional to transversional polymorphisms was approximately 1.82. We compared unigenes of B. hookeriana to those of Arabidopsis thaliana and Nelumbo nucifera through sequence homology, Gene Ontology （GO） annotation, and KEGG pathway analyses. The comparative analysis revealed that unigenes of B. hookeriana were closely related to those of N. nucifera. B. hookeriana, N. nucifera, and A. thaliana shared similar GO anno- tations but different distributions in KEGG pathways, indicating that B. hookeriana has adapted to dry-Mediterranean type shrublands via regulating expression of specific genes. In total 1927 potential simple sequence repeat （SSR） markers were discovered, which could be used in the genotype and genetic diversity studies of the Banksia genus. Our results provide valuable sequence resource for further study in Banksia.

Effects of Salvia miltiorrhiza Bunge extract and its single components on monosodium urate-induced pain in vivo and lipopolysaccharide-induced inflammation in vitro

OBJECTIVE:To investigate the possible antinociceptive effects of Salvia(S.)miltiorrhiza Bunge and its single components in monosodium urate(MSU)-induced pain model in mice and lipopolysaccharide(LPS)-induced inflammation model in RAW264.7 ***:Pretreatment of *** Bunge extract(from 1 to 50μg/mL)concentration-dependently attenuated LPS-induced nitric oxide(NO)*** extract of *** Bunge(50 or 100 mg/kg)also caused reversals of decreased threshold for pain in the MSU-treated group as measured by Von-Frey ***,we assessed the antinociceptive and anti-inflammatory properties of the active single components from *** Bunge such as 15,16-dihydrotanshinoneⅠ,tanshinoneⅠ,cryptotanshinone,miltirone,tanshinoneⅡA,and salvianolic acid *** of them showed an anti-inflammatory effect in LPS-induced NO release model and an antinociceptive effect in MSU-treated pain ***:Our results suggest that *** Bunge extract may exert anti-inflammatory effect by reducing LPS-induced NO release and an antinociceptive property in MSU-treated pain ***,tanshinoneⅡA,miltirone,cryptotanshinone,and 15,16-dihydrotanshinoneⅠ,not only appear to be responsible for LPS-induced NO release induced by *** Bunge,but also in the production of *** Bunge extract-induced antinociception in MSU-treated pain ***:Therefore,the analgesic and anti-inflammatory property of *** Bunge indicate it as a therapeutic potential candidate for the treatment of pain and inflammation.

A study on effects of ceramic foam filter on flow aspect through water modeling experiment

Casting defects that are closely related to entrapped air bubbles and metallic oxides occur very frequently in the sand mold casting process. Many researchers have shown that these defects can be reduced by adopting an appropriate gating system design. However, it is difficult for field engineers to identify a specific gating system that is more appropriate for their products. In this study, we tried to draw a comparison between two gating system designs with and without a ceramic foam filter. The ceramic foam filter was added to the horizontal runner just after the down sprue to prevent air bubble generation and reduce turbulence without a change of gating system design. The water modeling experiment was conducted with four different amounts of the initial volumes of water in the reservoir to verify the effects of initial pouring velocity. The results of the experiment applying the filter showed remarkably changed flow characteristics. The use of the filter was found to convert the flow pattern of water in the desired direction. The ceramic foam filter performed well to reduce flow velocity and stabilize the water *** flow pattern without a filter can be improved significantly even with the the use of just a 10 PPI irregular filter. Although the study confirmed that use of the filter may change the flow characteristics, it needs to be noted that the use of the ceramic filter alone cannot solve all the problems caused by a poorly designed gating system.

The peptide fraction of Bothrops jararaca snake venom induces toxicological effects on the male reproductive system after local envenomation in mice

Bothrops envenomation is complex and provokes prominent local tissue damage and systemic disturbances,but little is known about their effects on the male reproductive *** intratesticular injection,the bioactive peptide fraction(Bj-PF)obtained from Bothrops jararaca snake venom changes the structure of different stages of the seminiferous epithelium cycle in adult *** the first time,we investigated whether local envenomation of Bj-PF induces toxicological effects on the male reproductive system,particularly on the seminiferous epithelium and Sertoli *** adult mice were treated with 0.24 ***^(-1) by intramuscular(i.m.)injection for 24 *** testes samples were collected for morphological and morphometric *** toxicological effects of Bj-PF were also analyzed on mitochondrial metabolism and nitrite(NO2)production in 15P-1 Sertoli cell ***-PF changed the structure and function of the seminiferous epithelium,particularly the disruption of the epithelium and the presence of degenerated germ cells in the adluminal compartment,but there were no alterations in the basal ***-PF increased the thickness of the seminiferous epithelium and decreased the lumen diameter of the *** histological assessment of the degree of tubule degeneration revealed that Bj-PF also increased the number of hypospermatogenic tubules compared to ***-PF reduced NO2 levels in 15P-1 Sertoli cells without changing the mitochondrial ***,the fact that Bj-PF alters the structure and function of the seminiferous epithelium suggests that bioactive peptides found in *** snake venom can have toxicological effects on the reproductive systems of affected male mice,providing new insight into the biological characteristics of snake venom and therapeutic strategies for envenomation inflammation.

Compensating the impurities on the Cu surface by MOFs for enhanced hydrocarbon production in the electrochemical reduction of carbon dioxide

Copper(Cu)provides a cost-effective means of producing value-added fuels through the electrochemical reduction of carbon dioxide(CO_(2)RR).However,we observed the production of hydrocarbons via CO_(2)RR on commercial Cu films is less efficient because of the surface impurities,i.e.,*** monoxide(CO),a reaction intermediate of CO_(2)RR to hydrocarbons,binds strongly to the Fe sites and interrupts the production of hydrocarbons,resulting in an active hydrogen evolution reaction(HER).Herein,we report a method of blocking the effect of Fe impurities on the Cu surface through the preferential growth of nano-sized metal-organic frameworks(MOFs)on Fe *** zirconium(Zr)-based MOFs(UiO-66)forms a compensating layer on Cu film via the terephthalic acid(TPA)-Fe coordination bond,the Ui O-66 coated Cu film(UiO-66@Cu)presents significantly improved hydrocarbon Faradaic efficiency(FE)of 37.59%compared to 14.68%FE on commercial Cu film(99.9%purity)by suppressing *** to X-ray photoelectron spectroscopy(XPS)analysis,the UiO-66 ligand binds to entire metallic Fe site on the Cu surface,while metallic Cu is ***,UiO-66@Cu provides active sites of Cu for CO_(2)RR and leads to highly efficient and selective production of hydrocarbons.

Mobility of Arsenic and Heavy Metals in a Sandy-Loam Textured and Carbonated Soil

The continued effect of the pyrite-tailing oxidation on the mobility of arsenic, lead, zinc, cadmium, and copper was studied in a carbonated soil under natural conditions, with the experimental plot preserved with a layer of tailing covering the soil during three years. The experimental area is located in Southern Spain and was affected by a pyrite-mine spill. The climate in the area is typically Mediterranean, which determines the rate of soil alteration and element mobility. The intense alteration processes that occurred in the soil during three years caused important changes in its morphology and a strong degradation of the main soil properties. In this period, lead concentrated in the first 5 mm of the soil, with concentrations higher than 1500 mg kg?1, mainly associated to the neoformation of plumbojarosite. Arsenic was partially leached from the first 5 mm and mainly concentrated between 5–10 mm in the soil, with maximum values of 1239 mg kg-1; the retention of arsenates was related to the neoformation of iron hydroxysulfates (jarosite, schwertmannite) and oxyhydroxides (goethite, ferrihydrite), both with a variable degree of crystallinity. The mobility of Zn, Cd, and Cu was highly affected by pH, producing a stronger leaching in depth; their retention was related to the forms of precipitated aluminium and, in the case of Cu, also to the neoformation of hydroxysulfate.

An experimental study on tsunami inundation over complex coastal topography

In recent studies, the effects of complex costal topography on tsunami run-up has sparked heated discussion. This study mainly aims at investigating the effects of complex costal topography on the tsunami inundation distance and the effectiveness of sand dunes in dissipating tsunami wave energy. The experiments were carried out in a wave flume to investigate the potential reduction effects of wave run-up by non erodible sand dune like features. The results show that increasing dunes spacing could not significantly affect inundation distance. However, if the height of sand dunes is of the same order of magnitude as the incoming tsunami wave and the gaps between the dunes are large enough, successful tsunami mitigation could also be possible.

Roger Ames＇s Confucian Role Ethics.＂ A Vocabulary, is an ambitious and significant exposition of Confucianism. Despite my disagreements with some of Ames＇s interpretation of Confucius, I have both enjoyed this book and learned a lot from it. Due to the limited scope of this review, I restrict it to an evaluation of Ames＇s exposition of the highest Confucian virtue, ren 4Y-. By Ames＇s own account, this book does not offer an alternative ethical theory, but a Confucian account of human flourishing, which begins with the roles and relations in the family. These roles and relations provide the basis of morality for Ames, as they not only constitute us but also enable us to become virtuous, respond appropriately in any situation, and enhance our relationships. In Ames＇s own words, we are ＂interrelated persons living a multiplicity of roles that constitute who we are, and ... allow us to pursue a unique distinctiveness and virtuosity in our conduct＂（122） and ＂Confucian role ethics takes the substance of morality to be nothing more or less than positive growth in the constitutive relations of any particular situation＂（258）.

Diagnostic criteria for autoimmune hepatitis in children: A challenge for pediatric hepatologists

Autoimmune hepatitis (AIH) is a progressive inflammatory liver disorder that is rare in children and adolescents. AIH has a broad clinical spectrum and a quick response to treatment with corticosteroids and immunosuppressive medication. The available diagnosis criteria have limitations and should be evaluated in pediatric populations. Recently, some studies reported that the 2008 simplified diagnostic criteria for AIH could be used in children with high sensibility and specificity. In addition, the authors reported that globulin and immunoglobulin G levels can be used interchangeably for diagnostic purposes. They also demonstrated that the 2008 simplified criteria fail in identifying patients with fulminant hepatic failure. Here, we discuss the limitations of the use of these criteria in pediatric patients and the requirement of more studies to improve the diagnosis of AIH in children.
