Introduction: Malaria remains a public health priority in Senegal, particularly in Tambacounda, where it is one of the main causes of child mortality. The objective of this work was to evaluate the care of children under 10 years of age hospitalized at the Tambacounda Health Center and the factors associated with recovery. Methods: An analytical, retrospective, and descriptive cross-sectional study with exhaustive recruitment of children 0 to 120 months hospitalized at the Tambacounda reference health center for severe malaria (according to WHO criteria) between 1 January 2018 and 31 December 2021 was performed. Data collection was done through a questionnaire. Records, hospitalization records, and treatment records were the sources of collection. Data entry and analysis were performed on Epi Info 7.2 and R. Results: A total of 481 children hospitalized with severe malaria were recruited. The highest number of cases was recorded in 2018 (33.05%). In the four years of the study, peaks were always observed between October and November and the highest peak in November 2020 with 95 cases. The mean age was 65.64 months with a standard deviation of 29.28 months and a predominance of male (53.43%). The majority of people were admitted from the outpatient clinic (57.79%) and the rest (42.21%) on the recommendation of a peripheral health post. All hospitalized patients had a positive RDT and/or a positive thick drop. However, the sharp decline at admission or during hospitalization was positive in 93.80% of patients in our series, negative in 5.20% and not achieved in 1.00%. Seizures and severe anemia topped the list of signs of severity with 45.94% and 8.11%, respectively. In terms of evolution, for all hospitalized patients, there were 81.29% recovery, 10.19% referral to the Tambacounda regional hospital center for hospitalization, 4.99% death, 0.83% discharge and 2.70% unknown evolution. International disaster databases and catalogs provide a baseline for researchers,governments,communities,and organizations to understand the risk of a particular place,analyze broader trends in disaster risk,and justify investments in *** because Singapore is routinely identified as one of the safest countries in the world,Singapore’s past disasters have not been studied extensively with few events captured in major global databases such as *** this article,we fill the disaster data gap for postwar Singapore(1950–2020)using specified metrics through an archival search,review of literature,and analysis of secondary *** present four key lessons from cataloging these ***,we expand Singapore’s disaster catalog to 39 events in this time period and quantify the extent of this data ***,we identify the mitigating actions that have followed past events that contribute to Singapore’s present-day ***,we discuss how these past events uncover continuities among vulnerability bearers in ***,we identify limitations of a disaster catalog when considering future *** expanding the disaster catalog,this case study of Singapore supports the need for comprehensive understanding of past disasters in order to examine current and future disaster resilience.

Human dental pulp stem cells differentiate into neural precursors but not into mature functional neurons

Large numbers of neuronal cells are needed for regenerative medicine to treat patients suffering from central nervous system diseases and deficits such as Parkinson’s disease and spinal cord injury. one suggestion has been the utilization of human dental pulp stem cells (hDPSCs) for production of neuronal cells which would offer a patient-specific cell source for these treatments. Neuronal differentiation of hDPSCs has been described previously. Here, we tested the differentiation of DPSCs into neuronal cells with previously reported protocol and characterized the cells according to their morphology, gene and protein expressions and most importantly according to their spontaneous electrical functionality with microelectrode array platform (MEA). Our results showed that even though hDPSC-derived neural progenitor stage cells could be produced, these cells did not mature further into functional neuronal cells. Thus, utilization of DPSCs as a cell source for producing grafts to treat neurological deficits requires more efforts before being optimal.

High level HIV-1 DNA concentrations in brain tissues differentiate patients with post-HAART AIDS dementia complex or cardiovascular disease from those with AIDS

Highly active antiretroviral treatment(HAART) has had a significant impact on survival of individuals with acquired immunodeficiency syndrome(AIDS);however,with the longer life-span of patients with AIDS,there is increasing prevalence of AIDS dementia complex(ADC) and other non-AIDS-defining illness,and cardiovascular diseases(CVD) are also *** influence of these varied disease processes on HIV-1 DNA concentration in brain tissues has not been thoroughly assessed in the post-HAART *** purpose of the current study is to clarify the impacts of ADC and other complications of HIV disease on the viral load in the brains in AIDS patients with *** examined autopsy specimens from the brains of thirteen patients who died from complications of AIDS with quantitative polymerase chain reaction(QPCR).All but one patient had received HAART prior to death since *** patients died with severe CVD,multiple cerebrovascular atherosclerosis(CVA) throughout the brain and five patients died with *** patients had no ADC/CVA.A QPCR was used to measure the presence of HIV-1 DNA in six brain tissues(meninges,frontal grey matter,frontal white matter,temporal subcortex,cerebellum and basal ganglia).In the post-HARRT era,for non-ADC/CVA patients,HIV-1 DNA concentration in brain tissues was statistically higher than that in patients with *** a new finding,two patients who suffered from severe CVD,especially CVA,also had high concentrations of HIV-1 in brain compartments not showing ADC related *** our knowledge,this is the first report of a relationship between the CVA and HIV-1 viral burden in *** current observations suggest that HAART-resistant HIV reservoirs may survive within ADC lesions of the brain as well as the macrophage rich atherosclerosis,which needs to be confirmed by more AIDS cases with CVA.

Transcription is a highly regulated cellular process in which dysfunction leads to disease. one level of regulation is chromatin structure which protects promoters from transcription factor binding. To circumvent this blockade, histone chaperones aid in displacement of nucleosomes. In particular, the histone chaperone complex HUCA, consisting of Hira, Ubn1, Cabin1, and ASF1a, replaces histone variant H3.1 with H3.3 in front of actively transcribing RNA Polymerase II (RNAPII). The 26S proteasome is a major degrader of proteins within the cell and plays both proteolytic and non- proteolytic roles in transcriptional regulation. one major role is the degradation of irreversibly arrested RNAPII. Several interactions between HUCA, the 26S proteasome, and RNAPII have been characterized individually;we now present observations from our lab and others which directly associate elongating RNAPII with the degradation machinery through observations of involvement with the HUCA complex. Our short report presents these ideas and discusses their importance in transcriptional regulation as well as implications in disease manifestation.

Histone deacetylase 1(HDAC1)is an important epigenetic controller involvedin transcriptional regulation throughmodification of chromatin *** and epigenetic changes and deregulation of signal transduction pathways have been implicated in the development of breast *** of estrogen receptor a(ERa)expression is one of the mechanisms behind the acquisition of endocrine *** and increased hormone and growth factor receptor signaling in breast cancer cells contribute to resistance to endocrine *** HDACs and the PI3K/mTOR signaling pathway are becoming promising targets in breast cancer,reversing also acquired hormone *** we show how mitogens,activating the PI3K/mTOR pathway,trigger the phosphorylation of HDAC1 in breast cancer cells,which is completely dependent on the activity of the p70 S6 kinase(S6K1).Our findings show that S6K1,overexpressed in many breast cancers,controls HDAC1-dependent transcriptional regulation of ERa levels upon mitogenic stimuli,controlling HDAC1 recruitment to the ERa ***,cell treatment with both mTOR and HDACs inhibitors shows an additive effect in inhibiting breast cancer *** confirms the novel cross-talk between the HDAC1 and PI3K pathways with clinical implications towards the treatment of this malignant disease.

Two NGO-Run Youth-Centers in Multicultural, Socially Deprived Suburbs in Sweden—Who Are the Participants?

Objective: Leisure-time is an important part of young people’s lives. one way to reduce social differences in health is to improve adolescents’ living conditions, for example by enhancing the quality of after-school activities. Multicultural, socially deprived suburbs have less youth participation in organized leisure-time activities. This study explores who the participants are at two NGO-run youth-centers in multicultural, socially deprived suburbs in Sweden and whether socio-demographic, health-related, and leisure-time factors affect the targeted participation. Methods: The study can be seen as an explanatory mixed-methods study where qualitative data help explain initial quantitative results. The included data are a survey with youth (n = 207), seven individual interviews with staff, and six focus-groups interviews with young people at two youth-centers in two different cities. Results and Conclusions: The participants in the youth-centers are Swedish born youths having foreign-born parents who live with both parents, often in crowded apartments with many siblings. Moreover they feel healthy, enjoy school and have good contact with their parents. It seems that strategies for recruiting youths to youth-centers have a large impact on who participates. one way to succeed in having a more equal gender and ethnicity distribution is to offer youth activities that are a natural step forward from children’s activities. The youth-centers’ proximity is also of importance for participation, in these types of neighborhoods.

Men’s Experience of Difficulties during First Year Following Myocardial Infarction—Not Only Fatigue

Recovery from a myocardial infarction (MI) can be a very difficult process and some patients are also stricken by fatigue. The aim of the study was to describe the difficulties experienced by men with fatigue during the first year following their MI. The study was conducted using qualitative content analysis of semi-structured interviews with nine men in working age (mean age: 55 years) with verified fatigue (≥12 of 20 on the Multidimensional Fatigue Scale with a mean of 17) one year after their MI. During the first year after the MI the men suffering from fatigue experienced various difficulties in terms of both themselves and the care received. The analysis generated three themes with difficulties described by the interviewed men. 1) Going through involuntary change: not recognizing their body and the inability to recognize themselves. 2) Lacking individualized care: To be denied satisfactory health care treatment and to be denied respectful treatment. 3) Not being seen in the light of the social context: The MI affects the whole family and caregivers do not pay attention to the family. In order to make both the patient and relatives feel secure and cared for, it is essential that caregivers be aware of the importance of providing individualized care over time with particular attention for the patient’s social context.
