Background: Cirrhotic patients have higher rates of hypercoagulable disorders. We hypothesized that orthotopic liver transplant (OlT) recipients with pre-operative portal vein thrombosis (PVT) have more post-operative thrombotic events than those without PVT. Aims: To compare rates of post-op thrombotic events and outcomes between those with and without pre-op PVT. Methods: All OlT recipients between 1/02-4/09 were retrospectively reviewed. Outcome measures included survival, deep venous thrombosis, pulmonary embolism, hepatic artery thrombosis, and recurrent PVT. Minimum follow up was 6 months. Results: In 363 OlTs performed, mean recipient age was 53.1 yrs (±9.2);268 patients were male. Mean MElD at transplant was 22.1 (±6.2). The prevalence of pre-op PVT was 11.2% (41/350). There was no difference in the % of post-op thrombotic events between those with and without PVT (p = 0.77). MElD, recipient and donor age, and gender were similar in both groups. Mean survival in those with pre-op PVT was 85.2 months vs. 78.7 in those without PVT (p = 0.19). Conclusions: The rate of post-op thrombotic events was equivalent in OlT recipients with and without pre-op PVT. The presence of PVT did not adversely impact patient survival and should not be a contraindication to OlT.

Functional Inferences of Environmental Coccolithovirus Biodiversity

The cosmopolitan calcifying alga Emiliania huxleyi is one of the most abundant bloom forming coccolithophore species in the oceans and plays an important role in global biogeochemical cycling. Coccolithoviruses are a major cause of coccolithophore bloom termination and have been studied in laboratory, mesocosm and open ocean studies. However, little is known about the dynamic interactions between the host and its viruses, and less is known about the natural diversity and role of functionally important genes within natural coccolithovirus communities. Here, we investigate the temporal and spatial distribution of coccolithoviruses by the use of molecular fingerprinting techniques PCR, DGGE and genomie sequencing. The natural biodiversity of the virus genes encoding the major capsid protein （MCP） and serine palmitoyltransferase （SPT） were analysed in samples obtained from the Atlantic Meridional Transect （AMT）, the North Sea and the l4 site in the Westem Channel Observatory. We discovered nine new coccolithovirus genotypes across the AMT and l4 site, with the majority of MCP sequences observed at the deep chlorophyll maximum layer of the sampled sites on the transect. We also found four new SPT gene variations in the North Sea and at l4. Their translated fragments and the full protein sequence of SPT from laboratory strains EhV-86 and EhV-99B 1 were modelled and revealed that the theoretical fold differs among strains. Variation identified in the structural distance between the two domains of the SPT protein may have an impact on the catalytic capabilities of its active site. In summary, the combined use of ＇standard＇ markers （i.e. MCP）, in combination with metabolically relevant markers （i.e. SPT） are useful in the study of the phylogeny and functional biodiversity of coccolithoviruses, and can provide an interesting intracellular insight into the evolution of these viruses and their ability to infect and replicate within their algal hosts.

The United States Food and Drug Administration recently warned that the direct acting antiviral(DAA) combination hepatitis C virus(HCV) treatment of Paritaprevir, Ombitasvir, Dasabuvir, Ritonavir, and Ribavirin(PODr + R) can cause severe liver injury in patients with advanced liver disease. Drug induced liver injury was observed in a small number of patients with decompensated cirrhosis treated with other DAAs, but has not been reported in patients with compensated cirrhosis. We report a case of a 74-year-old woman with chronic HCV and Child-Pugh class A cirrhosis(compensated cirrhosis) treated with PODr + R. The patient presented on day 14 of PODr + R therapy with jaundice and new-onset ascites. Her total bilirubin level increased to 23 mg/dl and international normalized ratio rose to 1.65, while aminotransferase levels remained relatively stable. Hepatitis C treatment was discontinued on day 24 and she gradually recovered. Follow-up testing showed that she achieved a sustained virologic response. In conclusion, hepatic decompensation developed within two weeks of starting treatment withPODr + R in a patient with Child-Pugh class A cirrhosis and was characterized by jaundice and ascites with stable aminotransferase levels. Careful monitoring is warranted in patients with HCV-related cirrhosis treated with PODr + R.

AIM: Functional characterization of ATP1A2 mutations that are related to familial or sporadic hemiplegic migraine(FHM2, SHM). METHODS: cRNA of human Na+/K+-ATPase α2- and β1-subunits were injected in Xenopus laevis oocytes. FHM2 or SHM mutations of residues located in putative α/β interaction sites or in the α2-subunit's C-terminal region were investigated. Mutants were analyzed by the twoelectrode voltage-clamp(TEVC) technique on Xenopus oocytes. Stationary K+-induced Na+/K+ pump currents were measured, and the voltage dependence of apparent K+ affinity was investigated. Transient currents were recorded as ouabain-sensitive currents in Na+ buffers to analyze kinetics and voltage-dependent presteady state charge translocations. The expression of constructs was verified by preparation of plasma membrane and total membrane fractions of cRNA-injected oocytes. RESUlTS: Compared to the wild-type enzyme, the mutants G900R and E902K showed no significant dif-ferences in the voltage dependence of K+-induced currents, and analysis of the transient currents indicated that the extracellular Na+ affinity was not affected. Mutant G855R showed no pump activity detectable by TEVC. Also for l994del and Y1009X, pump currents could not be recorded. Analysis of the plasma and total membrane fractions showed that the expressed proteins were not or only minimally targeted to the plasma membrane. Whereas the mutation K1003E had no impact on K+ interaction, D999H affected the voltage dependence of K+-induced currents. Furthermore, kinetics of the transient currents was altered compared to the wild-type enzyme, and the apparent affinity for extracellular Na+ was reduced. CONClUSION: The investigated FHM2/SHM mutations influence protein function differently depending on the structural impact of the mutated residue.

The significance and ability for receptor targeted nanoliposomes(tNls)to bind to their molecular targets in solid tumors in vivo has been questioned,particularly as the efficiency of their tumor accumulation and selectivity is not always predictive of their efficacy or molecular *** study presents,for the first time,in situ near-infrared(NIR)molecular imaging-based quantitation of the in vivo specificity of tNls for their target receptors,as opposed to tumor selectivity,which includes influences of enhanced tumor permeability and *** show that neither tumor delivery nor selectivity(tumor-to-normal ratio)of cetuximab and IRDye conjugated tNls correlate with epidermal growth factor receptor(EGFR)expression in U251,U87,and 9l tumors,and in fact underrepresent their imaging-derived molecular specificity by up to 94.2%.Conversely,their in vivo specificity,which we quantify as the concentration of tNl-reported tumor EGFR provided by NIR molecular imaging,correlates positively with EGFR expression levels in vitro and ex vivo(Pearson’s r=0.92 and 0.96,respectively).This study provides a unique opportunity to address the problematic disconnect between tNl synthesis and in vivo *** findings encourage their continued adoption as platforms for precision medicine,and facilitates intelligent synthesis and patient customization in order to improve safety profiles and therapeutic outcomes.

Does Selection for Seedling Tiller Number in Perennial Biomass Feedstocks Translate to Yield and Quality Improvements in Mature Swards?

Breeding for seedling traits in herbaceous perennial biomass feedstocks that translate into increased biomass yield or quality in established swards could accelerate the development of perennial grass cultivars for bioenergy or forage. In previously reported research, breeding for single large tillers (ST) or multiple tillers (MT, ≥3) six weeks after planting for two generations in big bluestem (Andropogon gerardii Vitman) and switchgrass (Panicum virgatum l.) base populations produced ST and MT populations that differed significantly for seedling and mature plant traits including biomass yield in spaced planted nurseries. Our objective was to evaluate these ST and MT populations in sward trials to determine the effect of these genetic changes on biomass yield and quality when the plants were grown in competitive sward conditions. Big bluestem monocultures of the base, ST, and MT populations were evaluated at three locations in Nebraska in 2001 and 2002 as randomized complete block experiments with four replicates. Switchgrass monocultures of the base, ST, and MT populations were evaluated in 2003, 2004, and 2005 near Mead, NE as a randomized complete block with six replicates. In both big bluestem and switchgrass, the ST and MT populations did not consistently differ from the base population or each other for biomass yield or forage quality. These results demonstrate the importance of evaluating perennial grasses in sward trials and not relying solely on greenhouse-grown plants or space-planted nurseries to develop selection criteria and make selection decisions.

This study investigated the speciation of halogen-specific total organic halogen and disinfection byproducts(DBPs) upon chlorination of natural organic matter(NOM) in the presence of iopamidol and bromide(Br^-).Experiments were conducted with low bromide source waters with different NOM characteristics from Northeast Ohio,USA and varied spiked levels of bromide(2-30 μmol/l) and iopamidol(1-5 μmol/l).Iopamidol was found to be a direct precursor to trihalomethane(THM) and haloacetic acid formation,and in the presence of Br^-favored brominated *** concentration and speciation of DBPs formed were impacted by iopamidol and bromide concentrations,as well as the presence of *** iopamidol increased the concentration of iodinated DBPs(iodo-DBPs) and THMs ***,as Br^-concentrations increased,the concentrations of nonbrominated iodo-and chloro-DBPs decreased while brominated-DBPs *** of the concentration of either iopamidol or bromide,bromochloroiodomethane(CHBrClI) was the most predominant iodo-DBP formed except at the lowest bromide concentration *** relevant concentrations of iopamidol(1 μmol/l) and bromide(2 μmol/l),significant quantities of highly toxic iodinated and brominated DBPs were ***,the rapid oxidation and incorporation of bromide appear to inhibit iodoDBP formation under conditions relevant to drinking water treatment.
