Objective: To study the anti-diabetic activity of marine macroalgae extracts (n = 31), purification and characterization of sulphated galactopyran (SGP) from Gracilaria opuntia (FM4) in diabetic rats. Methods: The animals were separated into groups and STZ (55 mg/kg body weight) was used to induce diabetics. Glucose, HbA1c, insulin, c-peptide levels and in vivo antioxidant levels were estimated and histopathological studies were done in STZ-induced diabetic and marine macroalgae treated rats. Results: Based on glucose and HbA1c levels and in vivo antioxidant levels, among the 31 marine macroalgae extracts, FM4 has showed high anti-diabetic activity. Hence, FM4 was purified and characterized by 1H-NMR spectra and FT-IR as sulphated galactopyran. During the survival analysis, SGP at dose of 100 mg/kg showed significant (P c-peptide and insulin levels. The histopathological modulations of SGP were observed in diabetic rat tissues such as liver, kidney and brain. Hence obtained results reveal that SGP treated diabetic rats has significant changes in c-peptide and insulin levels which regulates the blood glucose levels and recovered the histopathological changes. conclusions: Marine macroalgae have significant anti-diabetic activity. Hence, they could be used as nutraceutical supplement or natural green remedy against diabetes mellitus.

在实验室条件下,试验了某种巨型水生植物及藻类,如水葫芦、铜绿微胞藻、镰刀形栅列藻、小球藻及紫茉莉属衣藻等对污染物的去除效率,评价它们在废水处理中的潜在作用。试验废水为 Varanasi 市的城市污水与1200家小型工业的混合废水。研究分三个阶段:先用风信子水培,接着进行藻类处理,最后用风信子二次水培。在第一阶段,10枝风信子植物在污水槽中生长15天;在第二阶段,藻类在污水槽中培养5天;第三阶段,再次用风信子水培9天。经三个阶段的处理,污染物量大幅度减少,溶解氧增高至70％,各种污染物去除率分别为:BOD96.9％,悬浮物78.1％,总碱度74.6％,PO_4-P 89.2％,NO_3-N 81.7％,酸度73.3％,NH_4-N 95.1％,cOD 77.9％,硬度68.6％,大肠杆菌99.2％。

The ability of seed extracts of Trigonella foenum-graecum (T. foenum-graecum) and cuminum cyminum (c. cyminum) to act as natural coagulants was tested using natural turbid water. Seed extracts were prepared using distilled water and Nacl (0.5 M and 1.0 M) solution. Only 1.0 M Nacl extract of T. foenum-graecum had coagulation capability and did not depend on pH values. Further it showed that natural coagulant obtained from T. foenum-graecum is temperature (upto 100?c) and pH stable (pH 4.0 - 10.0). Extract of c. cyminum had very minimal (16 ± 2) coagulation property. The seed extract of T. foenum-graecum showed about 80% coagulation properties, where as the best known natural coagulants such as Strychnos potatorum and Moringa oleifera, and chemical coagulant such as Al2(SO4)3 showed around 90%, 65% and 95% respectively, which are used as standards for the present study. When compared with pond water, T. foenum-graecum extract treated water shows decrease in alkalinity, turbidity, KMnO4 demand and total coli-form. This study reveals that seed extract of T. foenum-graecum can be used as natural water coagulant.

AIM:To investigate the chemopreventive iffects of 1α,25-dihydroxyvitamin D3 in diethylnitrosamine induced,phenobarbital promoted rat ***:The rats were randomly divided into 6 different groups(A-F).Group A,c and E rats received a single intraperitoneal (i.p) injection of diethylnitrosamine (DEN) at a dose of 200 mg/kg body mass in 9 g/L Nacl solution at 4 wk of age ,while group B served as normal vehicle control received normal saline *** a brief recovery of 2 WK, all the DEN treated rats were given phenobarbital (PB) at 0.5g/L daily in the basal diet till WK *** c was started 4 wk prior to DEN injection and continued thereafter till wk 20 at a dose (os) twice a *** E received the treatment of 1α,25-(OH)2D3 at the same dose mentioned as above for 15 *** rats in group D and F received 1α,25-(OH)2D3 alone as in group c without DEN ***:The comet assay showed statistically higher mean values for length to width ratios (L:W) of DNA mass and tailed cells (pctively) in DEN treated rats as compared to their normal *** supplementation of 1α,25-dihydroxyvitaminD3 showed a significant (pcrease in L:W ratio of DNA mass tailed ***,1α,25-(OH)2D3 supplementations elevated the super oxide dismutase (SOD) activety,hepatic malondialdehyde(MDA) levle,reduced glutathione (GSH) and glutathione S-transferase (GST) activety (pctively).As an endpoint marker histological changes were observed to establish the chemopreventive effects of 1α,***:Supplementations of 1α,25-(OH)2D3 has a marked protection against hepatic nodelogenesis,antioxidant enzymes and DNA damages in DEN induced rat hepatocarcinogenesis promoted by phenobarbital.

cD8+ cytotoxic T lymphocyte (cTL) exhaustion is one of the major obstacles for the effectiveness of virus control in chronic infectious diseases. We previously generated novel ovalbumin (OVA)-specific 41BBL-expressing OVA-TEXO and human immunodeficiency virus (HIV-1) Gag-specific Gag-TEXO vaccines, inducing therapeutic immunity in wild-type c57BL/6 (B6) mice, and converting cTL exhaustion in recombinant OVA-specific adenovirus AdVOVA-infected B6 (AdVOVA-B6) mice with chronic infection. IL-21 cytokine plays an important role in controlling chronic infections. Therefore, in this study, we constructed recombinant transgene IL-21-expressing AdVIL-21, and generated IL-21-expressing OVA-TEXO/IL-21 and Gag-TEXO/IL21 vaccines, or control vaccines (OVA-TEXO/Null and Gag-TEXO/Null) by infecting OVA-TEXO and Gag-TEXO cells with AdVIL-21 or the control AdVNull, lacking transgene, and assessed their effects in B6 or AdVOVA-B6 mice. We demonstrate that both OVA-TEXO/IL-21 and control OVA-TEXO/Null vaccines are capable of converting cTL exhaustion in chronic infection. However, the OVA-TEXO/IL-21 vaccine more efficiently rescues exhausted cTLs by increasing stronger cTL proliferation and effector cytokine IFN-γ expression than the control OVA-TEXO/Null vaccine in AdVOVA-B6 mice with chronic infection, though both vaccines stimulated comparable OVA-specific cTL responses and protective immunity against OVA-expressing BL6-10OVA melanoma lung metastasis in wild-type B6 mice. In vivo, the OVA-TEXO/IL-21-stimulated cTLs more efficiently up-regulate phosphorylation of mTORc1-controlled EIF4E and expression of mTORc1- regulated T-bet molecule than the control OVA-TEXO/Null-stimulated ones. Importantly, the Gag-TEXO/IL21 vaccine induces stronger Gag-specific therapeutic immunity against established Gag-expressing BL6-10Gag melanoma lung metastases than the control Gag-TEXO/Null vaccine in chronic infection. Hepatitis c virus is associated with Hepatitis c which can be acute or chronic. Acute hepatitis c is usually short term illness that can lead to chronic infection, while chronic hepatitis c is a chronic liver disease leading to cirrhosis, hepatocellular carcinoma and even death. The mode of transmission is mainly parenteral and vertical. Hepatitis c is the leading cause of morbidity and mortality and a serious public health problem, worldwide as well as in Pakistan. There is no vaccine available for hepatitis c so the only way to prevent hepatitis c is by changing behaviors which can spread the disease. Pakistan is also facing a huge burden of this disease. In Pakistan, the frequency of hepatitis c virus infection ranges from 8% - 15% in the general population with variations in different parts of the country. The general method of detecting infection with HcV is to observe the presence of antibodies to the virus. The aim of this study was to determine frequency of hepatitis c among general population and to see the increasing frequency in our part. Selection of Patients: All patients visiting outpatient clinics, Hepatitis clinic and indoor patients at civil Hospital Karachi, Pakistan from January 1, 2010 to December 31, 2014 were screened for anti HcV. Methods: Blood samples were collected from the patients in Red Topped vaccutainers and allowed to clot, then centrifuged and serum was screened for anti HcV by Immunochromatography (IcT) method. Results: A total of 156,990 patients were screened for anti-HcV, 23,511 (14.98%) patients were reported positive. During the year 2010, total 11,058 patients were screened for anti HcV, 1791 (16%) were positive, while in the year 2011 total 26,921 were screened and 3908 (14.5%) were positive, during the year 2012, total 23,321 were screened, 3036 (14%) were positive, in the year 2013 we screened total 40,483 patients 6070 (15%) were positive, while in 2014, total 55,484 patients were screened and 8584 were positive (15.5%). A Stability Indicating U-HPLc Method for Milnacipran in Bulk Drugs and Pharmaceutical Dosage Forms

The objective of the current study was to develop a validated, specific and stability-indicating reverse phase UHPLc method for the quantitative determination of Milnacipran and its related substances. The determination was done for active pharmaceutical ingredient and its pharmaceutical dosage forms in the presence of degradation products, and its process-related impurities. The drug was subjected to stress conditions of hydrolysis (acid and base), oxidation, pho- tolysis and thermal degradation per International conference on Harmonization (IcH) prescribed stress conditions to show the stability-indicating power of the method. Significant degradation was observed during acid, base, oxidative and neutral stress hydrolysis. The chromatographic conditions were optimized using an impurity-spiked solution and the samples generated from forced degradation studies. In the developed UHPLc method, the resolution between Milnacipran and its process-related impurities was found to be greater than 2.0. Regression analysis shows an r value (correlation coefficient) of greater than 0.999 for Milnacipran and it’s all the five impurities. The chromatographic separation was achieved on a c18 stationary phase. The method employed a linear gradient elution and the detection wavelength was set at 220 nm. The mobile phase consists of buffer and acetonitrile delivered at a flow rate of 0.2 mL?min–1. Buffer consists a mixture of 10 mM Sodium dihydrogen phosphate monohydrate and 10 mM hexane sulfonate sodium salt, pH adjusted to 2.5 using ortho phosphoric acid. The mobile phase A consists of buffer and acetonitrile (950:50, v/v) and mobile phase B consists of acetonitrile. The stress samples were assayed against a qualified reference standard and the mass balance was found to be close to 99.5%. The developed RP-Lc method was validated with respect to linearity, accuracy, precision and robustness.
