Ocimum sanctum extract preserves neuronal echotexture and controls seizure in lithium-pilocarpine induced status epilepticus rats

Objective:To investigate the effect of Ocimum sanctum hydroalcoholic extract(OSHE)on seizure control and neuronal injury in rats with lithium-pilocarpine-induced status epilepticus(SE).Methods:SE was induced by administering lithium chloride followed by pilocarpine 24 h later.OSHE was administered either alone or in combination with valproate(VPA)3 days before SE induction until 14 days post-SE induction.Seizure parameters were recorded on day 1(0-3 h),day 1-3 and day 4-14 post-SE.On day 14 post-SE,neurobehavioural tests(elevated plus maze and passive avoidance)were done followed by total antioxidant capacity,neuron-specific enolase,immunohistochemistry,and electron microscopic assessment in the hippocampus and cortex tissue.Results:OSHE+VPA provided more significant seizure protection(75%)than VPA(62.5%),OSHE(62.5%),or SE control(12.5%)(overall P=0.003).The latency to stage-3/4 seizures was increased and the number of stage-3/4 seizures was reduced in all treatment groups compared to the SE control group(P=0.002 and<0.001,respectively).The OSHE+VPA group also had better memory retention than other treatment groups(P<0.001)in the passive avoidance test.Total antioxidant capacity level was significantly higher and neuron-specific enolase was lower in the OSHE and OSHE+VPA groups compared to the SE control group.Electron microscopic study showed significant myelin sheath damage(67.5%,P<0.05)and axonal degeneration(51.8%,P<0.001)in the hippocampus of the SE control group,which were alleviated by OSHE or OSHE+VPA treatment.In immunohistochemical analysis,the OSHE,OSHE+VPA,and VPA groups had a significantly higher number of viable neurons and less neuronal loss compared to the SE control in the hippocampus(P<0.001).Conclusions:OSHE either alone or in combination with VPA shows better seizure control by preservation of neuronal echotexture and reducing oxidative stress in the hippocampus.

Liposome based drug delivery as a potential treatment option for Alzheimer’s disease

Alzheimer’s disease is a neurodegenerative condition leading to atrophy of the brain and robbing nearly 5.8 million individuals in the United States age 65 and older of their cognitive functions.Alzheimer’s disease is associated with dementia and a progressive decline in memory,thinking,and social skills,eventually leading to a point that the individual can no longer perform daily activities independently.Currently available drugs on the market temporarily alleviate the symptoms,however,they are not successful in slowing down the progression of Alzheimer’s disease.Treatment and cures have been constricted due to the difficulty of drug delivery to the blood-brain barrier.Several studies have led to identification of vesicles to transport the necessary drugs through the blood-brain barrier that would typically not achieve the targeted area through systemic delivered medications.Recently,liposomes have emerged as a viable drug delivery agent to transport drugs that are not able to cross the blood-brain barrier.Liposomes are being used as a component of nanoparticle drug delivery;due to their biocompatible nature;and possessing the capability to carry both lipophilic and hydrophilic therapeutic agents across the blood brain barrier into the brain cells.Studies indicate the importance of liposomal based drug delivery in treatment of neurodegenerative disorders.The idea is to encapsulate the drugs inside the properly engineered liposome to generate a response of treatment.Liposomes are engineered to target specific diseased moieties and also several surface modifications of liposomes are under research to create a clinical path to the management of Alzheimer’s disease.This review deals with Alzheimer’s disease and emphasize on challenges associated with drug delivery to the brain,and how liposomal drug delivery can play an important role as a drug delivery method for the treatment of Alzheimer’s disease.This review also sheds some light on variation of liposomes.Additionally,it emphasizes on the liposomal formulations

Application of light emitting diodes (LEDs) for food preservation, post-harvest losses and production of bioactive compounds: a review

Light-emitting diode (LED) technology is a new non-thermal food preservation method that works by converting light energy into heat. LED has potential to revolutionize crop production, protection and preservation. This technology is economical and environmentally friendly. LEDs have been shown to improve the nutritive quality and shelf life of foods, control the ripening of fruits, induce the synthesis of bioactive compounds and antioxidants and reduce the microbial contamination. This technology also has great scope in countries, where safety, hygiene, storage and distribution of foods are serious issues. While comparing this technology with other lighting technologies, LEDs can bring numerous advantages to food supply chain from farm to fork. In case of small growing amenities which exploit only LEDs, energy expenditure has been successfully reduced while producing nutritious food. LEDs can be used to give us better understanding and control over production and preservation of food with relation to spectral composition of light. LEDs also play significant role in food safety by inactivating the food borne pathogens. Therefore, LED lighting is a very effective and promising technology for extending shelf life of agricultural produce by increasing disease resistance and with increased nutritional values.

Semantic segmentation of landcover for cropland mapping and area estimation using Machine Learning techniques

The paper has focussed on the global landcover for the identification of cropland areas.Population growth and rapid industrialization are somehow disturbing the agricultural lands and eventually the food production needed for human survival.Appropriate agricultural land monitoring requires proper management of land resources.The paper has proposed a method for cropland mapping by semantic segmentation of landcover to identify the cropland boundaries and estimate the cropland areas using machine learning techniques.The process has initially applied various filters to identify the features responsible for detecting the land boundaries through the edge detection process.The images are masked or annotated to produce the ground truth for the label identification of croplands,rivers,buildings,and backgrounds.The selected features are transferred to a machine learning model for the semantic segmentation process.The methodology has applied Random Forest,which has compared to two other techniques,Support Vector Machine and Multilayer perceptron,for the semantic segmentation process.Our dataset is composed of satellite images collected from the QGIS application.The paper has derived the conclusion that Random forest has given the best result for segmenting the image into different regions with 99%training accuracy and 90%test accuracy.The results are cross-validated by computing the Mean loU and kappa coefficient that shows 93%and 69%score value respectively for Random Forest,found maximum among all.The paper has also calculated the area covered under the different segmented regions.Overall,Random Forest has produced promising results for semantic segmentation of landcover for cropland mapping.

Impact of ovariectomy and estradiol-17β(E_(2))replacement on the brain steroid levels of the Indian stinging catfish Heteropneustes fossilis

In the present study,effects of ovariectomy(Ovx)and estradiol-17β(E2)replacement on the brain steroid levels were investigated in the resting and prespawning phases of the female catfish Heteropneustes fossilis.The study showed that Ovx resulted in significant decreases in plasma levels of estradiol-17β(E2)and its precursors,and the effect varied with the reproductive stage and duration.Our study showed that E2 supplementation reversed the effect of Ovx in 3-week Ovx fish group,either restored or increased according to the dose.In contrast,E2 levels increased in the brain after Ovx in the resting stage and produced a biphasic effect of significant decreases on week 1 and 2,and significant increases on week 3,4 and 5 in the pre-spawning stage.The brain E2 levels significantly inhibited in E2 supplementation groups.The testosterone(T)levels showed biphasic effects,an initial decrease(up to 3 weeks Ovx)and an increase later(week 4,5)in the resting stage.In the pre-spawning stage,the T levels increased significantly after Ovx.The E2 supplementation significantly inhibited the T levels,more severely in the low dose group in the resting and pre-spawning stages.Brain 17-hydroxyprogesterone(17-OHP4)levels increased(week 1,2,3)but decreased on week 4 and 5 of Ovx in the resting stage and increased throughout Ovx in the pre-spawning stage except in the 5th week(though compared to other steroids,level was very low;almost negligible).The E2 replacement significantly decreased the steroid levels further down compared to the Ovx control group.Brain progesterone(P4)and pregnenolone(P5)levels decreased initially(week 1 and 2 Ovx)and recovered later.The E2 replacement significantly decreased P4 levels in the Ovx fish in the pre-spawning stage and in the lower dose group in the resting stage.The P5 levels remained inhibited after the E2 supplementation in the resting stage but unchanged or increased it in the pre-spawning stage.Brain cortisol(F)levels were inhibited initially(1 and 2 weeks and later increased after OVX in the rest

Assessment of terrace gardens as modifiers of building microclimate

Hard concrete roofs cause excessive heat gain impacting thermal comfort in buildings.Terrace gardens promoting greening at higher levels of built structures are seen as one of the key mitigating strategies for modifying building microclimate and improving urban health.We have undertaken a research project to quantitatively assess the value of a terrace garden in a residential scale.A garden patch of about 15 m 2 in area,a typical size available in most urban terraces is developed.Surface temperatures are measured over a period of 15 months between July 2018 to January 2020 using four thermocouple sensors,placed within and outside the garden bed,on and below the roof.We compare the thermal performance of the terrace garden across years,seasons,time of the day,presence or absence of garden bed and type and height of vegetation.The surface temperature data was found to correlate well with the ambient air temperature values.The results show that the terrace garden moderates and stabilises the ceiling temperatures and reduces it by about 2-3°C in winter months and 5-7°C in hot summers.The garden also provided nearly 400 g of fresh monthly vegetable harvest per m 2 of garden.Further,the cooling impact of the terrace garden with natural,tall,wild vegetation is higher as compared to planted vegetation.The study demonstrates a sustainable approach to terrace garden design at residential scale through quantified dual benefits of temperature control within buildings and urban farming.

Influence of slope position and aspect on the vegetation attributes and treewater relations in forests of the central Himalayas

While the need for understanding the effects of topographical factors on forest structure and function is well recognized,comprehensive studies are scarce.This study evaluates the effect of slope aspect and slope position on water relations and forest attributes across six forest types occurring between 400 m and 2600 m altitude in the Central Himalaya(27°-38°N).We found that predawn tree water potential and soil water potential were generally higher on moist north slope aspect(-0.78±0.05 MPa and-3.34±0.18 MPa,respectively)than dry south slope aspect(-0.82±0.18 MPa and-3.77±0.18 MPa,respectively).Across six different forests,these values were higher at hill base(-0.71±0.06 MPa and-2.77±0.19 MPa,tree predawn water potential and soil water potential,respectively)than other topographical positions.The favorable effect of north aspect and hill base was also observed in maintaining soil water and tree water potential during the dry season.Vegetation attributes,such as species richness,unique species and plant density were also generally higher on north slope and hill base than southern aspect and lowest at hill top.Across forest types,the hill base provided shelter to 46 unique species,compared to 16-18 at the other positions,thus emphasizing its importance as refugia for species to survive climate change induced perturbations.The favorable conditions of hill base position not only contribute to increase in alpha diversity,but also to extended species distributional range.

In this article,we are exploring the hemodynamics of nanofluid,flowing through a bifurcated artery with atherosclerosis in the presence of a catheter.For treating obstruction in the artery,one can use the catheter whose outer surface is carrying the drug coated with nano-particles.The resultant solvent is considered as blood nano-fluid.Blood being a complex fluid,is modeled by couple stress fluid.In the presence of nano-particles,the temperature and the concentration distribution are understood in a bifurcated stenotic artery.The concluded mathematical model is governed by coupled non-linear equations,and are solved by using the homotopy perturbation method.Consequently,we have explored is the effects of fluid and the embedded geometric parameters on the hemodynamics characteristics.It is also realized that high wall shear stress exists for couple stress nano-fluid when compared to Newtonian nano-fluid.which is computed at a location corresponding to maximum constriction(z=12.5)of the artery.

Detection of urinary antigens and their seroreactivity with serum of patients in Leishmania donovani infection

Objective:To detect leishmanial antigens in pre and post treated urine of visceral leishmaniasis (VL) patients.Methods:Urine and serum sample from three VL patients were collected. Ammonium sulphate precipitation and purification of urine sample was done for proteins isolation.SDS PAGE of proteins was done followed by western blotting,with the patient’s pre and post treatment serum.Results:Eight proteins of molecular weights 17 kDa,25 kDa,28 kDa,42 kDa, 47 kDa,54 kDa,60 kDa and 85 kDa were detected in the urine of VL patients before treatment. After treatment with miltefosine,none of the above proteins was detected in urine samples.The western blot analysis with pre treatment serum confirmed the antigenicity of four urinary proteins of molecular weights 25 kDa,28 kDa,54 kDa and 60 kDa.The seropositivity with 25 kDa and 28 kDa antigens was negative with serum obtained after the completion of treatment.Conclusions:In the context to unavailability of a prognostic tool,urinary leishmanial antigens may offer a better choice and may also be useful as immunoprophylactic candidates.

Natural compounds as potential inhibitors of SARS-CoV-2 main protease: An insilico study

Objective:To explore natural compounds as potential inhibitors against main protease(Mpro)of SARS-CoV-2.Methods:In the current study,systematic molecular docking analysis was conducted using AutoDock 4.2 to determine the binding affinities and interactions between natural compounds and Mpro.Selected natural compounds were further validated using a combination of molecular dynamic(MD)simulations and molecular mechanic Poisson-Boltzmann surface area(MM/PBSA)free energy c alculation s.Results:Out of twenty natural compounds,four natural metabolites namely,amentoflavone,guggulsterone,puerarin,and piperine were found to have strong interaction with Mpro of SARS-CoV-2 based on docking analysis.During MD simulations,all four natural compounds bound to Mpro at 50 ns and MM/G/P/BSA free energy calculations showed that all four shortlisted ligands had stable and favorable energies with strong binding to Mpro protein.Conclusions:Guggulsterone is a potential inhibitor of COVID-19 main protease Mpro.Further in vitro and pre-clinical studies are needed.
