In 2004, docetaxel was approved for the treatment of metastatic castration-resistant prostate cancer （mCRPC）. For the next several years, there was a lull in drug approvals. However, from 2010 onwards, 5 additional therapies have been approved on the basis of showing a survival benefit in phase III studies. These agents include sipuleuceI-T, cabazitaxel, abiraterone, enzalutamide and （most recently） radium-223. Amongst radiopharmaceuticals currently used for advanced prostate cancer （e.g. samarium-153 and strontium-89）, radium-223 possesses several unique properties. As an alpha-emitting compound, the agent produces a high-energy output over a short range, facilitating selective destruction of tissue within the bone in the region of osteoblastic lesions while sparing surrounding normal tissue. The current review will outline biological rationale for radium-223 and also provide an overview of preclinical and clinical development of the agent. Rational sequencing of radium-223 and combinations, in the increasingly complex landscape of mCRPC will be discussed, along with factors influencing clinical implementation.

Herein, we discuss 18-year follow-up data from the Scandinavian ProstateCancer Group-4 （SPCG-4） trial, a randomized study comparing observation and radical prostatectomy （RP） in patients with localized prostate cancer. The results of this study are contrasted with another study employing a similar randomization, the Prostate Cancer Intervention Versus Observation Trial （PIVOT）. We highlight several key differences in study eligibility and enrollment that may account for distinct results, and describe how these datasets impact the complex landscape of therapy for localized prostate cancer.

Despite great progress in the detection and treatment of prostate cancer, this disease remains an incredible health and economic burden. Although androgen receptor （AR） signaling plays a key role in the development and progression of prostate cancer, aberrations in other molecular pathways also contribute to the disease, making it essential to identify and develop drugs against novel targets, both for the prevention and treatment of prostate cancer. One promising target is the peroxisome proliferator-activated receptor gamma （PPARy） protein. PPARy was originally thought to act as a tumor suppressor in prostate cells because agonist ligands inhibited the growth of prostate cancer cells; however, additional studies found that PPARy agonists inhibit cell growth independent of PPARy. Furthermore, PPARy expression increases with cancer grade/stage, which would suggest that it is not a tumor suppressor but instead that PPARy activity may play a role in prostate cancer development and/or progression. Indeed, two new studies, taking vastly different, unbiased approaches, have identified PPARy as a target in prostate cancer and suggest that PPARy inhibition might be useful in prostate cancer prevention and treatment. These findings could lead to a new therapeutic weapon in the fight against prostate cancer.

Until recently, docetaxel-based therapy represented the only therapy shown to prolong survival in patients with metastatic castration-resistant prostate cancer （mCRPC）. The past year and a half has been marked by unprecedented progress in treatments for this disease. Three positive phase III clinical trials have emerged, each evaluating agents （sipuleuceI-T, cabazitaxel and abiraterone） with distinct mechanisms of action. Herein, the three pivotal trials are described alongside both past and current large phase III studies conducted in this mCRPC. The overall survival for patients with mCRPC treated in current clinical trials is considerably longer than noted in the past. We note that more recent trials with older agents have also shown improved survival and discuss potential non-therapeutic biases that influence this critical measure of outcome. The necessity for utilizing randomized trials when evaluating new therapeutics is emphasized given the changing prognosis in this mCRPC.

This study introduces a monitoring system that can accurately predict the quality of friction stir *** study involved four different sets of welding experiments using varying materials and tool *** goal was to create a universal monitoring system capable of predicting weld quality across these diverse experimental ***-time welding data was collected using instruments such as load cells and a power *** processing methods were then used to analyze this data and extract essential information about welding ***,federated learning(FL)was used to develop the universal monitoring *** method involves collective learning and model training,leading to a global model trained on decentralized data from the different experimental *** approach proved to be effective in forecasting weld quality,achieving a mean absolute error(MAE)of 10.44 for all welding ***,it offered benefits such as reduced latency and enhanced user data protection,while maintaining the accuracy of the global *** artificial neural network model was also developed for comparison with FL,achieving a MAE of 13.85 for the welding ***,this study demonstrates the effectiveness of training global and more reliable models,with multiple devices sharing their knowledge bases to train the global model effectively.

The Mw 9.0 Tohoku-Oki earthquake that hit the mainland Japan on 11 th March, 2011 had resulted a devastating Tsunami due to an active thrusting between the Pacific and the North American Plates. Static and kinematic offsets at the offshore epicentre of the Mw 9.0 event remain unanswered and being investigated along with their near and far field limiting distances from the epicentre. Accordingly, offset measurements from 60 continuously operating IGS and GEONET GNSS stations were radially classified from the epicentre and interpreted with analytical models to find their linear offset decay rates. Co-and post-seismic static positional anomaly offsets of sixty days show almost all near field stations had strong or appreciable eastward or south eastward static shifts. Near stations(km) showed both kinematic and static offsets. GEONET station ’0175’ showed maximum resultant static offset of-4.5 m, which diminishes approximately 1-2 cm at far sites like SMST and AIRA. Characteristic decay duration(’b’) of the mean kinematic co-seismic shift(’a’)of near field stations was 17.28 s during earthquake hours with an EW component shift >1.5 m. Spatial models of projected N-S static and kinematic offsets show their asymmetrical distributions around the epicentre with maximum model offset of-1.84 m displaced towards south at-45 km north of the epicentre. The Tohoku-Oki earthquake produced a resultant kinematic offset of-10.2 m towards East at its offshore epicentre;while the estimated near field static offset is ~9.82 m. However, both estimates are bigger than double the resultant offset measured value(~4.3 m) in the Japanese mainland using GPS. The difference in the kinematic and static near field offsets highlight that the near surface had elastic or in-elastic kinematic strain dissipation as against the lithospheric level viscoelastic static response, which resulted rapid kinematic strain release(1.12 cm/km)within the limiting radius of ~220 km from the Tohoku-Oki epicentr

Influence of Heat Input on Martensite Formation and Impact Property of Ferritic-Austenitic Dissimilar Weld Metals

The effect of heat input on martensite formation and impact properties of gas metal arc welded modified ferritic stainless steel （409M） sheets （as received） with thickness of 4 mm was described in detail in this work. The welded joints were prepared under three heat input conditions, i.e. 0.4, 0.5 and 0.6 kJ/mm using two different austenitic filler wires （308L and 316L） and shielding gas composition of Ar ＋ 5% CO2. The welded joints were evaluated by microstructure and charpy impact toughness. The dependence of weld metal microstructure on heat input and filler wires were determined by dilution calculation, Creq/Nieq ratio, stacking fault energy （SFE）, optical microscopy （OM） and transmission electron microscopy （TEM）. It was observed that the microstructure as well as impact property of weld metal was significantly affected by the heat input and filler wire. Weld metals prepared by high heat input exhibited higher amount of martensite laths and toughness compared with those prepared by medium and low heat inputs, which was true for both the filler wires. Furthermore, 308L weld metals in general provided higher amount of martensite laths and toughness than 316L weld metals.
