Erythropoietin modulates bone marrow stromal cell differentiation

Erythropoietin is essential for bone marrow erythropoiesis and erythropoietin receptor on non-erythroid cells including bone marrow stromal cells suggests systemic effects of erythropoietin. Tg6 mice with chronic erythropoietin overexpression have a high hematocrit, reduced trabecular and cortical bone and bone marrow adipocytes, and decreased bone morphogenic protein 2 driven ectopic bone and adipocyte formation. Erythropoietin treatment(1 200 IU·kg–1) for 10 days similarly exhibit increased hematocrit,reduced bone and bone marrow adipocytes without increased osteoclasts, and reduced bone morphogenic protein signaling in the bone marrow. Interestingly, endogenous erythropoietin is required for normal differentiation of bone marrow stromal cells to osteoblasts and bone marrow adipocytes.ΔEpoR_E mice with erythroid restricted erythropoietin receptor exhibit reduced trabecular bone, increased bone marrow adipocytes, and decreased bone morphogenic protein 2 ectopic bone formation. Erythropoietin treated ΔEpoR_E mice achieved hematocrit similar to wild-type mice without reduced bone, suggesting that bone reduction with erythropoietin treatment is associated with non-erythropoietic erythropoietin response. Bone marrow stromal cells from wild-type,Tg6, and ΔEpoR_E-mice were transplanted into immunodeficient mice to assess development into a bone/marrow organ. Like endogenous bone formation, Tg6 bone marrow cells exhibited reduced differentiation to bone and adipocytes indicating that high erythropoietin inhibits osteogenesis and adipogenesis, while ΔEpoR_E bone marrow cells formed ectopic bones with reduced trabecular regions and increased adipocytes, indicating that loss of erythropoietin signaling favors adipogenesis at the expense of osteogenesis. In summary, endogenous erythropoietin signaling regulates bone marrow stromal cell fate and aberrant erythropoietin levels result in their impaired differentiation.

Donkey milk is known for its beneficial aspects such as low fat and cholesterol,total protein and casein content,high lactose,whey protein,calcium,selenium and vitamin D3 *** also has nutraceutical and utilitarian properties that can boost invulnerability,regulate digestion and conveniently change gut *** current study was focused on the effect of probiotic bacterial metabolites from donkey milk against human bacterial *** effective isolate VITRAAJ14 was found to be non-hemolytic and was tolerant to acid,NaCl and bile *** activity by cross streak method showed maximum zone of *** diffusion method also exhibited effective inhibition against Escherichia coli(MTCC No.9721),Staphylococcus aureus(MTCC No.3160)and Salmonella enterica(MTCC No.8587)with fractions obtained from chloroform:ethyl *** metabolites capable of inhibiting pathogens were identified as oleyl alcohol by Gas chromatography-mass spectrometry(GCMS).Lead compounds detected in GCMS was studied for its functional group by Fourier transform infrared spectroscopy(FTIR),which revealed the presence of alcoholic stretch,carboxylic acid OH stretch,alkene and amide *** isolate VITRAAJ14 was identified to be the closest neighbour of Bacillus species(MH100985)by 16S rRNA gene *** our knowledge,current study is the first-time report on the presence of Bacillus *** donkey milk showing antibacterial activity due to its ability to produce oleyl *** electron microscopic images revealed the presence of rod-shaped morphology of the effective bacterium VITRAAJ14.

Do forest health threats affect upland oak regeneration and recruitment?Advance reproduction is a key co-morbidity

We analyzed national forest inventory data collected from circa 2000–2018 across 37 states in the eastern United States to better understand the influence of forest health related canopy disturbances on the regeneration and recruitment dynamics of upland oaks(Quercus).We found low levels of oak recruitment across all disturbance types examined but limited evidence of any direct effects from the type of disturbance on the population of regenerating *** general lack of differences in oak regeneration response between forest health disturbances and disturbances caused by harvested or non-disturbed plots does not indicate that the effects of forest health disturbances were benign,***,low level of oak recruitment across all disturbance types highlights the pervasiveness of the trend of shifting composition in once oak-dominated forests where oak is absent or sparse in the regeneration *** results show that oak recruitment was higher when oak was present as advance reproduction prior to disturbance from any cause ***,these results lead us to conclude that the widespread inadequacy of oak advance reproduction in mature oak-dominated forests is the prevailing threat to oak forest health and *** suggest the status of advance reproduction be treated as a co-morbidity when weighing the risk and potential outcomes from other threats to upland oak forests in the eastern United States.

Photolysis studies on HCOOH and HCOO^- in presence of TiO_2 photocatalyst as suspension in aqueous medium

Photolysis studies on formic acid （HCOOH） and formate ion （HCOO-） in presence of TiO2, a photocatalyst, as suspension in water were carried out separately using 350 nm ultraviolet light. The products, such as H2, CO, CO2 and CH4, generated during the experiments were monitored with varying the ambient, light exposure time, and the concentration of HCOOH/HCOO-. The yields of CO in all these systems increased with light exposure time. In aerated systems, CO yields were higher in contrast to the deoxygenated （Ar-purged） systems under identical conditions. It is proposed apparently that the formation of CO is taking place during the chemical reduction of in-situ generated CO2, a photo-mineralized product of HCOOH/HCOO-, but not through the direct photodecomposition or photodehydration （CO＋H2O） of solute molecules. The rates of CO formation during 1.3 M HCOOH photolysis in presence of TiO2 photocatalyst were evaluated to be 0.21 and 0.13 μl/min in aerated and Ar-purged systems, respectively. As compared with HCOOH systems, the CO yields are lower when 0.2 M HCOONa was exposed to light under identical conditions. The CO growth rates were evaluated to be 0.07 and 0.046 μl/min^-1 for aerated and deoxygenated HCOONa systems, respectively; moreover, the trend is quite similar to that of the HCOOH system. Under these conditions, the emission of H2 was also observed, and its yield was significantly higher in Ar-purged system as compared with the CO yields. However, in aerated system, the yields of these products were just opposite. The formation of low yield of methane was observed during photolysis of HCOOH/HCOO ions. In CO2 ambient, the yields of CO and H2 varied drastically with time.

High intensity interval training(HIIT)causes oxidative stress and haematological *** study was aimed to evaluate the effect of 8 weeks’supplementation of vitamin C and E on HIIT induced changes in lipid profile parameters and haematological *** six male adolescent players were randomly assigned into five age-matched groups,i.e.,Control(no exercise+placebo),HIIT(placebo),HIIT+vitamin-C(1000mg/day),HIIT+vitamin-E 400 IU/day)and combined HIIT+vitamin C and *** and evening sessions(90min)of HIIT included 4 phases(15min each)with 3 sets(4min each).Each 4min HIIT set consisted of 2min intense sprint workout(90%–95%of heart rate maximum[HRmax])followed by 1min active recovery(60%–70%HRmax)followed by 1min of complete rest(1:1 work-rest ratio).Lipid profile parameters,haematological variables,endurance capacity and vertical jump were evaluated by standard *** decrease in body weight,fat%,total cholesterol,triglyceride,Total Cholesterol/High Density Lipoprotein-Cholesterol and significant increase in High Density Lipoprotein-Cholesterol,maximal oxygen consumption,vertical jump were observed for all four intervention *** blood cell count,red blood cell count,haemoglobin percentage and haematocrit values were significantly decreased while platelet count and platelet-to-leukocyte ratio(PLR)ratio were increased significantly only for HIIT *** level of tocopherol and ascorbic acid was significantly increased(values were within the normal range)in all the respective vitamin supplemented *** of vitamin C and E secures health protection with suppressed haemolysis and improved inflammatory blood variables with enhanced explosive leg strength and lipid profile parameters without any concomitant change in endurance capacity.

Design of a Proportional-Integral-Derivative Controller for an Automatic Generation Control of Multi-area Power Thermal Systems Using Firefly Algorithm

Essentially, it is significant to supply the consumer with reliable and sufficient power. Since, power quality is measured by the consistency in frequency and power flow between control areas. Thus, in a power system operation and control,automatic generation control(AGC) plays a crucial role. In this paper, multi-area(Five areas: area 1, area 2, area 3, area 4 and area 5) reheat thermal power systems are considered with proportional-integral-derivative(PID) controller as a supplementary controller. Each area in the investigated power system is equipped with appropriate governor unit, turbine with reheater unit, generator and speed regulator unit. The PID controller parameters are optimized by considering nature bio-inspired firefly algorithm(FFA). The experimental results demonstrated the comparison of the proposed system performance(FFA-PID)with optimized PID controller based genetic algorithm(GAPID) and particle swarm optimization(PSO) technique(PSOPID) for the same investigated power system. The results proved the efficiency of employing the integral time absolute error(ITAE) cost function with one percent step load perturbation(1 % SLP) in area 1. The proposed system based FFA achieved the least settling time compared to using the GA or the PSO algorithms, while, it attained good results with respect to the peak overshoot/undershoot. In addition, the FFA performance is improved with the increased number of iterations which outperformed the other optimization algorithms based controller.
