Plasmid vector based generation of transgenic mesenchymal stem cells with stable expression of reporter gene in caprine

The production of cells capable of expressing gene(s) of interest is important for a variety of applications in biomedicine and biotechnology, including gene therapy and a novel method of stem cell therapy in the various diseases. Achieving high levels of transgene expression for the longer period of time, without adversely affecting cell viability and differentiation capacity of the cells, is crucial. In the present study, we investigated the efficiency of plasmid vector for the production of transgenic cMSCs and examined any functional change of cells after transfection. To do so first we have collected bone marrows from the adult goats and cultured them for isolation of mesenchymal stem cells (cBM-MSCs). These cells were characterized using MSC specific markers including differentiation into osteocytes and adipocytes. Transfection with plasmid vector did not adversely affect cBM-MSCs morphology, viability or differentiation potential, and transgene expression levels were unaffected beyond passage 12th. The results indicated that we have been able to generate transgenic caprine MSC (tcBM-MSC) and transfection of cBM-MSCs using plasmid vector resulted in very high and stable transfection efficiency. This finding may have considerable significance in improving the efficacy of MSC-based therapies and their tracking in animal model.

Networks form an integral part of our daily lives, comprising structures known as nodes connected via ties. These networks exhibit a complex interplay of random and structured features. We encounter networks regularly in our daily activities, whether it is logging into websites, commuting by train, going to work, or attending school. Networks are prevalent in various academic disciplines and practical applications, spanning biology, physics, engineering, social science, and numerous other fields. The early 21st century has witnessed a rapid expansion of techniques facilitating human interaction, significantly impacting behavior and social bonding in both urban and rural settings. The availability and accessibility of extensive population datasets have spurred ongoing research into understanding and shaping human inclinations and sociological phenomena in unconventional ways. This review delves into the nuanced aspects of social physics, exploring the rationale behind choosing this research topic, various conceptual frameworks, categorizations, and distinctions between machine learning and deep learning. The article briefly outlines the advantages of leveraging big data in social physics, highlighting its transformative potential. Lastly, the authors provide insights into several applications of social physics in subsequent sections, offering a glimpse into its real-world implications.

Nonlinear thermomechanical deformation behaviour of P-FGM shallow spherical shell panel

In the present article, the linear and the nonlinear deformation behaviour of functionally graded （FG） spherical shell panel are examined under thermomechanical load. The temperature- dependent effective material properties of FG shell panel are evaluated using Voigt＇s micro-mechanical rule in conjunction with power-law distribution. The nonlinear mathematical model of the FG shell panel is developed based on higher-order shear deformation theory and Green-Lagrange type geometrical nonlinearity. The desired nonlinear governing equation of the FG shell panel is computed using the variational principle. The model is discretised through suitable nonlinear finite element steps and solved using direct iterative method. The convergence and the val- idation behaviour of the present numerical model are performed to show the efficacy of the model. The effect of different parameters on the nonlinear deformation behaviour of FG spherical shell panel is highlighted by solving numerous examples.

This paper explains an integrated production inventory supply chain model,which consists of a supplier,a manufacturer and a retailer under two-level credit *** is manufacturer’s credit period offered by the supplier,and other is retailer’s credit period offered by the ***,the manufacturer replenishes raw materials from the supplier and produces non-deteriorating products in some cycles of equal *** is noted that here four inventory structures have been considered to show the flow of the materials as either raw materials or finished products from supplier to retailer via *** in this model,manufacturer’s imperfect production has been *** imperfect items are not *** retailer receives good finished products from the manufacturer and sells these to his/her customers during some cycles of equal length in total time *** main objective is to find the optimal number of production and business cycles of the manufacturer such that the integrated system can get the maximum *** above discussed model is elaborated with the help of a numerical example,and a sensitivity analysis is done with respect to some parameters used in this model.

Wear and performance analysis of a 4-stroke diesel engine employing nanolubricants

In the present study, the performance of a 4-stroke diesel engine was experimentally evaluated upon adding Al2O3 or SiO2 nanoparticles to the engine oil （SAEISW40）. The viscosity and density of the resulting nanolubricants were determined while varying both the nanoparticle volume fraction and the temperature. Field emission scanning electron microscopy （FE-SEM） showed that the nanoparticles had a spherical morphology and dynamic light scattering analysis determined some aggregation of the nanoparticles in the engine oil. A pin-on-disc test apparatus was used for friction and wear analysis in the presence of the nanolubricants. Examination of wear scars by FE-SEM and energy dispersive spectroscopy found evidence of ball bearing and surface polishing effects, which were responsible for improvements in the tribological properties of the oil. The performance of these nanolubricants in a 4- stroke diesel engine test rig was assessed, and the greatest improvements in the tribological behavior and engine performance were observed when employing 0.3 vol% Al2O3.

As fossil fuel stocks are being depleted,alternative sources of energy must be ***,traditional thermal power plants must coexist with renewable resources,such as wind,solar,and hydro units,and all-day planning and operation techniques are necessary to safeguard nature while meeting the current *** fundamental components of contemporary power systems are the simultaneous decrease in generation costs and increase in the available transfer capacity(ATC)of current *** units are linked to sources of renewable energy such as hydro,wind,and solar power,and are set up to run for 24 *** contrast,new research reports that various chaotic maps are merged with various existing optimization methodologies to obtain better results than those without the inclusion of *** seems to increase the performance and convergence properties of existing optimization *** this study,selfish animal tendencies,mathematically represented as selfish herd optimizers,were hybridized with chaotic phenomena and used to improve ATC and/or reduce generation costs,creating a multi-objective optimization *** evaluate the performance of the proposed hybridized optimization technique,an optimal power flow-based ATC was enforced under various hydro-thermal-solar-wind conditions,that is,the renewable energy source-thermal scheduling concept,on IEEE 9-bus,IEEE 39-bus,and Indian Northern Region Power Grid 246-bus test *** findings show that the proposed technique outperforms existing well-established optimization strategies.

Chemical Enhance oil Recovery(CEOR)technology is getting more attention since energy crises are getting worse and *** application of polymer flooding depends on the evaluation of rheological and solution properties of injected slug at reservoir conditions,which is function of polymer concentration,salinity,temperature and shear *** acryl amide(PAM)is commonly used polymer in polymer flooding ***,comparative study with other polymers such as xanthan gum(XG)is required while selection of appropriate polymer for *** present study,the recovery efficiency of PAM and XG as a polymer has been evaluated for application as chemical EOR agent for tertiary phase polymer flooding in Upper Assam *** comparative study between PAM and XG has been done in terms of rheological *** flooding experiment of polymer flooding has also been tested to determine the recovery efficiency of the PAM and XG polymer *** results showed that these polymers exhibit favorable salt tolerance,temperature resistance,and recoverable viscosity after shearing,reasonable thickening behavior and improved viscosity enhancement properties due to presence of hydrophobic association in the polymer main *** flooding experiment of polymer flooding has also been tested to determine the recovery efficiency of the PAM and XG polymer *** show that macroscopic sweep efficiency is the dominant factor during *** an injection of hydrogel polymer to the reservoir is to increase a viscosity of fluid containing water so that the fluid is more difficult to flow than the oil,and as a result,the oil production *** thinning behavior was observed in case of PAM indicating predominance of the frontal advance *** PAM seems to be following both piston like movement and frontal advance theory it can be efficiently enhancing the macroscopic sweep efficiency as well as microscopic displacement *** overall

Polytene chromosomes of an Indian Himalayan black fly Simulium (Nevermannia) praelargum (Diptera:Simuliidae)

High quality polytene chromosome maps （n=3） of a Himalayan Simuliid Simulium praelargum Datta, 1973 are presented and represent the first cytological description of a taxon found in the feuerborni group, subgenus Nevermannia. Polytene chromosomes one （I） and two （II） are metacentric, chromosome three （III） is submetacentric with the length of each chromosome occupying 37.25 %, 31.36 % and 31.34 % of the total complement length, respectively. Typical simuliid diagnostic intergeneric chromosomal markers are found within the polytene complement of this species. The nucleolar organizer （N.O.） is found at the base of the short arm of chromosome one （IS）, the Ring of Balbiani （R.B.）, double bubble （D.B.） and triad occur in the short arm of chromosome two （IIS）, the Parabalbiani Ring （EB.） and grey band （gb） occur in the long arm of chromosome two （IIL） and the Blister （BL） and Capsule （Ca） occur in the short arm of chromosome three （IIIS）.Terminal bands at the end of IIIS are heterochromatinized and present atypically with respect to other simuliid fauna. Populations studied so far are unique among the Simuliidae in that they exhibit chromosome structural monomorphism. These high resolution polytene chromosome maps will form the basis for future cytological characterization and phylogenetic comparisons amongst members of the feuerborni group

Molecular differentiation of cryptic stage of Echinococcus granulosus and Taenia species from faecal and environmental samples

Objective:To differentiate cryptic stage of Echinococcus granulosus(***) and Taenia by PCR-RFLP and sequence information of ***:DNA were isolated from metacestodes stage of Taenia and *** using DNA isolation kit(Q-BIOgene kit,USA), the amplified and purified DNA product was then cloned and sent for *** generating sequence information was used for amplicons ***:Out of 112 faecal and environmental samples,16 exhibited positive *** product size of amplicon positive for *** was 310 bp;whereas,for Taenia *** varied from 379 to 388 *** pr of ile of actinⅡwith Csp61 also differed Taenia *** ***:The result of the study indicated that,the primers were useful to differentiate cryptic stage of the two genera which is yet to be reported earlier.

Well-Defined Fluorinated Copolymers:Current Status and Future Perspectives
