Effects of amyloid precursor protein peptide App96-110,alone or with human mesenchymal stromal cells,on recovery after spinal cord injury

Delivery of a peptide(App96-110),derived from amyloid precursor protein(App),has been shown to elicit neuroprotective effects following cerebral stroke and traumatic brain *** this study,the effect of App96-110 or a mutant version of this peptide(mApp96-110)was assessed following moderate(200 kdyn,(2 N))thoracic contusive spinal cord injury(SCI)in adult Nude *** received a single tail vein injection of App96-110 or mApp96-110 at 30 minutes post-SCI and were then assessed for functional improvements over the next 8 weeks.A cohort of animals also received transplants of either viable or non-viable human mesenchymal stromal cells(hMSCs)into the SC lesion site at one week post-injury to assess the effect of combining intravenous App96-110 delivery with hMSC *** were perfused 8 weeks post-SCI and longitudinal sections of spinal cord analyzed for a number of factors including hMSC viability,cyst size,axonal regrowth,glial reactivity and macrophage *** of sensorimotor function revealed occasional significant differences between groups using Ladderwalk or ratwalk tests,however there were no consistent improvements in functional outcome after any of the ***96-110 alone,and App96-110 in combination with both viable and non-viable hMSCs significantly reduced cyst size compared to SCI *** treatments with donor hMSCs also significantly increased βIII tubulin^(+),glial fibrillary acidic protein(GFAp^(+))and laminin+expression,and decreased ED1^(+)expression in *** preliminary study demonstrates that intravenous delivery of App96-110 peptide has selective,modest neuroprotective effects following SCI,which may be enhanced when combined with hMSC ***,the effects are less pronounced and less consistent compared to the protective morphological and cognitive impact that this same peptide has on neuronal survival and behaviour after stroke and traumatic brain *** while the efficacy of a

In this study,nitrogen doped electrochemically exfoliated reduced graphene oxide and carbon black supported platinum(pt/Nr EGO_(2)-CB_(3))has been prepared to enhance the performance and durability of hightemperature pEMFCs with lower pt *** the one hand,pt/Nr EGO_(2)-CB_(3)with the strong interaction between the pt and nitrogen(N)prevent agglomeration of pt particles and pt particles is 5.46±1.46 nm,which is smaller than that of 6.78±1.34 nm in pt/***,ECSA of pt/Nr EGO_(2)-CB_(3)decrease 13.65%after AST,which is much lower than that of 97.99%in pt/*** the other hand,the Nr EGO flakes in MEAac act as a barrier to mitigate phosphoric acid redistribution,which improves the formation of triple-phase boundaries(TpBs)and gives stable operation of the MEAacwith a lower decay rate of 0.02 mV h^(-1)within100 *** steady-state operation,the maximum power density of pt/Nr EGO_(2)-CB_(3)(0.411 W cm^(-2))is three times higher than that of conventional pt/C(0.134 W cm^(-2))in high-temperature *** AST,the mass transfer resistance of pt/Nr EGO_(2)-CB_(3)electrode(0.560Ωcm^(2))is lower than that in pt/C(0.728Ωcm^(2)).

The doping of functionalized graphene oxide(GO)in the membranes becomes a promising method for improving the performance of high-temperature proton exchange membrane fuel cells(HT-pEMFC).phosphonated graphene oxide(pGO)with a p/O ratio of 8.5%was quickly synthesised by one-step electrochemical exfoliation based on a three-dimensiaonal(3D)printed reactor and natural graphite *** with the GO prepared by the two-step electrochemical exfoliation method,the pGO synthesized by the one-step electrochemical exfoliation can better improve the performance of the membrane-electrode-assembly(MEA)based on the polybenzimidazole(pBI)membrane in the *** doping of 1.5 wt%GO synthesised by electrochemical exfoliation with the 2-step method or reactor method in pBI increased the peak power density by 17.4%or 35.4%compared to MEA based on pure pBI membrane at 150℃,*** addition,the doping of pGO in pBI improves its durability under accelerated stress test(AST).

primary sclerosing cholangitis(pSC)is an orphan,cholestatic liver disease that is characterized by inflammatory biliary strictures with variable progression to end-stage liver *** pathophysiology is poorly *** biliary inflammation is likely driven by immune dysregulation,gut dysbiosis,and environmental exposures resulting in gutliver crosstalk and bile acid metabolism *** is no proven medical therapy that alters disease progression in pSC,with the commonly prescribed ursodeoxycholic acid being shown to improve liver biochemistry at low-moderate doses(15–23 mg/kg/day)but not alter transplant-free survival or liver-related *** transplantation is the only option for patients who develop end-stage liver disease or refractory complications of *** and antifibrotic agents have not proven to be effective,but there is promise for manipulation of the gut microbiome with fecal microbiota transplantation and *** acid manipulation via alternate synthetic bile acids such as norursodeoxycholic acid,or interaction at a transcriptional level via nuclear receptor agonists and fibrates have shown potential in phase II trials in pSC with several leading to larger phase III *** view of the enhanced malignancy risk,statins,and aspirin show potential for reducing the risk of colorectal cancer and cholangiocarcinoma in pSC *** patients who develop clinically relevant strictures with cholestatic symptoms and worsening liver function,balloon dilatation is safer compared with biliary stent insertion with equivalent clinical efficacy.

It is widely accepted that further development of pb-free solder alloys for improved processing and in-service properties of next-generation electronics,can be accelerated through a fundamental understanding of phase transformation and microstructure control in pb-free solder *** characterization techniques including synchrotron radiation provide a comprehensive toolset to measure the composition,crystallography,morphology,and properties of the major components of solder *** research using such techniques is reviewed in detail including the characterization of the eff ects of microalloy additions on the microstructure and properties of pb-free solder joints,especially those on the intermetallic *** discoveries outlined are of scientific and industrial relevance and have implications for new solder alloy composition design and the reliability of lead-free solder joints.

Search-Based Software Test Data Generation for path Coverage Based on a Feedback-Directed Mechanism

Automatically generating test cases by evolutionary algorithms to satisfy the path coverage criterion has attracted much research attention in software *** the context of generating test cases to cover many target paths,the efficiency of existing methods needs to be further improved when infeasible or difficult paths exist in the program under *** is because a significant amount of the search budget(i.e.,time allocated for the search to run)is consumed when computing fitness evaluations of individuals on infeasible or difficult *** this work,we present a feedback-directed mechanism that temporarily removes groups of paths from the target paths when no improvement is observed for these paths in subsequent *** fulfill this task,our strategy first organizes paths into ***,in each generation,the objective scores of each individual for all paths in each group are summed *** each group,the lowest value of the summed up objective scores among all individuals is assigned as the best aggregated score for a group.A group is removed when no improvement is observed in its best aggregated score over the last two *** experimental results show that the proposed approach can significantly improve path coverage rates for programs under test with infeasible or difficult paths in case of a limited search *** particular,the feedback-directed mechanism reduces wasting the search budget on infeasible paths or on difficult target paths that require many fitness evaluations before getting an improvement.
