Recent improvements in the speed and accuracy of DNA sequencing, together with increasingly sophisti- cated mathematical approaches for annotating gene networks, have revolutionized the field of human genetics and made these once time consuming approaches assessable to most investigators. In the field of bone research, a particularly active area of gene discovery has occurred in patients with rare bone disorders such as osteogenesis imperfecta （OI） that are caused by mutations in single genes. In this perspective, we highlight some of these technological advances and describe how they have been used to identify the genetic determinants underlying two previously unexplained cases of OI. The widespread availability of advanced methods for DNA sequencing and bioinformatics analysis can be expected to greatly facilitate identification of novel gene networks that normally function to control bone formation and maintenance.

Shenfu Dongsheng Coal Co. Ltd. of the Shenhua Group is a modern mining enterprise. To construct this first class world enterprise, Shendong Company placed em- phasis on construction and production, and complied the internal administration with the international standards spontaneously. In order to strengthen the administration of the en- terprise, enhance the running efficiency of the systems, achieve the best administrative performances and goals, the Shendong Company began to engineer the integrated man- aging system(SDIMS) in early 2002. This article, as a case study, introduces the planning and construction processes of the SDIMS’s, and sums up the empirical experiences for modern and international mining enterprise.

Cellular reprogramming has rapidly become a promising methodology to generate new cardiomyocytes from non-cardiomyocyte cell *** the transient expression of OSKM factors,Chen et *** a unique reprogramming strategy involving the modulation of the resident adult cardiomyocyte identity to an immature proliferative state(Science 373:1537–40,2021).This OSKM-mediated reversion results in the adoption by adult murine cardiomyocytes of a transcriptional profile similar to cardiomyocytes found in developing hearts,as well as increased proliferative capacity of these reprogrammed cardiomyocytes compared to mature ***,this novel approach enhances the regeneration of adult murine hearts post-myocardial *** concerns and questions remain,the encouraging results of this study advance the field of cardiac regeneration by providing a new technique to generate cardiomyocytes as well as insights into cardiomyocyte dedifferentiation and its relation to proliferation.

Lymphocyte activation leads to changes in chemokine receptor *** are limited data,however,on how lymphocyte activators can alter chemokine signaling by affecting downstream *** hypothesized that B cell-activating agents might alter chemokine responses by affecting downstream signal transducers,and that such effects might differ depending on the *** found that activating mouse B cells using either anti-IgM or lipopolysaccharide(LPS)increased the surface expression of CCR6 and CCR7 with large increases in chemotaxis to their cognate *** contrast,while anti-IgM also led to enhanced calcium responses,LPS-treated cells showed only small changes in calcium signaling as compared with cells that were freshly *** particular interest,we found that LPS caused a reduction in the level of B-cell phospholipase C(PLC)-b2 mRNA and *** obtained using PLC-b22/2 mice showed that the b2 isoform mediates close to one-half the chemokine-induced calcium signal in resting and anti-IgM-activated B cells,and we found that calcium signals in the LPS-treated cells were boosted by increasing the level of PLC-b2 using transfection,consistent with a functional effect of downregulating ***,our results show activator-specific effects on responses through B-cell chemokine receptors that are mediated by quantitative changes in a downstream signal-transducing protein,revealing an activity for LPS as a downregulator of PLC-b2,and a novel mechanism for controlling chemokine-induced signals in lymphocytes.

Injuries to the postnatal skeleton are naturally repaired through successive stepsinvolving specific cell types in a process collectively termed “bone regeneration”.Although complex, bone regeneration occurs through a series of well-orchestratedstages wherein endogenous bone stem cells play a central role. In most situations,bone regeneration is successful;however, there are instances when it fails andcreates non-healing injuries or fracture nonunion requiring surgical or therapeuticinterventions. Transplantation of adult or mesenchymal stem cells (MSCs) definedby the International Society for Cell and Gene Therapy (ISCT) as CD105+-CD90+CD73+CD45-CD34-CD14orCD11b-CD79αorCD19-HLA-DR- is beinginvestigated as an attractive therapy for bone regeneration throughout the *** isolated from adipose tissue, adipose-derived stem cells (ADSCs), aregaining increasing attention since this is the most abundant source of adult stemcells and the isolation process for ADSCs is straightforward. Currently, there isnot a single Food and Drug Administration (FDA) approved ADSCs product forbone regeneration. Although the safety of ADSCs is established from their usagein numerous clinical trials, the bone-forming potential of ADSCs and MSCs, ingeneral, is highly controversial. Growing evidence suggests that the ISCT definedphenotype may not represent bona fide osteoprogenitors. Transplantation of bothADSCs and the CD105- sub-population of ADSCs has been reported to induce bone regeneration. Most notably, cells expressing other markers such as CD146,AlphaV, CD200, PDPN, CD164, CXCR4, and PDGFRα have been shown torepresent osteogenic sub-population within ADSCs. Amongst other strategies toimprove the bone-forming ability of ADSCs, modulation of VEGF, TGF-β1 andBMP signaling pathways of ADSCs has shown promising results. The U.S. FDAreveals that 73% of Investigational New Drug applications for stem cell-basedproducts rely on CD105 expression as the “positive” marker for adult stem cell
