Identification of osteopontin as the most consistently over-expressed gene in intrahepatic cholangiocarcinoma: Detection by oligonucleotide microarray and real-time PCR analysis

AIM: To investigate the molecular pathways involved in human cholangiocarcinogenesis by gene expression profiling. METHODS: Oligonucleotide arrays (Affymetrix U133A) were used to establish a specific gene expression profile of intrahepatic CCC in comparison to corresponding non- malignant liver tissue. To validate the expression values of the most overexpressed genes, RT-PCR experiments were performed. RESULTS: Five hundred and fifty-two statistically differentially expressed genes/ESTs (221 probes significantly up-regulated, 331 probes down-regulated; P 2; ≥ 70%) were identified. Using these data and two-dimensional cluster analysis,a specific gene expression profile was obtained allowing fast and reproducible differentiation of CCC, which was confirmed by supervised neuronal network modelling. The most consistently overexpressed gene (median fold change 33.5, significantly overexpressed in 100%) encoded osteopontin. Furthermore, an association of various genes with the histopathological grading could be demonstrated. CONCLUSION: A highly specific gene expression profile for intrahepatic CCC was identified, allowing for its fast and reproducible discrimination against non- malignant liver tissue and other liver masses. The most overexpressed gene in intrahepatic CCC was the gene encoding osteopontin. These data may lead to a better understanding of human cholangiocarcinogenesis.

Nowadays the protection of the marine environment raises increasing academic and public *** issue of organic micropollutants is of equally high importance for the marine *** vessels are considered to significant sources of micropollutants especially if the ship carries many passengers,which is often true for cruise ships which frequent attractive and sensitive sea *** emission pathways for micropollutants include wastewater discharges and sewage sludge *** findings of the German research and development project NAUTEK contribute to bridging the knowledge gap about micropollutant emissions from cruise *** expected,micropollutants were detected in both the blackwater and greywater on board,emitted from either the passengers or certain ship *** total,16 out of 21 target substances were *** concentrations of pharmaceuticals could be found mainly in blackwater(peak *** 3.9 mg/L,Ibuprofen 29 mg/L,Diclofenac 0.04 mg/L),while greywater is mainly characterized by substances such as ointment residues,UV-filters and flame retardants(peak *** 0.65 mg/L,Bisphenol A 8 mg/L,Tris(1-chloro-2-propyl)phosphate 136 mg/L).Further analyses suggest a gradual removal of the micropollutants by the onboard MBR plant(MBR effluent peak *** 0.47 mg/L,Ibuprofen 6.8 mg/L,Diclofenac 0.3 mg/L).Findings of this research provide a critical stepstone for shaping technical solutions for onboard micropollutants removal and water resource recycling.

High-dose interferon-α2b induction therapy in combination with ribavirin for treatment of chronic hepatitis C in patients with non-response or relapse after interferon-a monotherapy

AIM： To evaluate the daily high-dose induction therapy with interferon-α2b （IFN-α2b） in combination with ribavirin for the treatment of patients who failed with interferon monotherapy and had a relapse, based on the assumption that the viral burden would decline faster, thus increasing the likelihood of higher response rates in this difficult-totreat patient group. METHODS： Seventy patients were enrolled in this study. Treatment was started with 10 NU IFN-α2b daily for 3 wk, followed by IFN-α2b 5 NU/TIW in combination with ribavirin （1 000-1 200 mg/d） for 21 wk. In case of a negative HCV RNA PCR, treatment was continued until wk 48 （IFN-α2b 3MU/TIW＋1000-1200 mg ribavirin/daily）. RESULTS： The dose of IFN-α2b or ribavirin was reduced in 16% of patients because of hematologic side effects, and treatment was discontinued in 7% of patients. An early viral response （EVR） was achieved in 60% of patients. Fifty percent of all patients achieved an end-oftreatment response （EOT） and d0% obtained a sustained viral response （SVR）. Patients with no response had a significantly lower response rate than those with a former relapse （SVR 30% vs 53%; P=0.049）. Furthermore, lower response rates were observed in patients infected with genotype la/b than in patients with non-1-genotype （SVR 28% vs7d%; P=0.001）. As a significant predictive factor for a sustained response, a rapid initial decline of HCV RNA could be identified. No patient achieving a negative HCV-RNA PCR at wk 18 or later eventually eliminated the virus. CONCLUSION： Daily high-dose induction therapy with interferon-α2b is well tolerated and effective for the treatment of non-responders and relapsers, when interferon monotherapy fails. A fast decline of viral load during the first 12 wk is strongly associated with a sustained viral response.

A recent study demonstrated that in small-scale prepolarized surface nuclear magnetic resonance(SNMR-PP)measurements with a footprint of a few square meters,customized PP switch-off ramps can serve as an efficient excitation mechanism,eliminating the requirement for a conventional oscillating excitation *** approach enables the detection of short relaxation signals from the unsaturated soil zone and can,therefore,be used to directly provide soil moisture and pore geometry *** ultimately such small-scale SNMR-PP setups are intended for a mobile application,it is necessary to develop strategies that allow for speedy measurement progress and do not require noise cancellation protocols based on reference ***,we developed a new concentric figure-of-eight(cFOE)loop layout that combines the direction independence of a circular loop with the intrinsic noise cancellation properties of a classical *** approach significantly decreases the measurement time because suitable signal-to-noise ratios are reached much faster compared to a classical circular loop and will bring us one step further toward fast and non-invasive soil moisture mapping applications.

Polarization of M2 Line Emitted Following Electron-Impact Excitation of Beryllium-Like Ions

Detailed calculations are carried out for the electron-impact excitation cross sections from the ground state to the individual magnetic sublevels of the 1s2s^(2)2p^(3)/2 J=2 excited state of highly-charged beryllium-like ions by using a fully relativistic distorted-wave(RDW)*** contributions of the Breit interaction to the linear polarization of the 1s2s^(2)2p^(3)/2 J=2→1s22s2 J=0 magnetic quadrupole(M2)line are investigated systematically for the beryllium isoelectronic sequence with 42≤Z≤*** is found that the Breit interaction depolarizes significantly the linear polarization of the M2 fluorescence radiation and that these depolarization effects increase as the incident electron energy and/or the atomic number is enlarged.

One fifth of the population report adverse reactions to food. Reasons for these symptoms are heterogeneous, varying from food allergy, food intolerance, irritable bowel syndrome to somatoform or other mental disorders. Literature reveals a large discrepancy between truly diagnosed food allergy and reports of food allergy symptoms by care seekers. In most studies currently available the characterization of patient groups is incomplete, because they did not distinguish between immunologic reactions and other kinds of food reactions. In analysing these adverse reactions, a thorough physical and psychological diagnostic approach is important. In our qualitative review, we present those diagnostic measures that are evidenced-based as well as clinically useful, and discuss the various psychological dimensions of adverse reactions to food. It is important to acknowledge the complex interplay between body and mind： Adults and children suffering from food allergy show impaired quality of life and a higher level of stress and anxiety. Pavlovian conditioning of adverse reactions plays an important role in maintaining symptoms. The role of personality, mood, or anxiety in food reactions is debatable. Somatoform disorders ought to be identified early to avoid lengthy and fiustrating investigations. A future task will be to improve diagnostic algorithms, to describe psychological aspects in clearly characterised patient subgroups, and to develop strategies for an optimized management of the various types of adverse reactions to food.

Helicobacter pyloriin gastric corpus of patients 20 years after partial gastric resection

AIM:To determine the long-term prevalence of Helicobacter pylori(H pylori)gastritis in patients after partial gastric resection due to peptic ulcer,and to compare the severity of Hpylori-positive gastritis in the corpus mucosa between partial gastrectomy patients and matched controls. METHODS:Endoscopic biopsies were obtained from 57 patients after partial gastric resection for histological examination using hematoxylin/eosin and Warthin-Starry *** was graded according to the updated Sydney *** of corpus gastritis was compared between Hpylori-positive partial gastrectomy patients and Hpylori-positive duodenal ulcer patients matched for age and gender. RESULTS:In partial gastrectomy patients,surgery was performed 20 years(median)prior to *** 25 patients(43.8%)Hpyloriwas detected histologically in the gastric *** atrophy was more common in H pylori-positive compared to H pylori-negative partial gastrectomy patients(P<0.05).The severity of corpus gastritis was significantly lower in Hpylori-positive partial gastrectomy patients compared to duodenal ulcer patients (P<0.01).There were no significant differences in the activity of gastritis,atrophy and intestinal metaplasia between the two groups. CONCLUSION:The long-term prevalence of Hpylorigastritis in the gastric corpus of patients who underwent partial gastric resection due to peptic ulcer disease is comparable to the general *** expression of Hpylorigastritis in the gastric remnant does not resemble the gastric cancer phenotype.

Hydrogen evolution was detected in an artificial system composed of light-harvesting unit of purified photosystem I, catalyst of hydrogenase, methyl viologen and electron donor under radiation. Absorption spectral features confirmed that electron transfer from electron donors to proton was via a photoinduced reductive process of methyl viologen.
