A novel LC–MS/MS assay for methylprednisolone in human plasma and its pharmacokinetic application

Methylprednisolone is a synthetic glucocorticoid. In our report, the authors proposed a sensitive and selective liquid chromatography/tandem mass spectrometry(LC–MS/MS) assay for the determination of methylprednisolone applying budesonide as internal standard. Liquid–liquid extraction(LLE) having tert–butyl methyl ether(TBME) have been employed to extract methylprednisolone from the plasma samples. Immediately after reconstitution, the samples were chromatographed on a C18 column using an isocratic mobile phase composed of 10 m M ammonium formate buffer and acetonitrile(35:65, v/v). A flow rate of 1.00 ml/min was used to elute the analyte form the column. Analysis was carried out with an API–4000 LC–MS/MS instrument operated in multiple reaction-monitoring(MRM) mode. The linearity has been proven within the concentration range of 10.1–804 ng/ml in plasma samples. The precision(%CV) and accuracy results in five validation batches across five concentration levels were well within the acceptance limits. The drug was stable under different conditions. The particular assay has been proficiently put on pharmacokinetic study in healthy male subjects.

Seven sampling sites in each of three biomes (Western Ghats, foothills of Western Ghats and west coast) of south- western India were investigated to study the distribution, abundance and ecology of pill millipedes (Arthrosphaera) and associated fauna in relation to edaphic features. Abundance and biomass of Arthrosphaera and other millipedes were the highest in Western Ghats, while earthworms were in foothills. Arthrosphaera magna and Arthrosphaera spp. were common in Western Ghats and foothills respectively, while no Arthrosphaera were found in the west coast. None of the sampling sites consisted of more than one species of Arthrosphaera. Biomass of Arthrosphaera, other millipedes and earthworms significantly differed in Western Ghats (P = 9.48 × 10-7) and foothills (P = 1.35 × 10-8), as did the biomass of species of Arthrosphaera (P = 2.76 × 10-7) between Western Ghats and foothills. Correlation analysis revealed that biomass of Arthrosphaera was significantly (P = 0.01, r = 0.45) correlated with soil organic carbon rather than other edaphic fea- tures (pH, phosphate, calcium and magnesium). Distribution pattern, abundance, biomass and ecology of Arthrosphaera of Western Ghats in relation to soil qualities were compared with millipedes of other regions of the world.

Involvement of gastrointestinal tract by cytomegalovirus(CMV) is common. CMV infections mainly run their course without any clinical signs in immunocompetent hosts. In contrast, CMV can cause severe infections with serious consequences in a immunocompromised state typically associated with organ transplants, highly immunosuppressive cancer chemotherapy, advanced HIV infection or treatment with corticosteroids. The incidence and severity of these manifestations of CMV is directly proportional with the degree of cellular immune dysfunction, i.e., CD8+ Cytotoxic T-cell response. Clinical manifestations of CMV can become apparent in different situations including reactivation of CMV from latency, primary infection in a seronegative host, or exposure of a seropositive host to a new strain of CMV. As the clinical signs of CMV in immunodeficient patients are usually sparse, physicians should be highly vigilant about CMV infection, a treatable condition that otherwise is associated with significant mortality. Here we report a rare case of severe gastrointestinal CMV infection with sustained immunodeficiency secondary to treatment with steroids manifesting as fatal duodenal diverticular bleeding.

About 400 pumice clasts collected from the Central Indian Ocean Basin（CIOB）were studied for their morphology and were classified based on their shape and size.A majority of the samples range between1 cm and 36 cm and in the Zinggs shape diagram plot in the equant and oblate *** Corey Shape Factor for most of the samples is close to 0.7,which is common for volcaniclastic material. The physical properties such as density,specific gravity,void ratio,porosity,moisture content and degree of saturation,were determined for 30 pumice *** varies from 0.21 to 0.74 g/cm^3 specific gravity 1.84 to 3.27,void ratio 2.21 to 10.67,porosity 67%to 91%,moisture content during sinking 0.44 to 2.35 and degree of saturation varies from 26.5%to 86%.Binocular and electron microscopy studies reveal that 60%of the vesicles are elongated,30%are spherical and 10%are *** of the pumices exhibits vitrophyric texture with phenocrysts of feldspars and clinopyroxenes.X-ray diffractrogram and mineral analyses confirm plagioclase to be a major phase, while quartz and orthoclase are not *** is commonly present in the ferromanganese oxide coating present over some of the *** paper also delves into some details concerning the controversial origin of the pumices and glass shards in the CIOB.

Tillage plays an important role in modifying soil hydraulic *** objective of the present study was to evaluate the effect of conservation tillage practices in a maize-wheat cropping system on nearsaturated soil hydraulic properties and pore characteristics in the North-West Himalayan region,*** treatments *** tillage(CT),minimum tillage(MT),and zero tillage(ZT)were evaluated in terms of field saturated hydraulic conductivity(ks),unsaturated hydraulic conductivity k(h),the inverse of capillary length(a),flow-weighted mean pore radius(r0),numbers of pores per square meter(n0)and water-conducting macroporosity(Ɛ).The above hydraulic conductivity and pore characteristics were derived from steady-state water flux(q)measured using hood infiltrometer at 0,1,and3 cm pressure head for each treatment after seven years of establishment of this tillage *** revealed significantly(p0.50 mm conducted about 63.60,68.01,and 75.97%of total flow(at 0 cm pressure head)in the corresponding water-conducting macroporosity of 0.00030,0.00044,and 0.00069%of soil volume under CT,MT,and ZT,***,zero-tillage based agriculture system was found to improve near-saturated soil hydraulic properties.

The conventional flexible pavements have been constructed such that the stiffness of the layer reduces with *** crust thickness becomes significantly high for heavy traffic corridors resulting in the consumption of large quantities of construction materials and also increasing environmental *** pavements with the aggregate interlayer(AIL)or stress absorbing membrane interlayer(SAMI)are considered to be one of the alternatives for thick conventional flexible pavements for heavy traffic *** AIL or SAMI is placed between a stiff cement-treated base and asphalt concrete layer to function as crack relief *** change in the composition alters the behaviour of inverted pavements compared to the conventional flexible *** the other hand,wide-base tires are being increasingly preferred by trucking industries due to increased fuel economy and cargo ***,the effect of wide-base tires on the performance of inverted pavements is yet to be *** this study,the 3D finite element(FE)models of inverted pavements considering different crack relief layers were developed,and load from dual-wheel and wide-base tires were *** stress-strain evolution in the various layers of inverted pavements was investigated and discussed in this *** results indicated the higher stress and strains due to wide base tires compared to the dual-wheel ***,pavement with SAMI was found to result in lower stress and strains in the asphalt concrete layer compared to AIL pavements.
