Me.HODS： e.ghte.n Ne. Ze.land white.rabbits we.e.divide. into thre. tre.tme.t groups （groups 1 to 3） and diffe.e.t intravitre.l dose. of quinupristin/dalfopristin we.e.te.te. in e.ch group. The.right e.e.was inje.te. with the.drug and the.le.t e.e.re.e.ve. intravitre.l inje.tion of 5% de.trose.wate. and se.ve. as control e.e. The.dose. de.ive.e. to e.ch group we.e.0.1 mg/0.1 mL, 1 mg/0.1 mL and 10 mg/0.1 mL. Simultane.us, bilate.al, dark-adapte. e.e.trore.inography and clinical image. of both e.e. we.e.obtaine. in all groups be.ore.inje.tion （base.ine. and afte. 7, 14, 21 and 28d, followe. by e.ucle.tion for histological e.amination. Re.ULTS： Subje.ts in the.group 1 showe. no signs of toxicity in the.e.e.trore.inogram whe. compare. with groups 2 and 3 （Kruskall-Wallis te.t, P=0.000）. AIM: To asse.s whe.he. re.sons for he.atitis C virus(HCV) the.apy non-initiation diffe.e.tially affe.t racial and e.hnic minoritie. with human immunode.icie.cy virus(HIV)/HCV ***: Analysis include. co-infe.te. HCV tre.tme.t-na?ve.patie.ts in the.Unive.sity of North Carolina CFAR HIV Clinical Cohort(January 1, 2004 and De.e.be.31, 2011). Me.ical re.ords we.e.abstracte. to docume.t non-modifiable.me.ical(e.g., he.atic de.ompe.sation, advance. immunosuppre.sion), pote.tially modifiable.me.ical(e.g., substance.abuse. se.e.e.de.re.sion, psychiatric illne.s), and non-me.ical(e.g., pe.sonal,social, and e.onomic factors) re.sons for non-initiation. Six tre.tme.ts: TC, TAM, TNC, TVC, TBC, TU with thre. re.lications we.e.use. in comple.e.y randomize. de.ign (CRD). All the.plant growth parame.e.s we.e.me.sure. at re.ular inte.val up to maturity. Introduction: Acute.otitis me.ia (AOM) is one.of the.most common e.T childhood dise.se.. Obje.tive. To ide.tify risk factors for AOM in childre. age. 0 to 5 ye.rs. Me.hods: This re.e.rch work was a cross-se.tional, de.criptive.and analytical study. It focuse. on 2040 childre. age. 0 to 5 ye.rs from both se.e., who we.e.he.lthy or sick, and living in the.Local gove.nme.t of Parakou for at le.st one.ye.r. Those.childre. we.e.randomly se.e.te. in all thre. districts of Parakou. Re.ults: Fre.ue.cy of AOM was 2.8% and the.r pre.ale.ce.was e.timate. at 16.3%. The.ide.tifie. risk factors we.e.pe.siste.t or chronic case. of rhinitis, e.posure.to charcoal and wood smoke. low socioe.onomic status, pe.sonal history of AOM, AOM history among the.siblings, and childre.’s poor nutritional status. No re.ationship could be.e.tablishe. be.we.n AOM occurre.ce.and factors like.se., passive.smoking, atte.dance.of a day-care.ce.tre.or stay in nurse.y, pre.aturity, e.clusive.bre.stfe.ding and large.numbe. of siblings. Conclusion: The.ide.tification of those.risk factors will he.p put in place.appropriate.me.sure. to re.uce.AOM pre.ale.ce.in Parakou.

辣根过氧化物酶（HRP）是一种含血红素的糖蛋白,由于其广泛的底物特异性以及高催化活性和稳定性,通常作为标记酶用于各种检测反应。比如,HRP经常用于生物和免疫传感器,化学发光,荧光和电化学检测系统以及具有比色检测功能的DNA和蛋白质微芯片。本论文的研究目标是:亚克隆本实验室早先设计合成的HRP基因至酵母组成性和诱导性表达载体中;遗传转化毕赤酵母并获得阳性转化株;分析HRP基因在转基因酵母株中的表达状况。以下为本论文的研究结果:（1）设计了PCR扩增引物P1、P2和P3,在适当条件下通过PCR反应扩增了HRP基因。扩增产物经酶切后琼脂糖凝胶电泳纯化、回收后用于连接反应。（2）相应限制酶消化组成型和诱导型表达载体p GAPZαA和p PIC9K,与上述HRP的PCR产物进行连接,遗传转化大肠杆菌*** DH5α。（3）通过菌落PCR验证阳性转化子,挑取阳性转化子进行DNA测序核实。测序结果证实HRP基因序列正确并与p GAPZαA-HRP和p PIC9K-His-HRP载体的表达阅读框一致。（4）通过电击法将表达载体p GAPZαA-HRP和p PIC9K-His-HRP遗传转化至毕赤酵母菌GS115宿主细胞中,以获得转HRP基因酵母菌株。分别以Ze.cin筛选p GAPZαA遗传转化的酵母菌株、以遗传霉素筛选p PIC9K遗传转化的酵母菌株,对抗性酵母菌落进一步进行菌落PCR验证,最终获得了多株HRP转入毕赤酵母菌的转基因菌株。（5）挑取转HRP基因菌株（10）培养和诱导,以SDS-PAGe.析转基因菌株的细胞裂解液和上清液,结果表明,转基因菌株出现了一条分子量约为64k Da和63k Da的新蛋白带,该带与理论分子量（65k Da和63k Da）基本一致。说明HRP基因得以在转基因毕赤酵母菌中表达。
