Transplantation of bone marrow-derived endothelial progenitor cells and hepatocyte stem cells from liver fibrosis rats ameliorates liver fibrosis

AIM To explore the effectiveness for treating liver fibrosisby combined transplantation of bone marrow-derived endothelial progenitor cells(BM-EPCs) and bone marrow-derived hepatocyte stem cells(BDHSCs) from the liver fibrosis *** The liver fibrosis rat models were induced with carbon tetrachloride injections for 6 wk. BM-EPCs from rats with liver fibrosis were obtained by different rates of adherence and culture induction. BDHSCs from rats with liver fibrosis were isolated by magnetic bead cell sorting. Tracing analysis was conducted by labeling EPCs with PKH26 in vitro to show EPC location in the liver. Finally, BM-EPCs and/or BDHSCs transplantation into rats with liver fibrosis were performed to evaluate the effectiveness of BM-EPCs and/or BDHSCs on liver *** Normal functional BM-EPCs from liver fibrosis rats were successfully obtained. The co-expression level of CD133 and VEGFR2 was 63.9% ± 2.15%. Transplanted BM-EPCs were located primarily in/near hepatic sinusoids. The combined transplantation of BM-EPCs and BDHSCs promoted hepatic neovascularization, liver regeneration and liver function, and decreased collagen formation and liver fibrosis degree. The VEGF levels were increased in the BM-EPCs(707.10 ± 54.32) and BM-EPCs/BDHSCs group(615.42 ± 42.96), compared with those in the model group and BDHSCs group(P 0.05) compared with those in the BDHSC(AST, TBIL and PT, P < 0.05) and BM-EPCs(TBIL and PT, P < 0.05) groups. Inhibiting SHP2 reduces glycolysis, promotes microglial M1 polarization, and alleviates secondary inflammation following spinal cord injury in a mouse model

Reducing the secondary inflammatory response, which is partly mediated by microglia, is a key focus in the treatment of spinal cord injury. Src homology 2-containing protein tyrosine phosphatase 2(SHP2), encoded by PTPN11, is widely expressed in the human body and plays a role in inflammation through various mechanisms. Therefore, SHP2 is considered a potential target for the treatment of inflammation-related diseases. However, its role in secondary inflammation after spinal cord injury remains unclear. In this study, SHP2 was found to be abundantly expressed in microglia at the site of spinal cord injury. Inhibition of SHP2 expression using siRNA and SHP2 inhibitors attenuated the microglial inflammatory response in an in vitro lipopolysaccharide-induced model of inflammation. Notably, after treatment with SHP2 inhibitors, mice with spinal cord injury exhibited significantly improved hind limb locomotor function and reduced residual urine volume in the bladder. Subsequent in vitro experiments showed that, in microglia stimulated with lipopolysaccharide, inhibiting SHP2 expression promoted M2 polarization and inhibited M1 polarization. Finally, a co-culture experiment was conducted to assess the effect of microglia treated with SHP2 inhibitors on neuronal cells. The results demonstrated that inflammatory factors produced by microglia promoted neuronal apoptosis, while inhibiting SHP2 expression mitigated these effects. Collectively, our findings suggest that SHP2 enhances secondary inflammation and neuronal damage subsequent to spinal cord injury by modulating microglial phenotype. Therefore, inhibiting SHP2 alleviates the inflammatory response in mice with spinal cord injury and promotes functional recovery postinjury.

EBD-YOLO：a lightweight auxiliary driving detection algorithm based on YOLOv5

A lightweight road-assistant detection algorithm， EBD-YOLO， based on YOLOv5s is proposed to address the problems of high model complexity， computation cost， and difficulty in deployment on resource-limited embedded terminals in existing assisted driving detection algorithms. First， lightweight Transformer model EfficientViT was used as the backbone feature extraction network of YOLOv5s model to reduce network parameters and calculation costs. Secondly， a Focal-GIoU Loss function is proposed for bounding box regression to accelerate model convergence and reduce loss. Thirdly， the feature pyramid structure is improved to a weighted bi-directional feature pyramid network (BiFPN) to enhance localization and semantic features. Then， a dynamic head framework is added to unify the attention mechanism with the object detection head to improve its performance. Finally， a Soft-CIoU＿NMS algorithm is proposed in the post-processing stage to enhance occluded targets' localization and detection ability and reduce the missed detection rate. We conducted experiments on the KITTI and BDD100K datasets for autonomous driving， and the results showed that the EBD-YOLO model reduced in size by 38.4% and 37.2%， respectively. In comparison， the computational cost was reduced by 48.1%. As measured by mAP@0.5， the detection accuracy improved by 0.5% and 5.8%， respectively， and mAP@0.5:0.95 improved by 2.8% and 7%， respectively. These improvements satisfied the requirements for deployment on embedded terminals in cars.

In the fields of biomedicine, detection, and anti-counterfeiting, there is a widespread demand for cost-effective, easily synthesized, and environmentally friendly near-infrared(NIR) materials. Carbon dots(CDs), a cheap and non-toxic zero-dimensional carbon nanomaterial exhibiting exceptional photoluminescence(PL) properties and facile structural modification, have emerged as a promising candidate for the development of novel NIR materials. This study presents the synthesis of CDs capable of emitting a wide range of wavelengths from blue to NIR(456–836 nm) as their concentrations increase. The incorporation of oxygen and nitrogen co-doped atoms facilitates the optimization of non-coplanar angles, thereby promoting favorable π-πstacking interactions. This leads to the formation of metal-free rigid clusters through interconnecting adjacent nanoparticles in a gear-like meshing pattern through self-assembly. Consequently, intramolecular motion is constrained, vibration coupling is mitigated, non-radiative transition losses are minimized, and efficient NIR emission from CD clusters in high concentrations is ensured. Furthermore, NIR light-emitting diode(NIR LED) based on the CD clusters was successfully fabricated and employed for various applications including information encryption, anti-counterfeiting, and penetration detection in the field of NIR imaging.
