Method for the extraction of circulating nucleic acids based on MOF reveals cell-free RNA signatures in liver cancer

Cell-free RNA(cf RNA) allows assessment of health, status, and phenotype of a variety of human organs and is a potential biomarker to non-invasively diagnose numerous diseases. Nevertheless, there is a lack of highly efficient and bias-free cf RNA isolation technologies due to the low abundance and instability of cf ***, we developed a reproducible and high-efficiency isolation technology for different types of cell-free nucleic acids(containing cf RNA and viral RNA) in serum/plasma based on the inclusion of nucleic acids by metal-organic framework(MOF) materials, which greatly improved the isolation efficiency and was able to preserve RNA integrity compared with the most widely used research kit method. Importantly, the quality of cf RNA extracted by the MOF method is about 10-fold that of the kit method, and the MOF method isolates more than three times as many different RNA types as the kit method. The whole transcriptome mapping characteristics of cf RNA in serum from patients with liver cancer was described and a cf RNA signature with six cf RNAs was identified to diagnose liver cancer with high diagnostic efficiency(area under curve = 0.905 in the independent validation cohort) using this MOF method. Thus, this new MOF isolation technique will advance the field of liquid biopsy, with the potential to diagnose liver cancer.

Network selection policy in multi-radio access environment using stochastic control theory

Network selection is crucial in improving the performance of heterogeneous wireless access systems. Most of previous work on network selection or radio resource allocation concentrates on the capability of each available network and ignores the time-varying nature of wireless media due to channel fading. However, the channel condition determines the state of each wireless network and plays a vital role in ensuring quality of service in multi-radio access environment. In this article, we propose a network selection policy using stochastic control theory considering the time-varying and stochastic character of wireless channels. The proposed scheme selects one network among different alternatives in each decision epoch according to the channel state of each network, which is modeled as finite-state Markov channel, with the objectives of increasing the data-rate, decreasing the bit error rate and minishing the delay. The procedure of network selection is formulated as a stochastic control problem, which can be solved using linear programming and primal-dual index heuristic algorithm. Simulation results are presented to show that network selection has great impact on the system performance, and the proposed scheme can improve the performance significantly.

High-resolution melting-based TILLING of γ ray-induced mutations in rice

Targeting Induced Local Lesions IN Genomes （TILLING） is a reverse genetics strategy for the high-throughput screening of induced mutations.γ, radiation, which often induces both insertion/deletion （Indel） and point mutations, has been widely used in mutation induction and crop breeding. The present study aimed to develop a simple, high-throughput TILLING system for screening γ ray-induced mutations using high-resolution melting （HRM） analysis. Pooled rice （Oryza sativa） samples mixed at a 1：7 ratio of Indel mutant to wild-type DNA could be distinguished from the wild-type controls by HRM analysis. Thus, an HRM-TILLING system that analyzes pooled samples of four M2 plants is recommended for screening γ, ray-induced mutants in rice. For demonstration, a γ, ray-mutagenized M2 rice population （n=4560） was screened for mutations in two genes, OsLCT1 and SPDT, using this HRM-TILLING system. Mutations including one single nucleotide substitution （G→A） and one single nucleotide insertion （A） were identified in OsLCT1, and one tdnucleotide （TTC） deletion was identified in SPDT. These mutants can be used in rice breeding and genetic studies, and the findings are of importance for the application of γ, ray mutagenesis to the breeding of rice and other seed crops.

Cellulose nanofiber-coated delignified wood as an efficient filter for microplastic removal

Microplastics（MPs） are a new type of emerging pollutant in the aquatic environment that pose a tremendous risk to aquatic organisms and human health. Although many studies have focused on the removal of MPs, most of the materials used are non-renewable, non-degradable, and relatively costly. The development of green, renewable,and cost-effective materials for the removal of MPs presents a significant challenge. In this work, cellulose nanofiber-coated, delignified wood（CNF-CDW） was prepared and used as an efficient filter to remove MPs for the first time. Balsa wood chip was delignified through three approaches using NaClO2, Na2SO3/NaOH, and a deep eutectic solvent（DES） consisting of choline chloride/lactic acid. Delignified wood（DW） with desirable mechanical properties and a high pore structure was successfully prepared by optimizing the delignification method and reaction conditions. Subsequently, the CNF-CDW was prepared through immersion of DW in a suspension of cellulose nanofiber（CNF）, followed by crosslinking with CaCl2solutions. The effects of CNF concentration, CaCl2concentration, and wood chip thickness on the removal efficiency and flux of CNF-CDW towards polystyrene（PS）MPs were systematically studied. Under the optimal conditions of 0.5 wt% CNF, 1 mol/L CaCl2, and 10 mm in thickness, the CNF-CDW exhibited superior MPs removal performance, with a removal efficiency of 95.97% and a flux of 1146 L/m2·h. Morphological analysis revealed that CNF formed compact films on the surface DW, and MPs were efficiently blocked both on the surface and internal cellular structure of CNF-CDW. Therefore, with its exceptional filtration efficiency, high flux, and sustainable nature, CNF-CDW has a broad application prospect in the field of removing MPs from aquatic environment, promoting a brighter and cleaner future for aquatic ecosystems and human well-being.

Study on carbon emission calculation method and carbon reduction indexes of urban building stock

Carbon emissions associated with buildings are a major source of urban emissions. To put forward the methods and strategies to curb carbon emissions from urban building stock, it is not only necessary to establish a carbon emission calculation method for fine statistical analysis, but also to evaluate carbon emissions of urban planning schemes with applicable indexes. Currently,researches mainly focus on carbon emissions of individual buildings. When expanded to urban building stock, the calculation faces the lack of basic data, inadequate spatial analysis and unspecific carbon reduction indexes. Therefore, this study proposes a bottom-up calculation method for urban building stock, conducts spatial analysis based on carbon balance of urban grids, reveals the coupling mechanism between urban carbon reduction indexes and grid carbon emissions, and systematically establishes a carbon-reduction-oriented urban planning method that comprises calculation, analysis and evaluation, which is applied to Xi'an,China. This study provides a theoretical reference for cities to formulate carbon reduction targets and implement planning strategies by evaluating and predicting carbon emissions from urban building stock.

Safety and energy density are significant for lithium-ion batteries(LIBs),and the flammable organic elec-trolyte is one of the most critical causes of the safety problem of *** LiNi0.8 Co 0.1 Mn 0.1 O 2(NCM811)cathode with high capacity can improve the energy density,the interface stability between NCM811 cathode and electrolytes needs to be ***,we report a multifunctional additive,diethyl(2-(triethoxysilyl)ethyl)phosphonate(DETSP),which can suppress the flammability of the elec-trolyte and enhance the cycling stability of NCM811 cathode with a capacity retention of 89.9%after 400 cycles at 1 C,while that of the blank electrolyte is merely 61.3%.In addition,DETSP is compati-ble well with the graphite anode without impairing the electrochemical ***,the performance and safety of NCM811/graphite full cells are also *** and theoretical re-sults demonstrate that DETSP can scavenge acidic byproducts and is beneficial to form a stable cathode-electrolyte interface(CEI).Accordingly,DETSP can potentially be an effective solution to ameliorating the safety of the commercial electrolyte and improving the stability of high-voltage cathodes.

A credible traffic prediction method based on self-supervised causal discovery

Next-generation wireless network aims to support low-latency, high-speed data transmission services by incorporating artificial intelligence(AI) technologies. To fulfill this promise, AI-based network traffic prediction is essential for pre-allocating resources, such as bandwidth and computing power. This can help reduce network congestion and improve the quality of service(QoS) for users. Most studies achieve future traffic prediction by exploiting deep learning and reinforcement learning, to mine spatio-temporal correlated variables. Nevertheless, the prediction results obtained only by the spatio-temporal correlated variables cannot reflect real traffic changes. This phenomenon prevents the true prediction variables from being inferred, making the prediction algorithm perform poorly. Inspired by causal science, we propose a novel network traffic prediction method based on self-supervised spatio-temporal causal discovery(SSTCD). We first introduce the Granger causal discovery algorithm to build a causal graph among prediction variables and obtain spatio-temporal causality in the observed data, which reflects the real reasons affecting traffic ***, a graph neural network(GNN) is adopted to incorporate causality for traffic prediction. Furthermore,we propose a self-supervised method to implement causal discovery to to address the challenge of lacking ground-truth causal graphs in the observed data. Experimental results demonstrate the effectiveness of the SSTCD method.
