Improvement in cryosurvival of buffalo bull (Bubalus bubalis) sperm by altering freezing rate within critical temperature range

Objective:To optimize the cryopreservation of buffalo bull semen by altering freezing rates within critical temperature range (4℃ to -60℃). Methods: A total of 20 ejaculates each from 5 Murrah buffalo bulls were cryopreserved using programmable biofreezer in 2 phases. In the 1st phase, 9 freezing rates were applied at -2, -5, -10, -20, -30, -40, -50, -60 or -4℃/min (control) from 4℃ to -15℃;at -40℃/min from -15℃ to -60℃. In the 2nd phase, a fixed freezing rate was applied at -30℃/min from 4℃ to -15℃. Six freezing rates were applied at -10, -20, -30, -40 (control), -50 or -60℃/min from -15℃ to -60℃. The freezing from -60℃ to -140℃ were fixed at -50℃/min in both the phases. Post thaw semen quality was assessed in terms of motility, viability, membrane integrity (hypo-osmotic swelling test), sperm abnormalities, and active mitochondria. Data were arc sine transformed and analyzed through one-way analysis of variance using SPSS software. Results: In the 1st phase, percent individual motility, progressive motility and viability were similar among various protocols. Percent hypo-osmotic swelling reactive sperm was higher with freezing at -30℃/min. In the 2nd phase, percent individual motility, viability and hypo-osmotic swelling reactive sperm was higher with freezing at -50℃/min. Sperm head abnormalities were lower at -30℃/min in the 1st phase, but were similar among the protocols of the 2nd phase. Percent active mitochondria were higher at -30℃/min in the 1st phase and at -50℃/min in the 2nd ***:The optimum post thaw semen quality of buffalo bull could be obtained by applying freezing rate at -30℃/min (4℃ to -15℃) and at -50℃/min (-15℃ to -140℃), followed by plunging of straws in into liquid nitrogen for storage.

Health Risk Assessment for Bromate (BrO3–) Traces in Ozonated Indian Bottled Water

For this study, bromide and bromate ions in various commercial brands of Indian bottled water samples were estimated using ion chromatography. The measured mean concentration of bromide and bromate ions in water samples was found to be 28.13 μg/L and 11.17 μg/L respectively. The average level of bromate in Indian bottled water was found to be slightly higher (~ 12%) than the acceptable limits (10 μg/L) recommended by USEPA (US Environmental Protection Agency). Though, kinetically, it is predicted that 62.5% (6.25 μg/L) of bromide in bottled water is needed to convert into bromate upon ozonation to exceed the minimum acceptable limits, but the average formation of bromate determined to be only 26.77% of the predicted concentration. Bromate concentration in bottled water showed a strong correlation with bromide suggesting that its formation in water is very much influenced and controlled by bromide content. The objective of the present study was to determine the BrO3–) content of commercially available different brands of bottled drinking water in India and to estimate the health risks to population due to ingestion. Results of estimated excess cancer risk and chemical toxicity risk to Indian population due to ingestion of bottled water were presented and discussed.

Gastroduodenal artery aneurysm - A rare complication of traumatic pancreatic injury

Aneurysm of gastroduodenal artery （CDA） is rare. Most reported cases are due to pancreatitis and atherosclerosis; however, those following pancreatic trauma have not been reported. We encountered GDA aneurysm in a patient of blunt abdominal trauma, who had pancreatic contusion and retroduodenal air on contrast enhanced computed tomography of abdomen. Emergency laparotomy for suspected duodenal injury revealed duodenal wall and pancreatic head contusion, mild hemoperitoneum and no evidence of duodenal perforation. In the postoperative period, the patient developed upper gastrointestinal hemorrhage on day 5. Repeat imaging revealed GDA aneurysm, which was managed successfully by angioembolization. This case highlights, one, delayed presen- tation of GDA aneurysm after blunt pancreatic trauma and two, its successful management using endovascular technique.

AIM:To determine the expression and clinical significance of transcriptional intermediary factor 1 gamma (TIF1γ),Smad4 and transforming growth factor-beta (TGFβR) across a spectrum representing colorectal cancer (CRC) ***:Tissue microarrays were prepared from archival paraffin embedded tissue,including 51 colorectal carcinomas,25 tubular adenomas (TA) and 26 HPs,each with matched normal colonic *** was performed using antibodies against TIF1γ,Smad4 and TGFβ RⅡ.The levels of expression were scored semi-quantitatively (score 0-3 or loss and retention for Smad4).RESULTS:Overexpression of TIF1γ was detected in 5/26 (19%) HP;however,it was seen in a significantly higher proportion of neoplasms,15/25 (60%) TAs and 24/51 (47%) CRCs (P<0.05).Normal colonic mucosa,HP,and TAs showed strong Smad4 expression,while its expression was absent in 22/51 (43%) ***-expression of TGFβ RⅡ was more commonly seen in neoplasms,13/25 (52%) TAs and 29/51 (57%) CRCs compared to 9/26 (35%) HP (P<0.05).Furthermore,there was a correlation between TIF1γ overexpression and Smad4 loss in CRC (Kendall tau rank correlation value=0.35,P<0.05).The levels of TIF1γ overexpression were significantly higher in stage Ⅲ than in stage Ⅰ and Ⅱ CRC (P<0.05).CONCLUSION:The findings suggest that over-expression of TIF1γ occurs in early stages of colorectal carcinogenesis,is inversely related with Smad4 loss,and may be a prognostic indicator for poor outcome.

Traumatic cardiac injury： Experience from a level-1 trauma centre

Purpose： Traumatic cardiac injury （TCI） is a challenge for trauma surgeons as it provides a short thera- peutic window and the management is often dictated by the underlying mechanism and hemodynamic status, The current study is to evaluate the factors influencing the outcome of TCI. Methods： Prospectively maintained database of TCI cases admitted at a Level-I trauma center from July 2008 to June 2013 was retrospectively analyzed. Hospital records were reviewed and statistical analysis was performed using the SPSS version 15. Results： Out of 21 cases of TCI, 6 （28.6%） had isolated and 15 （71.4%） had associated injuries. Ratio be- tween blunt and penetrating injuries was 2：1 with male preponderance. Mean ISS was 31.95. Thirteen patients （62%） presented with features suggestive of shock. Cardiac tamponade was present in 12 （57%） cases and pericardiocentesis was done in only 6 cases of them. Overall 19 patients underwent surgery, Perioperatively 8 （38,1%） patients developed cardiac arrest and 7 developed cardiac arrhythmia. Overall survival rate was 71.4%. Mortality was related to cardiac arrest （p = 0.014）, arrhythmia （p = 0.014）, and hemorrhagic shock （p = 0.04）. The diagnostic accuracy of focused assessment by sonography in trauma （FAST） was 95.24%. Conclusion： High index of clinical suspicion based on the mechanism of injury, meticulous examination by FAST and early intervention could improve the overall outcome.

Optimization of Synthesis Conditions of LiMn2–xFexO4Cathode Materials Based on Thermal Characterizations

We have synthesized LiMn2–xFexO4 (x = 0, 0.25, and 0.50) cathode materials for applications in Li ion rechargeable batteries via sol-gel method. We studied thermal characteristics of as synthesized materials using differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA). In order to optimize the synthesis conditions, we studied X-ray diffraction (XRD) of synthesized cathode materials at various temperatures, based on the transitions obtained from DSC/TGA. The XRD results can be co-related to the thermal behavior of the synthesized cathode materials and the synthesis conditions optimized.

Design of 2D-Bootlace Lens with Five Focal Feed for Multiple Beam Forming

Utility of microwave bootlace lens is well established. For broadband and wide scanning network, it is required to op-timize and use features of this lens for better communication and scanning device. This report presents the design of a compact 2D bootlace lens with five focal feed for multiple beam forming. This design will help in reducing phase error and will provide power efficiency and better resolution during target tracking. This design will provide better aperture efficiency and scanning angle.

Design of Enterprise Storage Architecture for Optimal Business Continuity

This paper presents a solution for optimal business continuity, with storage architecture for enterprise applications, which will ensure zero data loss and quick recovery. The solution makes use of Internet protocol storage area network （IPSAN）, which is used for data management without burdening the application server, as well as mix of synchronous and semi-synchronous replication techniques to replicate data to remote disaster recovery site. We have presented the detailed design of both synchronous and semi-synchronous with case study of using open source database postgres to prove our point for optimal business continuity. The theoretical presentation is also given for the same.
