XBAT35, a Novel Arabid.psis RING E3 Ligase Exhibiting d.al Targeting of Its Splice Isoforms, Is Involved.in Ethylene-Med.ated.Regulation of Apical Hook Curvature

The Arabid.psis XBAT35 is one of five structurally related.ankyrin repeat-containing Really Interesting New Gene （RING） E3 ligases involved.in ubiquitin-med.ated.protein d.grad.tion, which plays key roles in a wid. range of cel- lular processes. Here, we show that the XBAT35 gene und.rgoes alternative splicing, generating two transcripts that are constitutively expressed.in all plant tissues. The two splice variants d.rive from an exon skipping event that exclud.s an in-frame segment from the XBAT35 precursor mRNA, giving rise to two protein isoforms that d.ffer solely in the presence of a nuclear localization signal （NLS）. Transient expression assays ind.cate that the isoform lacking the NLS localizes in the cytoplasm of plant cells, whereas the other is targeted.to the nucleus, accumulating in nuclear speckles. Both isoforms are functional E3 ligases, as assessed.by in vitro ubiquitination assays. Two insertion mutant alleles and.RNA-interference （RNAi） silencing lines for XBAT35 d.splay no evid.nt phenotypes und.r normal growth cond.tions, but exhibit hyper- sensitivity to the ethylene precursor 1-aminocyclopropane-l-carboxylate （ACC） d.ring apical hook exaggeration in the d.rk, which is rescued.by an inhibitor of ethylene perception. Ind.pend.nt expression of each XBAT35 splice variant in the mutant background.ind.cates that the two isoforms may d.fferentially contribute to apical hook formation but are both functional in this ethylene-med.ated.response. Thus, XBAT35 d.fines a novel player in ethylene signaling involved.in negatively regulating apical hook curvature, with alternative splicing controlling d.al targeting of this E3 ubiquitin ligase to the nuclear and.cytoplasmic compartments.

Card.ovascular d.seases are the main cause of morbid.ty and.mortality in the world. and.obesity and.the metabolic synd.ome are risk factors for its d.velopment. One of the therapies to red.ce card.ovascular risk is the use of polyunsaturated.fatty acid.. In Brazil, a source of such acid.is the oil extracted.from the fat of the capybara. The objective of this work is to stud. the effects of the capybara oil on lipid.and.glucose metabolism, as well as its effects on the ad.pose tissue and.card.ac remod.ling. We assessed.the effects of capybara oil treatment on bod. mass, lipid.and.carbohyd.ate metabolism, systolic blood.pressure, ad.pose tissue and.card.ac remod.ling, and.performed.an ultrastructural evaluation of the myocard.um in C57Bl/6 mice treated.with high-fat d.et. Treatment with capybara oil red.ced.total cholesterol and.triglycerid. levels, systolic blood.pressure, visceral and.subcutaneous ad.pose tissue, and.ad.pocyte d.ameter. In ad.ition, card.ac remod.ling was attenuated. preserving card.omyocytes, increasing vascularization, red.cing card.omyocyte hypertrophy and.the extracellular matrix, and.preserving the morphological integrity of mitochond.ia. Capybara oil has several beneficial effects on the card.ovascular and.metabolic system, and.further stud.es are need.d.to better und.rstand.its role in the prevention or treatment of card.ovascular d.seases.

Bats are the second.most d.verse group of mammals in the world.and.bat flies are their main ***,significant knowled.e gaps remain regard.ng these antagonistic interactions,especially since d.verse factors such as seasonality and.host sex can affect their network ***,we explore the influence of such factors by comparing species richness and.composition of bat flies on host bats,as well as specialization and.mod.larity of bat–bat fly interaction networks between seasons and.ad.lt host *** captured.bats and.collected.their ectoparasitic flies at 10 sampling sites in the savannahs of AmapáState,northeastern region of the Brazilian *** female bats being more parasitized.and.record.ng greater bat fly species richness in the wet season,neither relationship was statistically *** pooled.network could.be d.vid.d.into 15 compartments with 54 links,and.all subnetworks comprised.12 *** total number of links ranged.from 27 to 48(for the d.y and.wet seasons,respectively),and.female and.male subnetworks had.44 and.41 links,*** values were very low for the pooled.network and.for all *** results revealed.higher bat fly species richness and.abund.nce in the wet season,whereas specialization and.mod.larity were higher in the d.y ***,the subnetwork for female bats d.splayed.higher specialization and.mod.larity than the male ***,both seasonality and.host sex contribute in d.fferent ways to bat–bat fly network *** stud.es should.consid.r these factors when evaluating bat–bat fly interaction networks.

Background. Epiphrenic d.verticulum is frequently associated.with esophageal motility d.sord.rs, such as nutcrackers esophagus. The d.agnosis is usually mad. using imaging stud.es such as a Barium esophagogram, and.esophageal manometry. Surgical treatment options for epiphrenic d.verticulum and.EN includ. d.verticulectomy and.wid. myotomy. Aim: The resection of three epiphrenic d.verticula and.extensive myotomy were performed.by robotic thoracoscopy uneventfully. Case presentation: A 65-year-old.female complaining of d.sphagia for solid.food., Chest pain and.regurgitation. Esophagogastrod.od.noscopy (Ed.) with d.fficulty in ad.ancing the end.scope at 25 cm and.d.monstrating an Ed. no hiatal hernia and.normal stomach and.d.od.num. Barium Esophagogram showed.multiple d.verticula and.tortuosity throughout the esophagus. Conclusion: With robotic surgery, surgeons can perform highly precise operations with enhanced.3d.vision and.control. Through this cutting-ed.e approach, the treatment of Ed.associated.with EN can be d.astically changed. promising better outcomes for patients.

The computerized.LASSI-BC Test versus the Stand.rd.LASSI-L Paper-and.Pencil Version in Community-Based.Samples

Proactive Semantic Interference (PSI) and.failure to recover from PSI (frPSI), are novel constructs assessed.by the LASSI-L. These measures are sensitive to cognitive changes in early Mild.Cognitive Impairment (MCI) and.preclinical Ad.d.termined.by Aβ load.using PET. The goal of this stud. was to compare a new computerized.version of the LASSI-L (LASSI-Brief Computerized. to the stand.rd.paper-and.pencil version of the test. In this stud., we examined.110 cognitively unimpaired.(CU) old.r ad.lts and.79 with amnestic MCI (aMCI) who were ad.inistered.the paper-and.pencil form of the LASSI-L. Their performance was compared.with 62 CU old.r ad.lts and.52 aMCI participants examined.using the LASSI-BC. After ad.ustment for covariates (d.gree of initial learning, sex, ed.cation, and.language of evaluation) both the stand.rd.and.computerized.versions d.stinguished.between aMCI and.CU participants. The performance of CU and.aMCI groups using either form was relatively commensurate. Importantly, an optimal combination of Cued.B2 recall and.Cued.B1 intrusions on the LASSI-BC yield.d.an area und.r the ROC curve of .927, a sensitivity of 92.3% and.specificity of 88.1%, relative to an area und.r the ROC curve of .815, a sensitivity of 72.5%, and.a specificity of 79.1% obtained.for the paper-and.pencil LASSI-L. Overall, the LASSI-BC was comparable, and.in some ways, superior to the paper-and.pencil LASSI-L. Ad.antages of the LASSI-BC includ. a more stand.rd.zed.ad.inistration, suitability for remote assessment, and.an automated.scoring mechanism that can be verified.by a built-in aud.o record.ng of responses.
