Female reproductive system of <i>Amaranthus</i>as the target for <i>Agrobacterium</i>-mediated transformation

Agrobacterium-mediated transformation through floral dip and rapid selection process after transgenic event had become a preference as it will overcome the difficulties faced in tissue culturing procedures and lengthy time for screening transformed progenies. Therefore, in this study, three constructs, p5b5 (14,289 bp), p5d9 (15,330 bp) and p5f7 (15,380 bp) in pDRB6b vector which has hygromycin as a selectable marker gene were introduced individually into Agrobacterium tumefaciens strain (AGL1). The cell suspension was applied to Amaranthus inflorescence by drop-by-drop technique and was left to produce seeds (T1). The T1 seeds were germinated and grown to produce seedlings under non-sterile condition. Hygromycin selection on seedling cotyledon leaves results in identification of 12 putative transformants, three from p5b5, four from p5d9 and five from p5f7. All positive putative transformants that were selected at the first stage through hygromycin spraying showed positive result in leaf disk hygromycin assay and in a construct specific polymerase chain reaction-based assay. A ~750 bp amplified hygromycin gene was further verified through sequencing. Our results suggest that Amaranthus inflorescences were able to be transformed and the transformed progenies could be verified through a combination of simple and rapid methods .

Relapse typhoid fever in north-eastern state in Malaysia

Objective:To discuss the prevalence,clinical and laboratory presentations of relapse typhoid ***:All relapse cases were reviewed to identify the clinical and laboratory presentation of the relapse typhoid ***:Two hundred and forty six patients were admitted to a teaching tertiary hospital in north-eastern state of Malaysia and fourteen(5.69%) relapse cases were *** duration of relapse after the patient was discharged was(25.0±9.9) *** patients presented with fever,diarrhoea,headache,abdominal pain and constipation. The duration of fever before admission in the initial episode[(8.6±4.2) d]was significantly longer than the relapse episode[(5.0±2.5) d](P=0.019).Four patients have hepatomegaly in initial episode and ten in relapse episode(P=0.852).The defervescence days of initial episodes was (3.2±2.2) d,comparing to relapse episode[(2.0±1.8) d]which was statistically not significant (P=0.124).Conclusion:Assumption of the relapse typhoid fever is milder comparing to original episodes based on observation and is not supported by statistical analysis.

Litsea elliptica Blume has been traditionally used to treat headache,fever,and stomach ulcer,and has also been used as an insect *** acute and subacute toxicities of *** essential oil were evaluated orally by gavage in female Sprague-Dawley *** the acute toxicity study,*** essential oil was administered in doses from 500 to 4 000 mg/kg(single dose),and in the subacute toxicity test,the following doses were used:125,250,and 500 mg/kg,for 28 consecutive *** the acute toxicity study,*** essential oil caused dose-dependent adverse behaviours and *** median lethal dose value was 3 488.86 mg/kg and the acute non-observed-adversed-effect level value was found to be 500 mg/*** subacute toxicity study of *** essential oil did not reveal alterations in body weight,and food and water *** haematological and biochemical analyses did not show significant differences between control and treated groups in most of the parameters examined,except for the hemoglobin,mean cell hemoglobin concentration,mean cell volume,mean cell hemoglobin,serum albumin,and serum ***,these differences were still within the normal *** abnormalities or histopathological changes were observed in the liver,pancreatic islet of Langerhans,and renal glomerulous and tubular cells of all treated *** conclusion,*** essential oil can be classified in the U group,which is defined as a group unlikely to present an acute hazard according to World Health Organization(WHO) classification.

Litsea elliptica Blume leaves have been traditionally used as medicinal herbs because of its antimutagenicity,chemopreventative and insecticidal properties. In this study,the toxic effects of L. elliptica essential oil against Sprague-Dawley rat’s red blood cells (RBCs) were evaluated. L. elliptica essential oil was given by oral gavage 5 times per week for 3 treated groups in the doses of 125,250,and 500 mg/(kg body weight),respectively,and the control group received distilled water. Full blood count,RBC osmotic fragility,RBC morphological changes,and RBC membrane lipid were analyzed 28 d after the treatment. Although L. elliptica essential oil administration had significantly different effects on hemoglobin (Hb),mean cell hemoglobin concentration (MCHC),mean cell volume (MCV),and mean cell hemoglobin (MCH) in the experimental groups as compared to the control group (Pnormal range. L. elliptica induced morphological changes of RBC into the form of echinocyte. The percentage of echinocyte increased significantly among the treated groups in a dose-response manner (P0.05). It is concluded that structural changes in the RBC membrane due to L. elliptica essential oil administration did not cause severe membrane damage.

Distribution of Chironomidae (Insecta: Diptera) in polluted rivers of the Juru River Basin, Penang, Malaysia

The influence of physical and chemical parameters on the abundance and diversity of chironomids was studied in six rivers with moderate to highly polluted water in the Juru River Basin. The rivers: Ceruk Tok Kun (CTKR) as reference site, and polluted rivers of Pasir (PR), Juru (JR), Permatang Rawa (PRR), Ara (AR) and Kilang Ubi (KUR) were sampled over a period of five months (November 2007-March 2008). Nine chirnomid species: Chironimus kiiensis, C. javanus, Polypedilum trigonus, Microchironomus sp., Dicrotendipes sp., Tanytarsus formosanus, Clinotanypus sp., Tanypus punctipennis and Fittkauimyia sp. were identified. Assessment of their relationships with several environmental parameters was performed using the canonical correspondence analysis (CCA). Tanytarsus formosanus was the most dominant in the relatively clean CTKR and moderately polluted JR with mean densities of 19.66 and 25.32 m 2 , respectively while C. kiiensis was abundant in more polluted rivers. Tanytarsus formosanus, Dicrotendipes sp. and Microchironomus sp. were grouped under moderate to high water temperature, total organic matter (TOM), total suspended solids (TSS), velocity, pH, phosphates and sulphates. However, Tanypus punctipennis, Fittkauimyia sp., and Clinotanypus sp. were associated with high contents of river sediment such as TOM, Zn and Mn and water ammonium-N and nitrate-N and they were associated with higher dissolved oxygen (DO) content in the water. Chironomus kiiensis, C. javanus and P. trigonus showed positive relationships with TOM, ammonium-N and nitrate-N as well as trace metals of Zn, Cu and Mn. These three species could be considered as tolerant species since they have the ability to survive in extreme environmental conditions with low DO and high concentrations of pollutants. Based on the water parameter scores in all rivers, the highest diversity of chironomid larvae was reported in CTKR. Nonlinear photovoltaic(PV)output is greatly affected by the nonuniform distribution of daily irradiance,preventing conventional protection devices from reliably detecting *** fault diagnosis and good maintenance systems are essential for optimizing the overall productivity of a PV system and improving its life ***,a multiscale smart fault diagnosis model for improved PV system maintenance strategies is *** study focuses on diagnosing permanent faults(open-circuit faults,ground faults,and line-line faults)and temporary faults(partial shading)in PV arrays,using the random forest algorithm to conduct time-series analysis of waveform length and autoregression(RF-WLAR)as the main features,with 10-fold cross-validation using Matlab/*** actual irradiance data at 5.86°N and 102.03°E were used as inputs to produce simulated data that closely matched the on-site PV output *** data from the maintenance database of a 2 MW PV power plant in Pasir Mas Kelantan,Malaysia,were used for field testing to verify the developed *** RF-WLAR model achieved an average fault-type classification accuracy of 98%,with 100%accuracy in classifying partial shading and line-line faults.
