Three extraction methods were compared for their efficiency to analyze sitagliptin and simvastatin in rat plasma by LC-MS/MS, including （1） liquid-liquid extraction （LLE）, （2） solid phase extraction （SPE） and （3） supported liquid extraction （SLE）. Comparison of recoveries of analytes with different extraction methods revealed that SLE was the best extraction method. The detection was facilitated with ion trap-mass spectrometer by multiple reactions monitoring （MRM） in a positive ion mode with ESI. The transitions monitored were m./z 441.1→325.2 for simvastatin, 408.2→235.1 for sitagliptin and 278.1→260.1 for the IS. The lower limit of quantification （LLOQ） was 0.2 ng/mL for sitagliptin and 0.1 ng/mL for simvastatin. The effective SLE offers enhanced chromatographic selectivity, thus facilitating the potential utility of the method for routine analysis of biological samples along with pharmacokinetic studies.

The production of extra cellular lipase in Solid State Fermentation (SSF) using Yarrowia lipolytica NCIM 3589 with Palm Kernal cake (Elaeis guineensis) has been studied. Different parameters such as incubation time, inoculum level, initial moisture content, carbon level and nitrogen level of the medium were optimized. Screening of various process variables has been accomplished with the help of Plackett-Burman design. The maximum lipase activity of 18.58 units per gram of dry fermented substrate (U/gds) was observed with the substrate of Palm Kernal cake in four days of fermentation.

The purpose of this study was to determine the publication rate and time to publication following abstract presentation at a major neurosurgical meeting, namely the 2005 annual meeting of the Congress of Neurological Surgeons (CNS). We reviewed all 754 abstracts presented at the 2005 CNS annual meeting to determine publication status and identify factors that are associated with publication. A standardized database search was conducted to determine publication status. Predictors of publication were examined. Of the 754 abstracts presented, 383 (50.8%) were published after 5.3 years. The median time to publication was 450 days (range-646 to 1953 days) with 16% being published prior to the meeting. Oral presentations had a publication rate of 64.0% compared to 48.3% for poster presentations. Of the abstracts not published prior to the meeting, 77% were published within 3 years of the meeting. In univariate analysis, neuro-oncology abstracts (OR 1.88;95% CI 1.34 - 2.64;p < 0.001) and abstracts selected for oral presentation (OR 1.97;95% CI 1.30 - 2.99;p = 0.001) had a significantly higher likelihood of publication. These associations remained independent in multivariate analysis. Neuro-oncology abstracts (p = 0.004), basic science research (p = 0.003) and abstracts selected for oral presentation (p = 0.036) were published in higher impact factor journals. Half of presentations at the annual CNS meeting will be published in peer-reviewed journals with a median time to publication of 450 days. Neuro-oncology abstracts and those abstracts selected for oral presentation have a significantly higher likelihood of publication.

Introduction: Human immunodeficiency virus (HIV) and acquired immune deficiency syndrome (AIDS) are among the most complex health problems of the 21st century. Young people aged 15 - 24 years are the HIV/AIDS at risk group. The objective of this study was to evaluate the effectiveness of an educational interventional program on knowledge on HIV/AIDS among adolescent students of higher secondary school in Kathmandu, Nepal. Methods: The study was the pre test - post test experimental study design with an experimental and a control group conducted in the randomly selected eighteen higher secondary schools. The intervention, educational package on HIV/AIDS, was provided to all grade twelve students in the intervention group from 1st September, 2017 to 2nd January, 2018. In total, 321 from the intervention group, and 283 from the control enrolled at baseline and over 95% of these were followed up at posttest. The data were collected from self-administered questionnaires in English version and analyzed by using independence t-test and paired t-test. Results: Overall pretest knowledge of both intervention and control groups was comparable with 27.58 ± 4.05 and 28.53 ± 3.77 mean ± standard deviation respectively. But statistical analysis showed significant higher knowledge (P = 0.03) among control group. After the educational intervention, the mean knowledge score of control group increased by only 0.47, whereas the same score increased by 11.57 and reached 39.15 ± 3.7 in post-test for intervention group. The difference in post-test score was statistically highly significant (P Conclusion: Educational intervention was efficacious in improving awareness of adolescent students on HIV/AIDS. The study has also indicated that remarkable numbers of adolescents are practicing high risk behavior for HIV/AIDS like having early initiation sexual intercourse, multiple sex partners, using alcohol before intercourse etc. There is need to promote education program among higher secondary

Objective： This study aims to investigate the clinicopathologic significance of lymphatic vessel invasion （LVI） labeled by D2-40 monoclonal antibody in esophageal squamous cell carcinoma （ESCC）. Methods： Immunohistochemical assay was used to detect the expression of D2-40 and LVI in 107 ESCC patients. Then, the correlation between the clinicopathologic feature and the overall survival time of the patients was analyzed. Results： The lymph node metastasis rates were 70% and 21% in the LVI-positive and LVI-negative groups, respectively. The nodal metastasis rate was higher in the LVI-positive group than in the LVI-negative group. Multivariate regression analysis showed that LVI was related to nodal metastasis （P〈0.001）. The median survival time of the patients was 26 and 43 months in the LVI-positive and LVI-negative groups, respectively. Mthough univariate regression analysis showed significant difference between the two groups （P=0.014）, multivariate regression analysis revealed that LVI was not an independent prognostic factor for overall survival in the ESCC patients （P=0.062）. Lymphatic node metastasis （P=0.031）, clinical stage （P=0.019）, and residual tumor （P=0.026） were the independent prognostic factors. Conclusion： LVI labeled by D2-40 monoclonal antibody is a risk factor predictive of lymph node metastasis in ESCC patients.

Growth rate of Yarrowia lipolytica NCIM 3589 was observed in a fermentation medium consisting of peptone, yeast extract, sodium chloride. Logistic equation was fitted to the growth data (time vs. biomass concentration) and compared with the prediction given by Artificial Neural Networks (ANN). ANN was found to be superior in describing growth characteristics. A single MATLAB programme is developed to fit the growth data by logistic equation and ANN.

The VTLLS microscopy studies were made on high purity gold and commercial grade zinc grains in a temperature range of 30-230?C. Differential area ω and surface activity Sa were estimated from photomicrographs. The ω vs dT/dt (rate of heating) curve was seen to differ from those of silver and titanium. The nature of curve between normalized ω and dT/dt was seen to be non-exponential. The characteristic relation between sectorized differential area ωsec and mean temperature was examined. The present study further establishes the simplicity and versatility of the VTLLS technique, in studying the defect-sub-structure of metal particles such as Au and Zn in presence of an imposed temperature gradient in a reasonable way. As such an attempt was made to connect the ω and defect-sub-structure related parameters.

Introduction: The objective of this study was to write the epidemiological, etiological and therapeutic aspects of shock in children in the paediatric medico-surgical emergency unit of the National Donka Hospital. Method: This was a forward-looking, descriptive 6-month study, including all children aged 0 to 15 who were received in the paediatric medico-surgical emergency unit at The Donka National Hospital, where the shock diagnosis was retained and the patients received care. Results: In our study, 26 out of 1123 patients were received in a state of shock. Hypovolemic shock was more common at 24 (92%). Vascular filling by Ringer’s milk was the basic treatment. The 6 to 11 month age group was the most affected and the sex-ratio M/F was 1.6. The acute febrile gastroenteritis and dehydration status were the most frequent causes. The outcome was favorable in 25 patients (96%). Conclusion: Rapid and effective management would improve the vital and functional prognosis of children in shock.

Prevalence and Risk Factors for Albuminuria among HIV Ivorian Children, Using the Dipstick Method

Context: Human immunodeficiency virus (HIV) infected children are at risk of developing chronic kidney disease defined by decreased glomerular filtration rate and or proteinuria. Studies on the prevalence of and risk factors for proteinuria among HIV-infected children in sub-Saharan Africa are scarce and in Cote d’Ivoire, there is paucity of reported data. We determined the prevalence of and the risk factors for proteinuria among HIV-infected Ivorian children in both ART (antiretroviral therapy)-na?ve and ART-exposed children. Method: It was a cross-sectional bi-centric study, conducted from July 2013 to April 2014. Each child’s urine was tested for proteinuria and microalbuminuria using multi test strips and mitral test, respectively. Proteinuria was defined as of significant of 1+ and above on the dipstick or the presence of microalbuminuria ≥ 20 mg/l. Results: Of the 155 HIV-infected children enrolled, there were 78 boys. Sixty seven point seven percent were treated with antiretroviral and 74.1% had treatment duration of less than 5 years. In the first dipstick test, the prevalence of proteinuria was 29% which persisted in 9.7% after 24 hour urinary protein examination. It was significantly different between the 2 groups treated and na?ve (2.8% versus 24%). The advanced CDC clinical stage emerged as the main independent determinant of proteinuria. Conclusion: Proteinuria is common in HIV-infected Ivorian children. Screening for proteinuria in HIV-infected children will help in early and treatment, and thus prevention and progression to chronic kidney disease to end-stage kidney disease.

The rat posterodorsal medial amygdala (MePD) is responsive to the orexigenic neuropeptide Y (NPY) and is a putative candidate to participate in neural circuits that modulate feeding behavior. Here, we studied the effects of intracerebroventricular (icv) microinjection of NPY on the appetitive and food intake behaviors of rats under the paradigm of the self-selection macronutrient isolated diets [high-carbohydrate (high-CHO), high-protein and high-lipid food pellets]. At the same time, Fos expression was also evaluated in the MePD as a marker of local cellular activation. Adult male rats received icv microinjections of NPY (1mg and10mg/5mL, n = 10 and 8, respectively) whereas the control groups either received icv microinjection of artificial cerebrospinal fluid (5mL, n = 8) or underwent sham procedure (n = 8). The data were obtained after a fasting protocol. Feeding behavior was evaluated during a 2 h test period of free access to the selective diets. Rats in all groups preferred the high-CHO diet. Compared to controls, both doses of NPY increased the appetitive behaviors (searching for food and the frequency of attempts to eat any diet) and the percentage of animals eating high-CHO diet. However, only NPY at a dose of 1 μg led to a significant increase in food intake and showed a strong positive correlation with Fos expression in the MePD (p 0.05 in all cases). These new data reveal a biphasic effect of NPY on the appetite and food intake behaviors and suggest that the MePD participates in the NPY-induced feeding behavior in rats.
