Cellular and molecular mechanisms of intestinal fibrosis

Fibrosis is a chronic and progressive process characterized by an excessive accumulation of extracellular matrix (ECM) leading to stiffening and/or scarring of the involved tissue. Intestinal fibrosis may develop in several different enteropathies, including inflammatory bowel disease. It develops through complex cell, extracellular matrix, cytokine and growth factor interactions. Distinct cell types are involved in intestinal fibrosis, such as resident mesenchymal cells (fibroblasts, myofibroblasts and smooth muscle cells) but also ECM-producing cells derived from epithelial and endothelial cells (through a process termed epithelialand endothelial-mesenchymal transition), stellate cells, pericytes, local or bone marrow-derived stem cells. The most important soluble factors that regulate the activation of these cells include cytokines, chemokines, growth factors, components of the renin-angiotensin system, angiogenic factors, peroxisome proliferator-activated receptors, mammalian target of rapamycin, and products of oxidative stress. It soon becomes clear that although inflammation is responsible for triggering the onset of the fibrotic proc-ess, it only plays a minor role in the progression of this condition, as fibrosis may advance in a self-perpetuating fashion. Definition of the cellular and molecular mechanisms involved in intestinal fibrosis may provide the key to developing new therapeutic approaches.

A topology-based bounded rationality day-to-day traffic assignment model

This paper analyzes the day-to-day adjustment process of users’behaviors in a transport network which is affected by relevant alterations such as disruptions due to critical events which cause the impossibility to use one or more *** representing the progressive adjustment of the flows on the network to reach a new equilibrium,a dayto-day discrete-time model is proposed,based on the idea that people are bounded rational in their choices,*** often do not behave according to the optimal solution but they accept solutions they consider ***,in their choice process,are influenced by the topological similarity between the route they are currently using and *** means that they tend to prefer the solutions that are more similar to the one they are already *** parallel,users exhibit a myopic behavior,i.e.,they tend to overestimate the goodness of a route if,when using it,they suddenly experience a significant reduction in travel time compared to what they are used *** the paper it is shown that such route choice behaviour implies that the steady state of the system corresponds to a Bounded Rational User Equilibrium,i.e.,a state that does not diverge from the user equilibrium more than a certain value which increases when the relative importance given to the topological similarity *** model also assumes that these biases vanish,at least with respect to those routes that are most frequently used by users,after a sufficient amount of *** certain conditions,it is then shown that the steady state can eventually collapse into a User *** effectiveness of the proposed model is assessed via simulation results in which two test networks are analyzed in detail to show the evolution of the users’behaviour in a transport network after a disruption.

A model-based approximate λ-policy iteration approach to online evasive path planning and the video game ***-Man

This paper presents a model-based approximate λ-policy iteration approach using temporal differences for optimizing paths online for a pursuit-evasion problem,where an agent must visit several target positions within a region of interest while simultaneously avoiding one or more actively pursuing *** method is relevant to applications,such as robotic path planning,mobile-sensor applications,and path *** methodology described utilizes cell decomposition to construct a decision tree and implements a temporal difference-based approximate λ-policy iteration to combine online learning with prior knowledge through modeling to achieve the objectives of minimizing the risk of being caught by an adversary and maximizing a reward associated with visiting target *** learning and frequent decision tree updates allow the algorithm to quickly adapt to unexpected movements by the adversaries or dynamic *** approach is illustrated through a modified version of the video game ***-Man,which is shown to be a benchmark example of the pursuit-evasion *** results show that the approach presented in this paper outperforms several other methods as well as most human players.

Clear cell hepatocellular carcinoma: origin,metabolic traits and fate of glycogenotic clear and ground glass cells

Clear cell hepatocellular carcinoma(CCHCC)has hitherto been considered an uncommon, highly differentiated variant of hepatocellular carcinoma(HCC) with a relatively favorable prognosis. CCHCC is composed of mixtures of clear and/or acidophilic ground glass hepatocytes with excessive glycogen and/or fat and shares histology, clinical features and etiology with common HCCs. Studies in animal models of chemical, hormonal and viral hepatocarcinogenesis and observations in patients with chronic liver diseases prone to develop HCC have shown that the majority of HCCs are preceded by, or associated with, focal or diffuse excessive storage of glycogen(glycogenosis) which later may be replaced by fat(lipidosis/steatosis). In ground glass cells, the glycogenosis is accompanied by proliferation of the smooth endoplasmic reticulum, which is closely related to glycogen particles and frequently harbors the hepatitis B surface antigen(HBs Ag).From the findings in animal models a sequence of changes has been established, commencing with preneoplastic glycogenotic liver lesions, often containing ground glass cells, and progressing to glycogen-poor neoplasms via various intermediate stages, including glycogenotic/lipidotic clear cell foci, clear cell hepatocellular adenomas(CCHCA) rich in glycogen and/or fat, and CCHCC. A similar process seems to take place in humans, with clear cells frequently persisting in CCHCC and steatohepatitic HCC, which presumably represent intermediate stages in the development rather than particular variants of HCC. During the progression of the preneoplastic lesions,the clear and ground glass cells transform into cells characteristic of common HCC. The sequential cellular changes are associated with metabolic aberrations, which start with an activation of the insulin signaling cascade resulting in preneoplastic hepatic glycogenosis. Triple antiviral therapy in HCV positive patients who failed prior combination therapy

AIM： To assess the efficacy of triple therapy （peginterferon or high dose standard interferon, plus ribavirin and amantadine） in nonresponders to prior combination therapy. METHODS： A total of 196 patients were enrolled in a multicenter, open, randomized study. Patients were given 180 μg/wk of peginterferon-alpha-2a （40 kD） plus ribavirin （800-1000 rag/d） and amantadine （200 rag/d） for 48 wk （group A） or interferon-alpha-2a （6 MU/d for 4 wk, 3 MU/d for 20 wk, and 3 MU tiw for 24 wk） plus ribavirin （800-1000 rag/d） and amantadine （200 rag/d） for 48 wk （group B）. RESULTS： Overall sustained virologic response （SVR） was 26.6% （32.1% and 19.5% in group A and B, P = 0.057）. Baseline ALT 〉120 UI/L （OR 2.4; 95% CI：1.11 to 5.20; P = 0.026） and HCV RNA negativity after 12 wk （OR 8.7; 95% CI： 3.87 to 19.74; P 〈 0.0001） were independently associated with SVR. Therapy discontinuation occurred less frequently in patients treated with peginterferon than standard interferon （P = 0.036）. CONCLUSION： More than 25% of nonresponders to combination therapy can eradicate HCV infection when retreated with triple therapy, especially if they have a high baseline ALT and are treated with pegylated interferon.

It is well known that the passive strategies applying in traditional buildings respond satisfactory to climatic requirements and succeed to provide maximum indoor comfort with minimum energy *** this point of view,it is interesting to quantitatively assess the effectiveness of the vernacular strategies to improve the environmental performance of the building's envelope under desert climate *** research tries to address this issue and was undertaken in southern Algeria where a very hot and arid climate *** effect of some selected passive cooling strategies on enhancing the building's envelope climate performance was *** strategies are inspired from the local vernacular architecture,and they are expected to provide satisfactory indoor thermal comfort for users and to reduce the energy cooling demand from residential *** field and computational investigations,two existing residential buildings were tested:a typical residential unit and a contemporary vernacular(neo-vernacular)*** the latter,climate responsive strategies inspired from vernacular architecture were applied.A comparison based on site measurements was carried out on the two selected buildings which differ from their envelope design properties and components.

Similarly to other adult tissues,a hierarchical structure has been established for the brain,where the differentiated cell types（neurons,oligodendrocytes and astrocytes）are generated from primary progenitor cells,known as type B astrocytes or neural stem cells （NSC）, through one or multiple stages of amplification （transient amplifying cells） that generate precursor cells （iPC） with more restricted potential nlPCs （neural）, alPCs （astrocytes）, olPC （oligodendrocytes）（Kriegstein and Alvarez-Buvlla, 2009）.
