Salivary metabolites are promising noninvasive biomarkers of druginduced liver injury

BACKGROUND Drug-induced liver injury(DILI)is one of the most common adverse events of medication use,and its incidence is ***,early detection of DILI is a crucial challenge due to a lack of biomarkers and noninvasive *** To identify salivary metabolic biomarkers of DILI for the future development of noninvasive diagnostic *** Saliva samples from 31 DILI patients and 35 healthy controls(HCs)were subjected to untargeted metabolomics using ultrahigh-pressure liquid chromatography coupled with tandem mass *** analyses,including partial least squares-discriminant analysis modeling,t tests and weighted metabolite coexpression network analysis(WMCNA),were conducted to identify key differentially expressed metabolites(DEMs)and metabolite *** we utilized least absolute shrinkage and selection operato and random fores analyses for biomarker *** use of each metabolite and metabolite set to detect DILI was evaluated with area under the receiver operating characteristic *** We found 247 differentially expressed salivary metabolites between the DILI group and the HC *** WMCNA,we identified a set of 8 DEMs closely related to liver injury for further prediction ***,the distinct separation of DILI patients and HCs was achieved with five metabolites,namely,12-hydroxydodecanoic acid,3-hydroxydecanoic acid,tetradecanedioic acid,hypoxanthine,and inosine(area under the curve:0.733-1).CONCLUsiON Salivary metabolomics revealed previously unreported metabolic alterations and diagnostic biomarkers in the saliva of DILI *** study may provide a potentially feasible and noninvasive diagnostic method for DILI,but further validation is needed.

The ocean is one of the essential fields of national defense in the future,and more and more attention is paid to the lightweight research of Marine equipment and *** study it is to develop a Machine learning(ML)-based prediction method to study the evolution of the mechanical properties of Al-Li alloys in the marine *** obtained the mechanical properties of Al-Li alloy samples under uniaxial tensile deformation at different exposure times through Marine exposure *** obtained the strain evolution by digital image correlation(DIC).The strain field images are voxelized using 2D-Convolutional Neural Networks(CNN)autoencoders as input data for Long Short-Term Memory(LSTM)neural ***,the output data of LSTM neural networks combined with corrosion features were input into the Back Propagation(BP)neural network to predict the mechanical properties of Al-Li *** main conclusions are as follows:*** variation law of mechanical properties of2297-T8 in the Marine atmosphere is *** the increase in outdoor exposure test time,the tensile elastic model of 2297-T8 changes slowly,within 10%,and the tensile yield stress changes significantly,with a maximum attenuation of 23.6%.*** prediction model can predict the strain evolution and mechanical response simultaneously with an error of less than 5%.*** study shows that a CNN/LSTM system based on machine learning can be built to capture the corrosion characteristics of Marine exposure *** results show that the relationship between corrosion characteristics and mechanical response can be predicted without considering the microstructure evolution of metal materials.

BACKGROUND Chronic heart failure is a complex clinical *** Chinese herbal compound preparation jianpi Huatan Quyu recipe has been used to treat chronic heart failure;however,the underlying molecular mechanism is still not *** To identify the effective active ingredients of jianpi Huatan Quyu recipe and explore its molecular mechanism in the treatment of chronic heart *** The effective active ingredients of eight herbs composing jianpi Huatan Quyu recipe were identified using the Traditional Chinese Medicine Systems Pharmacology Database and Analysis *** target genes of chronic heart failure were searched in the Genecards *** target proteins of active ingredients were mapped to chronic heart failure target genes to obtain the common drugdisease targets,which were then used to construct a key chemical componenttarget network using Cytoscape 3.7.2 *** protein-protein interaction network was constructed using the String *** Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses were performed through the Metascape ***,our previously published relevant articles were searched to verify the results obtained via network *** A total of 227 effective active ingredients for jianpi Huatan Quyu recipe were identified,of which quercetin,kaempferol,7-methoxy-2-methyl isoflavone,formononetin,and isorhamnetin may be key active ingredients and involved in the therapeutic effects of TCM by acting on STAT3,MAPK3,AKT1,JUN,MAPK1,TP53,TNF,HSP90AA1,p65,MAPK8,MAPK14,IL6,EGFR,EDN1,FOS,and other *** pathways identified by KEGG enrichment analysis include pathways in cancer,IL-17 signaling pathway,PI3K-Akt signaling pathway,HIF-1 signaling pathway,calcium signaling pathway,cAMP signaling pathway,NF-kappaB signaling pathway,AMPK signaling pathway,*** studies on jianpi Huatan Quyu recipe suggested that this Chinese compound preparation can regulate the TNF-α,IL-6,MAPK

●AIM:To identify the differential methylation sites(DMS)and their according genes associated with diabetic retinopathy(DR)development in type 1 diabetes(T1DM)children.●METHODS:This study consists of two surveys.A total of 40 T1DM children was included in the first *** no participant has DR,retina thinning was used as a surrogate indicator for *** lowest 25%participants with the thinnest macular retinal thickness were included into the case group,and the others were *** DNA methylation status was assessed by the Illumina methylation 850K array BeadChip assay,and compared between the case and control *** DMS with a potential role in diabetes were *** second survey included 27 T1DM children,among which four had *** methylation patterns of the four DMS identified by 850K were compared between participants with and without DR by pyrosequencing.●RESULTS:In the first survey,the 850K array revealed 751 sites significantly and differentially methylated in the case group comparing with the controls(|Δβ|>0.1 and Adj.Psignificance of Adj.Psites were hypermethylated and 432 were hypomethylated in the case group relative to the *** revealed that the transcription elongation regulator 1 like(TCERG1L,cg07684215)gene was hypermethylated in the four T1DM children with DR(P=0.018),which was consistent with the result from the first *** methylation status of the other three DMS(cg26389052,cg25192647,and cg05413694)showed no difference(all P>0.05)between participants with and without DR.●CONCLUsiON:The hypermethylation of the TCERG1L gene is a risk factor for DR development in Chinese children with T1DM.

Shale oil of the Qingshankou Formation of the Gulong Sag,the northern Songliao Basin,represents the first attempt at large-scale development of pure-shale-type shale oil in *** integrating the multiscale refined reservoir characterization with macro-micro-scale mechanical testing,it is clarified that the Gulong shale is characterized by high clay mineral content,high rock plasticity,highly-developed bedding,and prominent mechanical anisotropy.A three-dimensional(3D)fracture propagation model of hydraulic fracturing was built for the Gulong shale,which fully captures the hydraulic fracture distribution pattern affected by the high bedding density,in-situ stress,and fracturing treatment *** research showed that due to influences of bedding,hydraulic fracturing in the Gulong shale forms a complex fracture morphology featuring the main fracture with multiple perpendicular branches that have different lengths(like the outdoor directional TV antenna);however,the vertical propagation of fractures is inhibited,and the fracture height is commonly less than 10 *** limited stimulated reservoir volume(SRV)is the main problem facing the fracturing stimulation of the Gulong shale *** density has vital effects on fracture morphology,so case-specific fracturing designs shall be developed for shale intervals with different bedding development *** reservoirs with welldeveloped bedding,it is suggested to properly increase the perforation cluster spacing and raise the volume and proportions of viscous fluids of the pad,so as to effectively promote vertical fracture propagation and improve reservoir stimulation *** study integrates multi-scale fine reservoir characterization and macro-micro-scale mechanical testing,as well as the construction and numerical simulation of hydraulic fracturing models for high-density layered shale reservoirs,providing a new approach and methodological framework for the fracturing research of high-density layer

Hydrodynamic characteristics and particle tracking of 90° lateral intakes at an inclined river slope

Lateral intakes are common in *** pump effciency and sediment deposition are determined by the local hydrodynamic characteristics and mainstream division *** hydraulic characteristics of lateral withdrawal from inclined river slopes at different intake elevations should be ***,the division width exhibits significant vertical non-uniformity at an inclined river slope,which should be ***,a three-dimensional(3-D)hydrodynamic and particle-tracking model was developed with the Open Source Field Operation and Manipulation(Open FOAM),and the model was validated with physical model tests for 90°lateral withdrawal from an inclined side *** flow fields,withdrawal sources,and division widths were investigated with different intake bottom elevations,withdrawal discharges,and main channel *** study showed that under inclined side bank conditions,water entered the intake at an oblique angle,causing significant 3-D spiral flows in the intake rather than two-dimensional closed recirculation.A lower withdrawal discharge,a lower bottom elevation of the intake,or a higher main channel velocity could further strengthen this *** average division width and turbulent kinetic energy were smaller under inclined side bank conditions than under vertical bank *** a low intake bottom elevation,a low withdrawal discharge,or a high main channel velocity,the sources of lateral withdrawal were in similar ranges near the local inclined bank in the vertical *** inclined slope conditions,sediment deposition near the intake entrance could be reduced,compared to that under vertical slope *** results provide hydrodynamic and sediment references for engineering designs for natural rivers with inclined terrains.

Adhesive Contact of Elastic Solids with Self-Affine Fractal Rough Surfaces

The elastic adhesive contact of self-affine fractal rough surfaces against a rigid flat is simulated using the finite element method. An array of nonlinear springs, of which the force-separation law obeys the Lennard–Jones potential, is introduced to account for the interfacial adhesion. For fractal rough surfaces, the interfacial interaction is generally attractive for large mean gaps, but turns repulsive as the gap continuously shrinks. The interfacial interactions at the turning point corresponding to the spontaneous contact are shown for various surfaces. For relatively smooth surfaces, the probability density distributions of repulsion and attraction are nearly symmetric. However, for rougher surfaces, the simulation results suggest a uniform distribution for attraction but a monotonously decreasing distribution with a long tail for repulsion. The pull-off force rises with increasing ratio of the work of adhesion to the equilibrium distance, whereas decreases for solids with a higher elastic modulus and a larger surface roughness. The current study will be helpful for understanding the adhesion of various types of rough solids.

The experimental realization of Rydberg dressing technology in ultracold atomic systems provides another superior platform for studying novel states of matter and macroscopic quantum *** this work,based on the mean-field theory,we have investigated the ground-state phases of a two-component Bose–Einstein condensate with Rydberg interaction and confined in a toroidal *** effects of the Rydberg interaction and external potential,especially the Rydberg blockade radius,on the ground-state structure of such a system have been investigated in full parameter *** results show that the Rydberg blockade radius,which can be regarded as another controllable parameter,can be used to obtain a variety of ground-state *** interestingly,it is found that for weak Rydberg interactions,the Rydberg blockade radius breaks the spontaneous rotational symmetry of the system,leading to the formation of a discrete unit cell *** strongly interacting cases,it can be used to realize different orders of discrete rotational symmetry breaking.
