补气消脂方调控PI3K/AKT/mTOR通路改善自噬减轻小鼠非酒精性脂肪肝机制研究

目的：围绕磷酸肌醇3-激酶（phosphoinositide 3-kinase,PI3K）/蛋白激酶B(protein kinase,AKT)/动物雷帕霉素靶蛋白（mechanistic target of rapamycin,mTOR）信号通路和自噬研究补气消脂方对高脂饮食诱导的非酒精性脂肪肝（nonalcoholic fatty liver disease,NAFLD）的干预作用及其可能机制。方法：采用高脂饮食16周，诱导NAFLD小鼠模型，于第13～16周以补气消脂方和易善复灌胃，检测血清肝功能、血脂及炎性细胞因子水平，HE和油红O染色观察肝组织病理形态学，测定肝组织血脂和脂质过氧化损伤指标，Western blot法检测脂肪代谢、自噬以及PI3K/AKT/mTOR相关蛋白表达水平。结果：补气消脂方降低NAFLD模型小鼠肝功能丙氨酸氨基转移酶和谷氨酸氨基转移酶活性，降低甘油三酯、胆固醇、低密度脂蛋白水平，改善肝组织炎症损伤和脂滴堆积，降低肝组织三酰甘油、胆固醇和游离脂肪酸含量，提高超氧化物歧化酶、谷胱甘肽过氧化物酶活性，降低丙二醛含量；下调脂质代谢相关蛋白乙酰辅酶A羧化酶，脂肪酸合成酶表达，上调肉碱棕榈酰转移酶蛋白表达，上调自噬标志物LC3表达，降低自噬底物P62表达，下调PI3K/AKT/mTOR信号通路蛋白表达水平。结论：补气消脂方可有效防治高脂饮食诱导的NAFLD形成，其机制与下调PI3K/AKT/mTOR信号通路改善自噬有关。

Calcium(Ca)is an essential nutrient for plant growth and Ca concentrations in forage have important implications for ruminant diet and *** remains an open question whether forage Ca concentration would be decreased by increasing nitrogen(N)*** manipulated the increasing rates of N addition(2008–2015)in a semiarid grassland,northern *** Ca concentrations for all species were examined in each plot under N *** Ca concentrations at functional group and community levels were calculated based on the concentration of each species presented and their relative biomass in each *** found that community-level Ca concentration remained stable across a gradient of wide-ranged N addition rates,although Ca concentration at both species and functional group levels showed negative responses to N *** that forbs had higher Ca concentration than grasses,the increasing relative biomass of forbs canceled out the negative responses of species-level and functional group-level Ca *** results further showed that community Ca pool showed a positive but saturating response to N addition,with a threshold at the rate of 10 g N m^(−2)yr^(−1).Our findings highlight the role of changes in plant relative biomass in controlling the responses of forage Ca concentration and stock to N enrichment.

Influences of the cooling rate after solution treatment on microstructural evolution and mechanical properties of Rene 80 nickel-based superalloy were investigated by optical microscopy, scanning electron microscopy, transmission electron microscopy and mechanical test. Results showed that the high cooling rate decreased the size of secondary γ＇particles in the supersaturated matrix, but led to a high coarsening rate of γ＇ particles during subsequent aging treatment. Despite various coarsening rates, the size and morphology of γ＇ particles in the final microstructures of all samples were close due to the long enough holding time to an equilibrium state. During the aging of 870 ℃/2 h, primary MC started to decompose with the carbide reaction： MC ＋ γ＇ --＊ MrC or M23C6 -t- γ＇. And a number of observations showed that the coarsening of γ＇ particles on grain boundaries resulted in the depletion of γ＇ during aging treatment. The test results indicated that high cooling rate resulted in the presence of quench crack, and the air cooling method following solution treatment was an optimum heat treatment method for Rene 80.

An infrared detector with high responsivity based on graphene-PbSe thin film heterojunction was ***-quality PbSe thin film and graphene were prepared by molecular beam epitaxy and chemical vapor deposition,*** physical characteristics of PbSe thin film and graphene were performed using X-ray diffraction(XRD),X-ray photoelectron spectroscopy(XPS)and Raman *** photo transistor using PbSe thin film as a sensitizer and graphene as a channel to transport excitons exhibits peak responsivity and detectivity up to~420 A·W^(-1) and 5.9×10^(11) Jones(radiation intensity:0.75 mW·cm^(-2))at room temperature in the near-infrared(NIR)region,*** high optical response is attributed to the photo-excited holes transferring from PbSe film to graphene under ***,it is revealed that the responsivity of graphene-PbSe photo transistor is gate-tunable which is important in photodetectors.

Perturbation to Symmetries and Adiabatic Invariants for Generalized Birkhoffian Systems on Time Scales

Time scale is a new and powerful tool for dealing with complex dynamics problems. The main result of this study is the exact invariants and adiabatic invariants of the generalized Birkhoffian system and the constrained Birkhoffian system on time scales. Firstly, we establish the differential equations of motion for the above two systems and give the corresponding Noether symmetries and exact invariants. Then, the perturbation to the Noether symmetries and the adiabatic invariants for the systems mentioned above under the action of slight disturbance are investigated, respectively. Finally, two examples are provided to show the practicality of the findings.

Tumor hypoxia is one of the major factors restricting the photodynamic therapy(PDT)*** address this problem,we designed an arginine-peptide complex,namely Fluorenylmethoxycarbonyl-Leucine-Leucine-Leucine-Arginine-OH(Fmoc-L_(3)-Arg),which is able to co-assemble with 5,10,15,20-Tetrakis(4-hydroxyphenyl)porphyrin(THPP)into stable nanoparticles(NPs)with uniform and spherical *** THPP/Fomc-L_(3)-Arg NPs were ultra-sensitive to tumorous acidic and oxidative conditions,and could rapidly release photosensitizers in tumor ***,the co-loaded Fmoc-L_(3)-Arg could efficiently generate nitric oxide(NO),inhibiting mitochondrial cellular respiration and increasing oxygen in tumor cells to support the profound improvement of reactive oxygen species(ROS)yield and PDT *** intravenous injection,the THPP/Fomc-L_(3)-Arg NPs greatly accumulated at tumor tissue and significantly inhibited tumor growth upon *** conclusion,such an arginine-peptide complex-based nanoassembly addresses the inevitable problem of hypoxia-induced tumor resistance to PDT.
