Clinical manifestations and spermatogenesis outcomes in Chinese patients with congenital hypogonadotropic hypogonadism caused by inherited or de novo FGFR1 mutations

Fibroblast growth factor receptor 1(FGFR1)mutations are associated with congenital hypogonadotropic hypogonadism(CHH)through inheritance or spontaneous *** detected FGFR1 mutations in a Chinese cohort of 210 CHH patients at Peking Union Medical College Hospital(Beijing,China)using next-generation and Sanger *** assessed missense variant pathogenicity using six bioinformatics tools and compared clinical features and treatment outcomes between inherited and de novo mutation *** 19 patients with FGFR1 mutations,three were recurrent,and 16 were novel *** of the novel mutations were likely pathogenic according to the American College of Medical Genetics and Genomics(ACMG)guidelines,with the prevalent P366L *** majority of FGFR1 mutations was inherited(57.9%),with frameshift mutations exclusive to the de novo mutation *** inherited mutation group had a lower incidence of cryptorchidism,short stature,and skeletal *** the inherited mutation group,luteinizing hormone(LH)levels were 0.5 IU l−1,follicle-stimulating hormone(FSH)levels were 1.0 IU l−1,and testosterone levels were 1.3 nmol l−*** contrast,the de novo group had LH levels of 0.2 IU l−1,FSH levels of 0.5 IU l−1,and testosterone levels of 0.9 nmol l−1,indicating milder hypothalamus–pituitary–gonadal axis(HPGA)functional deficiency in the inherited *** inherited mutation group showed a tendency toward higher spermatogenesis *** conclusion,this study underscores the predominance of inherited FGFR1 mutations and their association with milder HPGA dysfunction compared to de novo mutations,contributing to our understanding of the genetic and clinical aspects of FGFR1 mutations.

VPD modifies CO_(2) fertilization effect on tomato plants via abscisic acid and jasmonic acid signaling pathways

Atmospheric CO_(2)concentration is elevated globally,which has“CO_(2)fertilization effects”and potentially improves plant photosynthesis,yield,and *** the beneficial effect of CO_(2)fertilization being modulated by vapor pressure deficit(VPD),the underlying mechanism is highly *** the present study,the potential roles of hormones in determining CO_(2)fertilization effects under contrasting high and low VPD conditions were investigated by integrated physiological and transcriptomic *** CO_(2)fertilization effects were offset under high VPD conditions and were constrained by plant water stress and photosynthetic CO_(2)*** VPD induced a large passive water driving force,which disrupted the water balance and consequently caused plant water *** water potential,turgor pressure,and hydraulic conductance declined under high VPD *** physiological evidence combined with transcriptomic analyses demonstrated that abscisic acid(ABA)and jasmonic acid(JA)potentially acted as drought-signaling molecules in response to high VPD *** foliar ABA and JA content triggered stomatal closure to prevent excessive water loss under high VPD stress,which simultaneously increased the diffusion resistance for CO_(2)uptake from atmosphere to leaf intercellular *** VPD also significantly increased mesophyll resistance for CO_(2)transport from stomatal cavity to fixation site inside *** chloroplast“sink”CO_(2)availability was constrained by stomatal and mesophyll resistance under high VPD stress,despite the atmospheric“source”CO_(2)concentration being ***,ABA-and JA-mediated drought-resistant mechanisms potentially modified the beneficial effect of CO_(2)fertilization on photosynthesis,plant growth,and yield *** study provides valuable information for improving the utilization efficiency of CO_(2)fertilization and a better understanding of the physiological processes.

Prevalence and clinical impact of sarcopenia in liver transplant recipients:A meta-analysis

BACKGROUND The prevalence of sarcopenia in patients undergoing liver transplantation(LT)remains to be determined partly because of different diagnostic *** has recently been recognized as a new prognostic factor for predicting outcomes in LT *** To estimate the prevalence of sarcopenia and evaluate its clinical effect on LT *** This systematic search was conducted in PubMed,Web of Science,Embase,and Cochrane Library for original English-language articles that investigated the prevalence and influence of sarcopenia in patients undergoing LT from database inception to November 30,*** studies of the definition of sarcopenia that estimate sarcopenia prevalence and evaluate its effect on clinical outcomes and the risk of mortality were *** Twenty-five studies involving 7760 patients undergoing LT were *** pooled prevalence of sarcopenia in patients undergoing LT was 40.7%[95%confidence intervals(95%CI):32.1-49.6].The 1-,3-,and 5-year cumulative probab-ilities of post-LT survival in patients with preoperative sarcopenia were all lower than those without sarcopenia(P<0.05).Sarcopenia was associated with an increased risk of post-LT mortality in patients undergoing LT(adjusted hazard ratio:1.58;95%CI:1.21-2.07).Patients with preoperative sarcopenia had a longer intensive care unit stay,a high risk ratio of sepsis,and serious post-LT complications than those without *** Sarcopenia is prevalent in a substantial proportion of patients undergoing LT and is strongly and independently associated with higher a risk of mortality risk.

Metal-free two-dimensional phosphorene-based electrocatalyst with covalent P-N heterointerfacial reconstruction for electrolyte-lean lithium-sulfur batteries

The use of lithium-sulfur batteries under high sulfur loading and low electrolyte concentrations is severely restricted by the detrimental shuttling behavior of polysulfides and the sluggish kinetics in redox ***-dimensional(2D)few layered black phosphorus with fully exposed atoms and high sulfur affinity can be potential lithium-sulfur battery electrocatalysts,which,however,have limitations of restricted catalytic activity and poor electrochemical/chemical *** resolve these issues,we developed a multifunctional metal-free catalyst by covalently bonding few layered black phosphorus nanosheets with nitrogen-doped carbon-coated multiwalled carbon nanotubes(denoted c-FBP-NC).The experimental characterizations and theoretical calculations show that the formed polarized P-N covalent bonds in c-FBP-NC can efficiently regulate electron transfer from NC to FBP and significantly promote the capture and catalysis of lithium polysulfides,thus alleviating the shuttle ***,the robust 1D-2D interwoven structure with large surface area and high porosity allows strong physical confinement and fast mass ***,with c-FBP-NC as the sulfur host,the battery shows a high areal capacity of 7.69 mAh cm^(−2) under high sulfur loading of 8.74 mg cm^(−2) and a low electrolyte/sulfur ratio of 5.7μL mg^(−1).Moreover,the assembled pouch cell with sulfur loading of 4 mg cm^(−2) and an electrolyte/sulfur ratio of 3.5μL mg^(−1) shows good rate capability and outstanding *** work proposes an interfacial and electronic structure engineering strategy for fast and durable sulfur electrochemistry,demonstrating great potential in lithium-sulfur batteries.

Discharge and mass transfer characteristics of atmospheric pressure gas-solid two-phase gliding arc

In this work,a gas-solid two-phase gliding arc discharge(GS-GAD)reactor was *** arc was formed in the gap between the blade electrodes,and solid powder was deposited on the sieve plate positioned beneath the blade electrodes.A range of experimental parameters,including the inter-electrode spacing,gas flow rate,applied voltage,and the type of the powder,were systematically varied to elucidate the influence of solid powder matter on the dynamics of gliding arc discharge(GAD).The discharge images were captured by ICCD and digital camera to investigate the mass transfer characteristics of GS-GAD,and the electrical parameters,such as the effective values of voltage,current,and discharge power were record to reveal the discharge characteristics of *** results demonstrate that powder undergoes spontaneous movement towards the upper region of the gliding arc due to the influence of electric field *** the discharge voltage,decreasing relative dielectric constant of the powder and reducing the electrode-to-sieve-plate distance all contribute to a greater involvement of powder in the GAD process,subsequently resulting in an enhanced powder concentration within the GAD ***,powder located beneath the gliding arc experiences downward resistance caused by the opposing gas flow and *** gas flow rate notably hampers the powder concentration within the discharge region,and the velocity of powder motion in the upper part of the GAD region is *** the condition of electrode-to-sieve-plate distance of 30 mm,gas flow rate of 1.5 L/min,and peak-to-peak voltage of 31 kV,the best combination of arc gliding and powder spark discharge phenomena can be achieved with the addition of Al_(2)O_(3) powder.

The gut microbiota significantly influences host physiology and provides essential ecosystem *** diet can affect the composition of the gut microbiota,the gut microbiota can also help the host adapt to specific dietary *** carrion crow(Corvus corone),an urban facultative scavenger bird,hosts an abundance of pathogens due to its scavenging *** this,carrion crows infrequently exhibit illness,a phenomenon related to their unique physiological *** present,however,the role of the gut microbiota remains incompletely *** this study,we performed a comparative analysis using 16S rRNA amplicon sequencing technology to assess colonic content in carrion crows and 16 other bird species with different diets in Beijing,*** findings revealed that the dominant gut microbiota in carrion crows was primarily composed of Proteobacteria(75.51%)and Firmicutes(22.37%).Significant differences were observed in the relative abundance of Enterococcus faecalis among groups,highlighting its potential as a biomarker of facultative scavenging behavior in carrion ***,*** isolated from carrion crows was transplanted into model mice to explore the protective effects of this bacterial community against Salmonella enterica *** showed that *** down-regulated the expression of pro-inflammatory cytokines tumor necrosis factor alpha(TNF-α),interferon gamma(IFN-γ),and interleukin 6(IL-6),prevented *** colonization,and regulated the composition of gut microbiota in mice,thereby modulating the host’s immune regulatory ***,*** exerts immunoregulatory and anti-pathogenic functions in carrion crows engaged in scavenging behavior,offering a representative case of how the gut microbiota contributes to the protection of hosts with specialized diets.

＜正＞Neurons develop extensive axonal and dendritic arbors and form numerous *** heterochronic coculture method,we studied the sequential events of the pre- and postsynaptic *** data showed that functional synapses could be formed between young and old neurons within 24 h in culture *** using GAD-GFP mice,we found that in GABAergic synapses,the number of new neurons that function as postsynaptic parters is much more that new neurons act as presynaptic *** together,our results suggest that the postsynaptic components of GABAergic synapses maturate earlier than presynaptic *** sequential development of inhibitory and excitatory synapses may play important role in the circuit *** studies mainly focused on excitatory synapses development and rarely on inhibitory *** the difference between the development process of inhibitory and excitatory synapses and if they share the same developmental mechanism or not is still unclear *** using GAD-GFP mice and dual patch technique,we can clearly pick the GABAergic and Glutamatergic neurons to test if they for a functional synapse or not during the developmental *** far,our results suggest that functional GABAergic synapses appears earlier than Glutamatergic *** there’s a few evidence indicate that certain type of neurons tend to form synapses with a different type of neuron as a post-synaptic tarket.
